The role of the CNOT1 subunit of the CCR4-NOT complex in mRNA deadenylation and cell viability

The human CCR4-NOT deadenylase complex consists of at least nine enzymatic and non-enzymatic subunits.Accumulating evidence suggests that the non-enzymatic subunits are involved in the regulation of mRNA deadenylation,although their precise roles remain to be established.In this study,we addressed the function of the CNOT1 subunit by depleting its expression in HeLa cells.Flow cytometric analysis revealed that the sub G1 fraction was increased in CNOT1-depleted cells.Virtually,the same level of the sub G1 fraction was seen when cells were treated with a mixture of siRNAs targeted against all enzymatic subunits,suggesting that CNOT1 depletion induces apoptosis by destroying the CCR4-NOT-associated deadenylase activity.Further analysis revealed that CNOT1 depletion leads to a reduction in the amount of other CCR4-NOT subunits.Importantly,the specific activity of the CNOT6L immunoprecipitates-associated deadenylase from CNOT1-depleted cells was less than that from control cells.The formation of P-bodies,where mRNA decay is reported to take place,was largely suppressed in CNOT1-depleted cells.Therefore,CNOT1 has an important role in exhibiting enzymatic activity of the CCR4-NOT complex,and thus is critical in control of mRNA deadenylation and mRNA decay.We further showed that CNOT1 depletion enhanced CHOP mRNA levels and activated caspase-4,which is associated with endoplasmic reticulum ER stress-induced apoptosis.Taken together,CNOT1 depletion structurally and functionally deteriorates the CCR4-NOTcomplex and induces stabilization of mRNAs,which results in the increment of translation causing ER stress-mediated apoptosis.We conclude that CNOT1 contributes to cell viability by securing the activity of the CCR4-NOT deadenylase.

Identification of sources of lead in the atmosphere by chemical speciation using X-ray absorption near-edge structure(XANES) spectroscopy

Sources of Pb pollution in the local atmosphere together with Pb species, major ions, and heavy metal concentrations in a size-fractionated aerosol sample from Higashi-Hiroshima （Japan） have been determined by X-ray absorption near-edge structure （XANES） spectroscopy, ion chromatography, and ICP-MS/AES, respectively. About 80% of total Pb was concentrated in fine aerosol particles. Lead species in the coarse aerosol particles were PbC204, 2PbCO3-Pb（OH）2, and Pb（NO3）2, whereas Pb species in the fine aerosol particles were PbC204, PbSO4, and Pb（NO3）2. Chemical speciation and abundance data suggested that the source of Pb in the fine aerosol particles was different from that of the coarse ones. The dominant sources of Pb in the fine aerosol particles were judged to be fly ash from a municipal solid waste incinerator and heavy oil combustion. For the coarse aerosol particles, road dust was considered to be the main Pb source. In addition to Pb species, elemental concentrations in the aerosols were also determined. The results suggested that Pb species in size-fractionated aerosols can be used to identify the origin of aerosol particles in the atmosphere as an alternative to Pb isotope ratio measurement.

Axial charges of N(1535) and N(1650) in two-flavor lattice QCD

We show the lattice QCD results on the axial charge gAN＊-N＊ of negative parity nucleon resonances, N＊（1535） and N＊（1650）, which are key clues to the chiral structure in baryon sector. The measurements are performed with up and down dynamical quarks employing the renormalization-group improved gauge action at β=1.95 and the O（a） improved clover quark action with the hopping parameters, κ=0.1375, 0.1390 and 0.1400. In order to properly separate signals of N＊（1535） and N＊（1650）, we construct 2 × 2 correlation matrices and diagonalize them. Wraparound contributions in the correlator, which can be another source of signal contaminations, are eliminated by imposing the Dirichlet boundary condition in the temporal direction. We find that the axial charge of N＊（1535） takes small values as gAN＊N＊-O（0.1）, whereas that of N＊（1650） is about 0.5, which is found independent of quark masses and consistent with the predictions by the naive nonrelativistic quark model.

Experimental study on the in-plane thermal conductivity of Au nanofilms

The in-plane thermal conductivity of Au nanofilms with thickness of 23 nm, which are fabricated by the electron beam-physical vapor deposition method and a suspension technology, is experimentally measured at 80-300 K by a one-dimensional steady-state electrical heating method. Strong size effects are found on the measured nanofilm thermal conductivity. The Au nanofilm in-plane thermal conductivity is much less than that of the bulk material. With the increasing temperature, the nanofilm thermal conductivity increases. This is opposite to the temperature dependence of the bulk property. The Lorenz number of the Au nanofilms is about three times larger than the bulk value and decreases with the increasing temperature, which indicates the invalidity of the Wiedemann-Franz law for metallic nanofilms.

Background: Medical staff provide care to spouses of terminal cancer patients through trial and error by meeting their various support needs and spousal factors regarding their anticipatory grief. Studies on the association between spousal characteristics and anticipatory grief have been inconclusive;additionally, there has been insufficient research on support needs for anticipatory grief of spouses. This study aimed to explore the spousal characteristics and social support needs predicting anticipatory grief in spouses of patients with cancer at the end of life. Methods: This was a cross-sectional study. Eligible spouses (n = 102) completed a self-report questionnaire in two hospitals with palliative care units in Japan. The questionnaire included demographic information, a tool assessing social support needs of spouses, and the Anticipatory Grief Scale for Family Caregivers. Results: Simple regression analyses indicated that patient age, chemotherapy, no treatment, ECOG PS3, children aged under 20 years, total score of “social support needs regarding the disease and treatment of the patients” and subscale scores (“medical condition and cure,” “daily life and social support,” and “intimacy and employment”), and total score of “social support needs of the spouses” and subscale scores (“family psychological issues and social support” and “intimacy, employment, and society”) were significant variables (all p p Conclusions: Patients having no experience of “chemotherapy” and higher “social support needs of the spouses” in Model 1, and greater spousal needs of “family psychological issues and social support” in Model 2 were significant predictors of severe anticipatory grief. Medical staff should pay attention to these risk factors that predict anticipatory grief among spouses.

Fluid-induced high-temperature metasomatism at Rundv?gshetta in the Lützow-Holm Complex, East Antarctica: Implications for the role of brine during granulite formation

We report new petrological, phase equilibria modeling, and fluid inclusion data for pelitic and mafic granulites from Rundv?gshetta in the highest-grade region of the Neoproterozoic Lützow-Holm Complex(LHC),East Antarctica, and provide unequivocal evidence for fluid-rock interaction and high-temperature metasomatism in the presence of brine fluid. The studied locality is composed dominantly of well-foliated pelitic granulite(K-feldspar+quartz+sillimanite+garnet+ilmenite) with foliation-parallel bands and/or layers of mafic granulite(plagioclase+orthopyroxene+garnet+ilmenite+quartz+biotite). The boundary between the two lithologies is defined by thin(about 1 -20 cm in thick) garnet-rich layers with a common mineral assemblage of garnet+plagioclase+quartz+ilmenite+biotite ? orthopyroxene. Systematic increase of grossular and decrease of pyrope contents in garnet as well as decreasing Mg/(Fe+Mg) ratio of biotite from the pelitic granulite to garnet-rich rock and mafic granulite suggest that the garnet-rich layer was formed by metasomatic interaction between the two granulite lithologies. Phase equilibria modeling in the system NCKFMASHTO demonstrates that the metasomatism took place at 850 -860℃, which is slightly lower than the peak metamorphism of this region, and the modal abundance of garnet is the highest along the metapeliteemetabasite boundary(up to 40%), which is consistent with the field and thin section observations. The occurrence of brine(7.0 -10.9 wt.% Na Cleqfor ice melting or 25.1 -25.5 wt.% NaC leqfor hydrohalite melting) fluid inclusions as a primary phase trapped within plagioclase in the garnet-rich layer and the occurrence of Cl-rich biotite(Cl = 0.22 -0.60 wt.%) in the metasomatic rock compared to that in pelitic(0.15 -0.24 wt.%) and mafic(0.06-0.13 wt.%) granulites suggest infiltration of brine fluid could have given rise to the high-temperature metasomatism. The fluid might have been derived from external sources possibly related to the formation of major suture zones formed during the Gondwan

Lipophilic extractives of the inner and outer barks from six different Pinus species grown in Indonesia

The chemical compositions of the dichloromethane extracts of inner and outer barks from six Pinus species（P.elliotii,P.oocarpa,P.caribeae,P.merkusii,P.montezumae,and P.insularis） grown in Indonesia were investigated by GC and GC–MS.Generally,the amounts of extractive contents were higher in the inner bark than in the outer bark except for P.merksuii.Fatty acids,monoterpenes,sesquiterpenes,resin acids,triterpenoids,and steroids were detected and quantified.Inner and outer barks differed not only in content of these compounds but also in their composition.Fatty acids and alcohols were the major classes of lipophilic compounds in the outer bark of P.caribeae, P.insularis,and P.montezumae.Steroids and triterpenoids were the dominant compounds identified in the inner bark of P.elliotii,P.insularis,and P.merkusii.Resin acids were the most abundant group in the inner bark of P.oocarpa whereas monoterpenes and sesquiterpenes were recorded in minor quantities in both bark layers of all species.

Structure and Bonding in Some Gd(Ⅲ) Metal Complexes Studied by Three-Dimensional X-Ray Analysis and ^(155)Gd Mssbauer Spectroscopy

Some functional lanthanide metal complexes, such as acetylacetonato complexes, ethylenediaminetetraacetato complexes, were successfully applied for diagnostic technique. The authors are interested in investigating the structure and bonding in lanthanide and actinide metal complexes using 166Er, t55Gd, and 237Np Mtissbauer spectroscopies in connection with single-crystal and/or powder X-ray diffraction, making clear the differences on their structures as well as the differences in the participation of 4f and 5f orbitals in the chemical bonds. In this article, the crystal structures of two novel Gd（Ⅲ） acetylacetonato complexes, Gd（pta）3 · 2H2O （pta = 1,1,1 -trifluoro-5,5-dimethy 1-2,4-hexanedione） and Gd（bfa）3 · 2H2O （bfa = 1, 1, 1 -trifluoro-4-phenyl-2-4-butanedione） were reported. Though both of them were dihydrate and had distorted square antiprismatical structure, Gd（pta）3 · 2H2O crystallizes in the P 2 1/n （#14） monoclinic space group and its lattice parameters are a = 1.4141（6） nm, b = 1.0708（3） nm, c =2.2344（4） nm, β =952.4（2）°, and Gd（bfa）3· 2H2O crystallizes in P 212121 orthorhombic space group and its lattice parameters were a = 1.322 （1） nm, b = 2.295 （1） nm, c = 1. 0786（8） nm. In the meantime, the authors had finished a systematic investigation on the ^155Gd Mossbauer isomer shift （δ） of various Gd（Ⅲ） metal complexes having a different coordination number （C.N.） and different ratios coordinating oxygen to nitrogen. A tendency for the 6 value to decrease with an increase in the C.N, and the number of the nitrogen atom coordinating to Gd was confirmed. This indicated that the Gd-O and/or Gd-N bond in the investigated Gd（Ⅲ） metal complexes had a small covalent contribution, which was possible to be deduced from the O and/or N atoms of the lisands donating electrons to 6s, 5d, and 4f orbitals of Gd.

Different effects of grain boundary scattering on charge and heat transport in polycrystalline platinum and gold nanofilms

The in-plane electrical and thermal conductivities of several polycrystalline platinum and gold nanofilms with different thicknesses are measured in a temperature range between the boiling point of liquid nitrogen （77K） and room temperature by using the direct current heating method. The result shows that both the electrical and thermal conductivities of the nanofilms reduce greatly compared with their corresponding bulk values. However, the electrical conductivity drop is considerably greater than the thermal conductivity drop, which indicates that the influence of the internal grain boundary on heat transport is different from that of charge transport, hence leading to the violation of the Wiedemann-Franz law. We build an electron relaxation model based on Matthiessen＇s rule to analyse the thermal conductivity and employ the Mayadas ＆ Shatzkes theory to analyse the electrical conductivity. Moreover, a modified Wiedemann-Franz law is provided in this paper, the obtained results from which are in good agreement with the experimental data.
