HPTLC-MS as a Neoteric Hyphenated Technique for Separation and Forensic Identification of Drugs

Drugs are traditionally been identified on basis of chromatographic-spectroscopic hyphenated techniques in instrumental analysis. Gas chromatography (GC) and Liquid chromatography (LC) hyphenated with mass spectroscopy (MS) i.e. GC-MS and LC-MS give reliable and confirmatory results in drugs identification. In the present work the novel hyphenated technique High Performance Thin Layer Chromatography-Mass Spectroscopy (HPTLC-MS) has been used. This technique provides efficient, quick and simple method for identification and separation of Narcotic drugs and psychotropic substances. The drugs under study are Papaverine, Methadone, Cocaine, Ketamine, Caffeine, Codeine, Diazepam, Thebaine, Heroin, Methamphetamine, Carbamazepine, Morphine, Narcotine and Ephedrine. The present study comprising of sixteen drugs has been carried out on CAMAG HPTLC instrument with automatic sampling. Thin layer chromatography (TLC) plates were developed in various solvent systems, scanned under TLC scanner and the results in terms of Retention Factor (Rf value) and UV spectrum (λmax) are presented in the manuscript. Using hyphenated technique of HPTLC-MS (MS 2020 SHIMADZU) spots of these drugs from TLC plate was lifted with CAMAG TLC-MS interface and confirmed by the mass spectrum of the individual drugs by their m/z values thus delivering fast and accurate confirmatory result on the TLC plate.

Use of FFT in Protein Sequence Comparison under Their Binary Representations

The paper considers Voss type representation of amino acids and uses FFT on the represented binary sequences to get the spectrum in the frequency domain. Based on the analysis of this spectrum by using the method of inter coefficient difference (ICD), it compares protein sequences of ND5 and ND6 category. Results obtained agree with the standard ones. The purpose of the paper is to extend the ICD method of comparison of DNA sequences to comparison of protein sequences. The topic of discussion is to develop a novel method of comparing protein sequences. The main achievements of the work are that the method applied is completely new of its kind, so far as protein sequence comparison is concerned and moreover the results of comparison agree with the previous results obtained by other methods for the same category of protein sequences.

Condensed Matrix Descriptor for Protein Sequence Comparison

The present paper develops a novel way of reducing a protein sequence of any length to a real symmetric condensed 20 × 20 matrix. This condensed matrix can be nicely applied as a protein sequence descriptor. In fact, with such a condensed representation, comparison of two protein sequences is reduced to a comparison of two such 20 × 20 matrices. As each square matrix has a unique Alley Index/normalized Alley Index, such index is conveniently used in getting distance matrix to construct Phylogenetic trees of different protein sequences. Finally protein sequence comparison is made based on these Phylogenetic trees. In this paper three types viz., NADH dehydrogenase subunit 3 (ND3), subunit 4 (ND4) and subunit 5 (ND5) of protein sequences of nine species, Human, Gorilla, Common Chimpanzee, Pygmy Chimpanzee, Fin Whale, Blue Whale, Rat, Mouse and Opossum are used for comparison.

BACKGROUND Scoring systems have not been evaluated in oncology *** aimed to assess the performance of Acute Physiology and Chronic Health Evaluation(APACHE)II,APACHE III,APACHE IV,Simplified Acute Physiology Score(SAPS)II,SAPS III,Mortality Probability Model(MPM)II0 and Sequential Organ Failure Assessment(SOFA)score in critically ill oncology *** To compare the efficacy of seven commonly employed scoring systems to predict outcomes of critically ill cancer *** We conducted a retrospective analysis of 400 consecutive cancer patients admitted in the medical intensive care unit over a two-year *** outcome was hospital mortality and the secondary outcome measure was comparison of various scoring systems in predicting hospital *** In our study,the overall intensive care unit and hospital mortality was 43.5%and 57.8%,*** of the seven tested scores underestimated *** mortality as predicted by MPM II0 predicted death rate(PDR)was nearest to the actual mortality followed by that predicted by APACHE II,with a standardized mortality rate(SMR)of 1.305 and 1.547,*** best calibration was shown by the APACHE III score(χ^(2)=4.704,P=0.788).On the other hand,SOFA score(χ^(2)=15.966,P=0.025)had the worst calibration,although the difference was not statistically *** of the seven scores had acceptable discrimination with good efficacy however,SAPS III PDR and MPM II0 PDR(AUROC=0.762),had a better performance as compared to *** correlation between the different scoring systems was significant(P<0.001).CONCLUSION All the severity scores were tested under-predicted mortality in the present *** the difference in efficacy and performance was not statistically significant,the choice of scoring system used may depend on the ease of use and local preferences.

Comparative Study of Sublingual and Vaginal Misoprostol in Second Trimester Induced Abortion

Background: To identify an effective misoprostol-only regime for the termination of second trimester pregnancy. Objectives: To compare the efficacy, safety and acceptability of sublingual and vaginal misoprostol for second trimester pregnancy termination. Methods: In a prospective randomized comparative study, over 138 pregnant women at 13 - 20 weeks (91 - 140 days) of gestation requiring medical abortion were randomly assigned to the sublingual or vaginal route for misoprostol administration with dose schedule of 400 mcg every 3 hours up to 5 doses within 24 hours. The course of misoprostol was repeated if the woman did not abort within 24 hours. Primary outcome was the efficacy of the treatment to terminate pregnancy completely at 24 and 48 hours. Secondary outcomes measured were induction-abortion interval, side effects, failure rate, and women’s perception to these treatments. Results: At 24 h, the complete abortion rate was?87.88% in the vaginal administration group and 79.41% in sublingual group (difference 8.5%, 95%?CI: 3.8 to 13.2). No significant difference in the complete abortion rates was observed at 48 h(90.91% versus 88.24% difference: 2.7%, 95% CI: ?0.04 to 5.4) when vaginal and sublingual?groups were compared. Mean induction-abortion interval in sublingual and vaginal groups was 12.28 h (95% CI of mean 11.019 - 13.541 h) and 13.11 h (95% CI of mean 12.0301 - 14.1899 h) respectively;p = 0.485. The rates of side effects were similar in both groups except for fever, which was more common in vaginal group. Significantly more women in the sublingual group preferred the route as compared to vaginal administration (RR 1.618. 95% CI: 1.277 - 2.050;p < 0.0001). Conclusion: Both sublingual and vaginal routes of misoprostol are equally effective in medical termination of pregnancy in second trimester but sublingual route was preferred by the women.

Evaluation of Topical Application of <i>Azadirachta indica</i>(Neem) in Prevention of Gingivitis: A Randomized Control Trial

Background: The purpose of the present study was to envisage the anti-plaque and anti-inflammatory properties of neem (Azadirachta) in the form of a mouth gel. Materials and Methods: A total of 60 subjects (33 males and 27 females) from age group 19 - 35 years, suffering from mild to moderate gingivitis were randomly divided into two groups. Group A (n = 30) participants were advised experimental (neem) gel and Group B (n = 30) participants were advised 0.2% chlorhexidine gel. All the clinical parameters (the gingival index by Loe and Silness [1], Plaque index by Silness and Loe [2] and sulcus bleeding index by Muhlemann H. R. and Sen S. [3]) were recorded at baseline, on the 14th day and 21st day by the same clinician in both groups. Results: on intragroup comparison the gingival index, plaque index and sulcus bleeding index decrease significantly in both the groups in all assessed periods. On intergroup comparison the indices did not differ significantly at baseline but after 2nd and 3rd week, the indices in Group B decrease more than the group A, which was statistically significant. Conclusion: Based on the observations of our study, it can be concluded that chlorhexidine gluconate as well as neem gel can be effectively used as an adjunct to mechanical plaque control in prevention of plaque and gingivitis. chlorhexidine gluconate gel has been found to be more effective when antiplaque and anti-inflammatory properties were considered. The effect of neem observed maybe because of its anti-inflammatory action. The antiplaque action of chlorhexidine gluconate is due to its substantivity. Substantivity of neem is required to be further studied.

The aim of this study was to modify the sago starch and evaluate its efficacy as tablet disintegrant. Cross-linked car-boxymethylated sago starch (CMSS) was synthesized using native sago starch (SS) and monochloroacetic acid (MCA) with sodium hydroxide in microwave radiation environment. FT-IR analysis of the sample confirmed the carboxy-methylation by showing absorption peak at 1607.2 cm-1. CMSS with degree of substitution (DS) of 0.31 was formed and, it was further evaluated as disintegrant in Ondasetron based tablets. The results revealed that CMSS could be used as disintegrant in tablet formulation in concentration dependant manner.

In this paper measurement of real part of permittivity and loss tangent of (1-x) ***3 (ZMT) material in powder form in S-band of microwave frequencies is presented and the associated accuracy estimated. This approach is based on the measurement of transmitted power from the cavity at and off resonance. Details of the design and fabrication of the rectangular cavity and the input and output coupling are given. The variation of dielectric properties of (1-x) ***3 (ZMT) with x-value and calcination temperature (for x = 0.1) is presented. The effect of doping of V2O5 is also studied. The results of present work may provide useful design guidelines for development of microwave compo-nents including dielectric resonator antennas.

A 45 year old woman was diagnosed as having anteromedial radial head dislocation and distal radius fracture five months after her injury on fight forearm. The radial head dislocation led to ulnar nerve compression. She had severe restriction of her elbow movements. She was treated with arthrolysis, decompression of the ulnar nerve and radial head resection. The reverse Essex Lopresti injury and radial head dislocation compressing the ulnarnerve has not been reported in English language literature to the best of our knowledge. A mechanism is proposed for the injury. In acute presentations, restoration of both the radioulnar joints should be done and neglected nature of such injury leads to suboptimal outcomes.
