Enhanced organic degradation and microbial community cooperation by inoculating Bacillus licheniformis in low temperature composting

Microbial activity and interaction are the important driving factors in the start-up phase of food waste composting at low *** aim of this study was to explore the effect of inoculating Bacillus licheniformis on the degradation of organic components and the potential microbe-driven mechanism from the aspects of organic matter degradation,enzyme activity,microbial community interaction,and microbial metabolic *** results showed that after inoculating ***,temperature increased to 47.8℃ on day2,and the degradation of readily degraded carbohydrates(RDC)increased by 31.2%,and the bioheat production increased by 16.5%.There was an obvious enhancement of extracellular enzymes activities after inoculation,especially amylase activity,which increased by 7.68 times on day *** inoculated *** colonized in composting as key genus in the start-up *** network analysis and Mantel test indicated that inoculation drove the cooperation between microbial network modules who were responsible for various organic components(RDC,lipid,protein,and lignocellulose)degradation in the start-up *** function prediction suggested that carbohydrate metabolisms including starch and sucrose metabolism,glycolysis/gluconeogenesis,pyruvate metabolism,etc.,were improved by increasing the abundance of related functional genes after *** conclusion,inoculating *** accelerated organic degradation by driving the cooperation between microbial network modules and enhancing microbial metabolism in the start-up phase of composting.

针对餐厨废弃物堆肥腐殖酸不稳定易降解的问题,研究生物炭添加对餐厨废弃物堆肥腐殖酸形成的影响。一共设置两个处理(CK,不添加生物炭;BC,按照堆体干质量添加5%生物炭),整个堆肥周期持续15 d。堆肥结束时,BC的还原糖、氨基酸、多酚分别为2.05、1.57、2.25 mg/g,分别比CK低1.97、0.68、0.43 mg/g;BC的胡敏酸(HA)、HA/富里酸(FA)(质量比)分别为17.04 mg/g、1.85,较CK提升了61.21%、110.23%。BC的气单胞菌(Aerococcus)、土地芽孢杆菌(Terribacillus)、蜡样芽孢杆菌(Cerasibacillus)、巨球菌(Macrococcus)、Tepidanaerobacter相对丰度比CK高0.30、0.04、0.01、0.01、0.03百分点,土芽孢杆菌(Geobacillus)、小多孢菌(Micropolyspora)相对丰度比CK分别低0.79、0.16百分点。结果表明添加生物炭有利于:(1)还原糖、氨基酸、多酚向腐殖酸的转化;(2)促腐微生物(气单胞菌、土地芽孢杆菌、蜡样芽孢杆菌、巨球菌、Tepidanaerobacter)的生长;(3)堆肥的腐殖化。本研究对生物炭在餐厨废弃物肥料化上的应用以及堆肥腐殖化具有重要意义。

The white-blotched river stingray(Potamotrygon leopoldi)is a cartilaginous fish native to the Xingu River,a tributary of the Amazon River *** a rare freshwater-dwelling cartilaginous fish in the Potamotrygonidae family in which no member has the genome sequencing information available,*** provides the evolutionary details in fish phylogeny,niche adaptation,and skeleton *** this study,we present its draft genome of 4.11 Gb comprising 16,227 contigs and 13,238 scaffolds,with contig N50 of 3937 kb and scaffold N50 of 5675 kb in *** analysis shows that *** is a slow-evolving fish that diverged from elephant sharks about 96 million years ***,two gene families related to the immune system(immunoglobulin heavy constant delta genes and T-cell receptor alpha/delta variable genes)exhibit expansion in *** *** also identified the Hox gene clusters in *** and discovered that seven Hox genes shared by five representative fish species are missing in *** RNA sequencing data from *** and other three fish species demonstrate that fishes have a more diversified tissue expression spectrum when compared to *** functional studies suggest that lack of the gc gene encoding vitamin D-binding protein in cartilaginous fishes(both *** and Callorhinchus milii)could partly explain the absence of hard bone in their ***,this genome resource provides new insights into the niche adaptation,body plan,and skeleton formation of ***,as well as the genome evolution in cartilaginous fishes.

阜阳市是农业大市,农作物常年种植面积123万hm2,大宗农作物主要有小麦、玉米、大豆、水稻、薯类等,由此产生的秸秆资源储量十分丰富。本研究以主要种植作物为研究对象,通过查阅阜阳市统计年鉴、政府工作报告和公开发表文献资料,系统估算和分析了阜阳市农业秸秆资源量、养分供应潜力、空间分布特征和主要利用方式,为合理利用秸秆养分资源,实现“减施化肥”和“化肥零增长目标”提供参考。结果表明,全市秸秆有机质资源量为370.62万t,氮、磷、钾总养分为64.82万t秸秆还田养分贡献率约20%,建议秸秆还田后下茬作物化肥施用量减少20%。

耐高温油脂降解菌株的筛选、鉴定及其在好氧堆肥中的应用

针对餐厨垃圾油脂含量高抑制堆肥腐熟的问题,从餐厨垃圾高温期堆肥样品中采集样品,以大豆油为唯一碳源,进行富集驯化、划线分离获得菌株,并经中性红培养基筛选、耐高温筛选、油脂降解能力测定培养基油脂降解率验证,采用16S rDNA对菌株进行初步鉴定,并将菌株以1%(体积分数)接种至餐厨垃圾中进行21 d的好氧堆肥。结果表明:筛选获得2株耐高温油脂降解菌分别记为HBT-2、HBT-12,分别归属于枝芽孢杆菌属(Virgibacillus)和芽孢杆菌属(Bacillus),最高耐受温度分别为50、60℃,油脂降解率分别为30.40%和27.45%。复配后接种组油脂降解率为38.62%,发芽指数(GI)为85.90%,达到腐熟标准(参考《有机肥料》(NY 525—2012));对照组油脂降解率为26.51%,GI为76.68%,未达到腐熟标准。接种耐高温油脂降解菌可以促进餐厨垃圾堆肥中油脂的降解,加快堆肥腐熟进程,为基于生物强化技术的餐厨垃圾资源化处理工艺奠定基础。

Among the four prostaglandin E2 receptors,EP3 receptor is the one most abundantly expressed in white adipose tissue(WAT).The mouse EP3 gene gives rise to three isoforms,namely EP3α,EP3β,and EP3γ,which differ only at their C-terminal *** date,functions of EP3 receptor and its isoforms in WAT remain incompletely *** this study,we found that the expression of all EP3 isoforms were downregulated in WAT of both db/db and high-fat diet-induced obese *** ablation of three EP3 receptor isoforms(EP3^(−/−)mice)or EP3αand EP3γisoforms with EP3βintact(EP3βmice)led to an obese phenotype with increased food intake,decreased motor activity,reduced insulin sensitivity,and elevated serum *** the differentiation of preadipocytes and mouse embryonic fibroblasts to adipocytes was markedly facilitated by either pharmacological blockade or genetic deletion/inhibition of EP3 receptor via the cAMP/PKA/PPARγpathway,increased adipogenesis may contribute to obesity in EP3^(−/−)and EP3β***,both EP3^(−/−)and EP3βmice had increased lipolysis in WAT mainly due to the activated cAMP/PKA/hormone-sensitive lipase *** together,our findings suggest that EP3 receptor and itsαandγisoforms are involved in both adipogenesis and lipolysis and influence food intake,serum lipid levels,and insulin sensitivity.

Effect of Multiple Forging on Microstructure and Mechanical Properties of Powder Metallurgy FGH4096 Superalloy

FGH4096 is an important damage-tolerance-type powder metallurgy （PM） superalloy, which is also the optimized turbine material for high performance aero-engine. To turbine disc, bore and rim are used in high temperature-low stress condition and low temperature-high stress condition, and dual-microstructure is introduced. Coarse-grained microstructure is suit for rim region and fine-grained is no doubt most suitable for bore region. But fine-grained ingot can not make by PM technique, so that gradient heat treatment can not be developed to coarsen the grain of rim region. Under the confine of PM technique, several treatment processir, g should be taken place to refine the coarse grains of PM alloy. Multiple forging with large accumulate deformation was carried cat on PM FGH4096 superalloy, and fine grains of 3.5 μm were obtained by this treatment processing. Mechanical properties testing were ake carried out on both fine-grained and other statuses alloys, Microstrucmre evolution was observed by optical microscopy, scanning electron microscopy and transmission electron microscopy. As a result of repeated dynamic recrystallizafion, coarse grains could be completely refined into fine grains. And the increase of strength was mainly attributed to refinement effect and improvement of defects such as previous particle boundaries. Successful development of fine-grained treatment processing will promote the research of dual-microstructure turbine disc.

神经突起生长导向因子(Netrin)蛋白家族是分泌型蛋白质,其通过与不同的受体相互作用,在神经发育过程中起轴突导向和细胞迁移中发挥着双重导向功能.本文分别对涉及到细胞相互作用的Netrin基因家族的起源与进化进行研究.以进化史不同阶段的代表物种作为研究对象,通过使用MEGA的邻接法和似释然法分别对Netrin基因家族构建系统发育树;使用PAML 4.7工具以核苷酸序列作为分析材料进行选择压分析,筛选出发生正选择作用的位点;使用DIVERGE3.0软件对Netrin氨基酸序列进行功能分化分析,筛选出Ⅰ型和Ⅱ型功能分化位点.

Automatic Keyphrase Extraction from Scientific Chinese Medical Abstracts Based on Character-Level Sequence Labeling

Purpose:Automatic keyphrase extraction(ake)is an important task for grasping the main points of the *** this paper,we aim to combine the benefits of sequence labeling formulation and pretrained language model to propose an automatic keyphrase extraction model for Chinese scientific ***/methodology/approach:We regard ake from Chinese text as a character-level sequence labeling task to avoid segmentation errors of Chinese tokenizer and initialize our model with pretrained language model BERT,which was released by Google in *** collect data from Chinese Science Citation Database and construct a large-scale dataset from medical domain,which contains 100,000 abstracts as training set,6,000 abstracts as development set and 3,094 abstracts as test *** use unsupervised keyphrase extraction methods including term frequency(TF),TF-IDF,TextRank and supervised machine learning methods including Conditional Random Field(CRF),Bidirectional Long Short Term Memory Network(BiLSTM),and BiLSTM-CRF as *** are designed to compare word-level and character-level sequence labeling approaches on supervised machine learning models and BERT-based ***:Compared with character-level BiLSTM-CRF,the best baseline model with F1 score of 50.16%,our character-level sequence labeling model based on BERT obtains F1 score of 59.80%,getting 9.64%absolute *** limitations:We just consider automatic keyphrase extraction task rather than keyphrase generation task,so only keyphrases that are occurred in the given text can be *** addition,our proposed dataset is not suitable for dealing with nested *** implications:We make our character-level IOB format dataset of Chinese Automatic Keyphrase Extraction from scientific Chinese medical abstracts(Cake)publicly available for the benefits of research community,which is available at:https://***/possible1402/***

Genome Wide Association Study: Searching for Genes Underlying Body Mass Index in the Chinese

Objective Obesity is becoming a worldwide health problem. The genome wide association （GWA） study particularly for body mass index （BMI） has not been successfully conducted in the Chinese. In order to identify novel genes for BMI variation in the Chinese, an initial GWA study and a follow up replication study were performed. Methods Affymetrix 500K SNPs were genotyped for initial GWA of 597 Northern Chinese. After quality control, 281 533 SNPs were included in the association analysis. Three SNPs were genotyped in a Southern Chinese replication sample containing 2 955 Chinese Han subjects. Association analyses were performed by Plink software. Results Eight SNPs were significantly associated with BMI variation after false discovery rate （FDR） correction （P=5.45×10-7-7.26×106, FDR q=0.033-0.048）. Two adjacent SNPs （rs4432245 ＆ rs711906） in the eukaryotic translation initiation factor 2 alpha kinase 4 （EIF2AK4） gene were significantly associated with BMI （P=6.38×10-6＆ 4.39×106, FDR q=0.048）. In the follow-up replication study, we confirmed the associations between BMI and rs4432245, rs711906 in the EIF2ake gene （P=0.03 ＆ 0.01, respectively）. Conclusion Our study suggests novel mechanisms for BMI, where EIF2AK4 has exerted a profound effect on the synthesis and storage of triglycerides and may impact on overall energy homeostasis associated with obesity. The minor allele frequencies for the two SNPs in the EIF2AK4 gene have marked ethnic differences between Caucasians and the Chinese. The association of the EIF2AK4 gene with BMI is suggested to be ＇ethnic specific＇ in the Chinese.
