Isolation, purification and characterization of an N-acetyl-D-lactosamine binding mitogenic and anti-proliferative lectin from tubers of a cobra lily Arisaema utile Schott

Lectins are the carbohydrate-binding proteins of non-immune origin which have been the subject of intense investigation over the last few decades owing to the variety of interesting biological properties. Most of the lectins which have been purified and characterized from plants have been obtained from dicotyledons. In the present study a lectin was purified from tubers of a monocot plant Arisaema utile (AUL) Schott by affinity chromatography on asialofetuin-linked amino activated silica beads. AUL gave a single band in SDS-PAGE at pH 8.3 corresponding to subunit Mr 13.5 kDa. The native molecular mass of AUL was 54 kDa suggesting a homotetrameric structure. AUL gave multiple bands in isoelectric focusing and in native PAGE at pH 8.3. AUL was inhibited by N-acetyl-D-lactosamine (Lac NAc), a disaccharide and asialofetuin, a complex desialylated serum glycoprotein. When treated with denaturing agents, the lectin was stable in the presence of urea (3 M), thiourea (4 M) and guanidine HCl (4 M). AUL was a glycoprotein with a carbohydrate content of 1.2%. Complete loss of activity was observed upon modification of tryptophan residues of the lectin. The activity was reduced to 25% after modification of tyrosine. Chemical modification of arginine, histidine, serine and cysteine residues of AUL did not affect its activity. Using Far UV CD spectra the estimated secondary structure was 37% α-helix, 25% β-sheet and 38% random contributions. The lectin showed potent mitogenic response towards human lymphocytes. In vitro anti-proliferative assay using 11 human cancer cell lines resulted in 50% inhibition of six cell lines viz. SW-620, HCT-15, SK-N-SH, IMR-32, Colo-205 and HT-29 at 38, 42, 43, 49, 50 and 89 µg/ml, respectively.

Software Development Life Cycle (SDLC) is one of the major ingredients for the development of efficient software systems within a time frame and low-cost involvement. From the literature, it is evident that there are various kinds of process models that are used by the software industries for the development of small, medium and long-term software projects, but many of them do not cover risk management. It is quite obvious that the improper selection of the software development process model leads to failure of the software products as it is time bound activity. In the present work, a new software development process model is proposed which covers the risks at any stage of the development of the software product. The model is named a Hemant-Vipin (HV) process model and may be helpful for the software industries for development of the efficient software products and timely delivery at the end of the client. The efficiency of the HV process model is observed by considering various kinds of factors like requirement clarity, user feedback, change agility, predictability, risk identification, practical implementation, customer satisfaction, incremental development, use of ready-made components, quick design, resource organization and many more and found through a case study that the presented approach covers many of parameters in comparison of the existing process models. .

Data security is a very important part of data transmission over insecure channels connected through high-speed networks. Due to COVID-19, the use of data transmission over insecure channels has increased in an exponential manner. Hybrid cryptography provides a better solution than a single type of cryptographical technique. In this paper, nested levels of hybrid cryptographical techniques are investigated with the help of Deoxyribonucleic Acid (DNA) and Paillier cryptographical techniques. In the first level, information will be encrypted by DNA and at the second level, the ciphertext of DNA will be encrypted by Paillier cryptography. At the decryption time, firstly Paillier cryptography will be processed, and then DAN cryptography will be processed to get the original text. The proposed algorithm follows the concept of Last Encryption First Decryption (LEFD) at the time of decryption. The computed results are depicted in terms of tables and graphs.

Software Defect Prediction Using Hybrid Machine Learning Techniques: A Comparative Study

When a customer uses the software, then it is possible to occur defects that can be removed in the updated versions of the software. Hence, in the present work, a robust examination of cross-project software defect prediction is elaborated through an innovative hybrid machine learning framework. The proposed technique combines an advanced deep neural network architecture with ensemble models such as Support Vector Machine (SVM), Random Forest (RF), and XGBoost. The study evaluates the performance by considering multiple software projects like CM1, JM1, KC1, and PC1 using datasets from the PROMISE Software Engineering Repository. The three hybrid models that are compared are Hybrid Model-1 (SVM, RandomForest, XGBoost, Neural Network), Hybrid Model-2 (GradientBoosting, DecisionTree, LogisticRegression, Neural Network), and Hybrid Model-3 (KNeighbors, GaussianNB, Support Vector Classification (SVC), Neural Network), and the Hybrid Model 3 surpasses the others in terms of recall, F1-score, accuracy, ROC AUC, and precision. The presented work offers valuable insights into the effectiveness of hybrid techniques for cross-project defect prediction, providing a comparative perspective on early defect identification and mitigation strategies. .

The present work investigates the effect of ultrasonic power(%)and the time of ultrasonic vibration on the sintered density and ultimate compressive strength(UCS)of Mg15Nb3Zn1Ca fabricated using ultrasonic assisted conventional sintering(UACS).The customized UACS setup was designed and manufactured to conduct the experimentations.A customized ultrasonic stepped horn assembly was used for providing vibrations to the sample during ***,to evaluate the efficacy of ultrasonic vibration parameters,the developed setup was used to sinter Mg15Nb3Zn1Ca composite *** study unveiled an increased sintered density and UCS of the fabricated sample by the increase in ultrasonic power(%).Moreover,a decrease in sintered density and UCS was observed with an increase in the time of ultrasonic vibration beyond a certain *** sintered with the assistance of ultrasonic vibration at 100%ultrasonic power,and 20 min of ultrasonic vibration resulted in a sintered density of 1.928±0.062 g·cm^(-3)and UCS of 234.9±12.3 *** obtained mechanical properties of the fabricated sample were comparable to the properties of cortical *** surface morphology and elemental compositions of samples fabricated using UACS declared a fair dispersion of reinforcement in the matrix containing merely the source *** results of corrosion test have showed that the assistance of ultrasonic vibration suppressed the degradation behaviour of the sintered sample after performing electrochemical study of samples using 3-electorde cell ***15Nb3Zn1Ca fabricated using UACS showed a 50.18%and 9.08%of reduction in corrosion rate over conventionally sintered pure Mg and Mg15Nb3Zn1Ca *** addition,electrochemical impedance spectroscopy(EIS)results indicated an enhanced corrosion resistance of the Mg15Nb3Zn1Ca composite material when fabricated at 100%of ultrasonic power with 20 min of vibration *** from that,electrochemical equivalent circuits also result

It is well recognized that the voltage stability of a power system is affected by the load model and hence, to effectively analyze the reactive power compensation of an isolated hybrid wind-diesel based power system, the loads need to be considered along with the generators in a transient analysis. This paper gives a detailed mathematical modeling to compute the reactive power response with small voltage perturbation for composite load. The composite load is a combination of the static and dynamic load model. To develop this composite load model, the exponential load is used as a static load model and induction motors （IMs） are used as a dynamic load model. To analyze the dynamics of IM load, the fifth, third and first order model of IM are formulated and compared using differential equations solver in Matlab coding. Since the decentralized areas have many small consumers which may consist large numbers of IMs of small rating, it is not realistic to model either a single large rating unit or all small rating IMs together that are placed in the system. In place of using a single large rating IM, a group of motors are considered and then the aggregate model of IM is developed using the law of energy conservation. This aggregate model is used as a dynamic load model. For different simulation studies, especially in the area of voltage stability with reactive power compensation of an isolated hybrid power system, the transfer function AQ/AV of the composite load is required. The transfer function of the composite load is derived in this paper by successive derivation for the exponential model of static load and for the fifth and third order IM dynamic load model using state space model.
