A selective and rapid high-performance liquid chromatography–tandem mass spectrometry method was developed and validated for the quantification of raltegravir using raltegravir-d3 as an internal standard（IS）. The analyte and IS were extracted with methylene chloride and n-hexane solvent mixture from 100 mL human plasma. The chromatographic separation was achieved on a Chromolith RP-18 e endcapped C18（100 mm 4.6 mm） column in a run time of 2.0 min. Quantitation was performed in the negative ionization mode using the transitions of m/z 443.1-316.1 for raltegravir and m/z 446.1-319.0 for IS. The linearity of the method was established in the concentration range of 2.0–6000 ng/m *** mean extraction recovery for raltegravir and IS was 92.6% and 91.8%, respectively, and the IS-normalized matrix factors for raltegravir ranged from 0.992 to 0.999. The application of this method was demonstrated by a bioequivalence study on 18 healthy subjects.

BACKGROUNd Around 1 million cases of medical termination of pregnancy(MTP)take place yearly in the United States of America with around 2 percent of this population developing *** cardiovascular(CVd)complications occurring post MTP or after stillbirth is not very well *** To help the reader better understand,prepare,and manage these complications by reviewing various cardiac comorbidities seen after *** We performed a literature search in PubMed,Medline,RCA,and google scholar,using the search terms“abortions”or“medical/legal termination of pregnancy”and“cardiac complications”or“cardiovascular complications”.RESULTS The most common complications described in the literature following MTP were infective endocarditis(IE)(n=16),takotsubo cardiomyopathy(TTC)(n=7),arrhythmias(n=5),and sudden coronary artery dissection(SCAd)(n=4).The most common valve involved in IE was the tricuspid valve in 69%(n=10).The most observed causative organism was group B Streptococcus in 81%(n=12).The most common type of TTC was apical type in 57%(n=4).Out of five patients de veloping arrhythmia,bradycardia was the most common and was seen in 60%(3/5)of the *** four cases of SCAd-P type presented as acute coronary syndrome 10-14 d post termination of pregnancy with predominant involvement of the right coronary *** was only reported following IE in 6.25%.Clinical recovery was reported consistently after optimal medical management following all these *** In conclusion,the occurrence of CVd complications following pregnancy termination is infrequently documented in the existing *** this review,the most common CVd complication following MTP was noted to be IE and TTC.

For better rock mass characterization and support design, 3d engineering geological mapping was carried for the heading portion of the under construction 200.00 m long, 68.75 m high and 20.20 m wide underground additional surge pool cavern of a Pranahitha-Chevella Sujala Sravanthi lift irrigation scheme package 8, India. To study cavern behavior, 3d geologic mapping of heading portion is very important for large cavern for predicting geologic conditions in benching down up to invert level, planning support system, selecting inclination for best location of supplemental rock bolt and choosing strategic locations for various types of instrumentation. The assessment of Tunnel Quality Index “Q” and Geomechanics classification for the granitic rock mass was done based on the information available of the rock joints and their nature and 3d geological logging. Hoek-Brown parameters were also determined by the statistical analysis of the results of a set of triaxial tests on core samples. On basis of geological characteristics and NMT Q-system chart, support system is recommended which includes rock bolt, steel fibre reinforced shotcrete and grouting. To evaluate the efficacy of the proposed support system, the capacity of support system is determined.

AIM: The pathogenesis of occurrence of liver inflammation and fibrosis in patients with nonalcoholic steatohepatitis (NASH) is not completely understood. Other than insulin resistance, iron abnormalities have been thought to be one of the triggering factors. Therefore, our aim was to study the role of iron abnormalities and HFE gene mutations in patients with NASH. METHOdS: Thirty-one patients of NASH diagnosed on the basis of clinical examination biochemistry, ultrasonography and liver biopsy (n = 14) were included in the study. Serum iron parameters (n = 23) (iron, ferritin, total iron-binding capacity and transferrin saturation), Perls' iron staining on liver biopsies (n = 14) and HFE gene mutations (C282Y and H63d) (n = 16) were studied in these patients. The association between iron staining, necroinflammatory activity and fibrosis stage on liver biopsies was also determined. RESULTS: Elevated serum iron, ferritin and transferrin saturation above 55% were observed in 4.3% of patients. On histology, 71% of the patients had negative iron staining, 21.4% had 1+ staining, 7.2% had 2+ staining and none had 3+ or 4+ staining. There was no association between the degree of iron staining and necroinflammatory activity (P=0.55) and fibrosis stage (P= 0.09) on histology. None of the patients had C282Y HFE gene mutation and four patients (25%) were found to be heterozygotes for H63d gene mutation. CONCLUSION: Our study does not favor iron overload and HFE gene mutations as major factors in the pathogenesis of NASH in Asian Indians.

Arabidopsis cryptochrome 2 （CRY2） is a blue light receptor that mediates light inhibition of hypocotyl elongation and long-day promotion of floral initiation, CRY2 is known to undergo blue light-dependent phosphorylation, which is believed to serve regulatory roles in the function of CRY2. We report here on a biochemical and genetics study of CRY2 phosphorylation. Using mass spectrometry analysis, we identified three serine residues in the CCE domain of CRY2 （S588, S599, and S605） that undergo blue light-dependent phosphorylation in Arabidopsis seedlings. A study of serine-substitution mutations in the CCE domain of CRY2 demonstrates that CRY2 contains two types of phosphorylation in the CCE domain, one in the serine cluster that causes electrophoretic mobility upshift and the other outside the serine cluster that does not seem to cause mobility upshift. We showed that mutations in the serine residues within and outside the serine cluster diminished blue light-dependent CRY2 phosphorylation, degradation, and physiological activities. These results support the hypothesis that blue light-dependent phosphorylation of the CCE domain determines the photosensitivity of Arabidopsis CRY2.

AIM:To identify the frequency of iron overload and study the three mutations in the HFE gene (C282Y,H63d,and S65C) in patients with chronic liver disorders (CLd) and controls. METHOdS:To identify patients with iron overload (transferrin saturation > 45% in females and > 50% in males and serum ferritin > 1000 ng/mL) we evaluated 236 patients with CLd,including 59 with non-alcoholic steatohepatitis (NASH),22 with alcoholic liver disease (ALd),19 of cirrhosis due to viruses (HBV,HCV),and 136 with cryptogenic cirrhosis. Mutations of the HFE gene were analyzed by PCR-RE. hundred controls were screened for iron status and the mutations. RESULTS:Seventeen patients with CLd showed evidence of iron overload. Fifteen cases of iron overload had cryptogenic cirrhosis and two had ALd. None of the controls showed iron overload. We did not find any individual with 282Y or 65C either in the cases or in the controls. The prevalence of H63d heterozygosity was 12% in normal individuals,14.8% in 236 patients (16.9% in NASH,13.6% in ALd,26.3% in viral and 12.5% in cryptogenic cirrhosis) and the overall prevalence was 13.98%. Only two of the 17 patients with primary iron overload were heterozygous for H63d. One patient with NASH and one normal individual who were homozygous for H63d showed no iron ***:Primary iron overload in Indians is nonHFE type,which is different from that in Europeans and further molecular studies are required to determine the defect in various iron regulatory genes.
