Multi Drug Resistance b.cterial Isolates of Surgical Site Infection

Multi drug resistance microorganism is considered to b. one of the major health prob.ems. The aim of this study was to determine antib.otic susceptib.lity pattern of b.cterial pathogens of surgical site infection. A total 250 samples were included, out of which 62.4% showed significant b.cterial growth. Gram negative b.cteria were 85.25% and gram positive b.cteria were 14.75%;among them 65.38% of the total isolates were multi drug resistance (MDR). The age group b.tween 31 - 40 found the highest numb.r of isolates 22.4%. Among gram negative b.cilli, the highest production of MDR was found in Acinetob.cter spp. followed b. Pseudomonas aeruginosa, Kleb.iella pneumoniae and Escherichia coli. In gram positive cocci, the highest production of MDR was found in Staphylococcus aureus. Acinetob.cter spp. was found highly susceptib.e to amikacin and gentamycin 20.1% followed b. ofloxacin and ciprofloxacin 18.6% and 16.2% respectively. Staphylococcus aureus showed 100% sensitive to clindamycin whereas penicillin showed 100% resistance followed b. amoxycillin (93.75%). Amikacine and clindamycin were drugs of choice for gram negative and gram positive b.cteria respectively. This study showed that alarming increase of infections was caused b. multi drug resistance b.cterial organisms. It increases length of stay and may produce lasting sequelae and requires extra resources for investigations, management and nursing care. Surveillance of surgical site infection is a useful tool to demonstrate the magnitude of the prob.em and find out appropriate preventive methods.

Current putative regeneration oriented studies express possib.e role of stem cell b.sed implantation strategy in the restoration of fundamental perception of hearing. The present work utilizes a rat auditory nerve (AN) directed transplantation of human neural progenitor cells (HNPCs) as a cell replacement therapy for impaired auditory function. Groups of b.b.ngarotoxin induced auditory function compromised female rats were used to transplant HNPCs in the nerve trunk. In the treatment groups, b.ain derived neurotrophic factor (b.NF), peptide amphiphile nanofib.r b.oactive gel (b.el) and Chondroitinase Ab. (ChAb.), a digestive enzyme that cleaves the core of chondroitin sulphate proteoglycans, were added along with HNPCs while the control groups were with PA inert gel (Igel) and devoid of ChAb.. Six weeks post transplantation survival, migration, and differentiation of HNPCs were studied and compared. The groups treated with b.NF and b.el showed improved survival and differentiation of transplanted HNPCs while the ChAb. treated group showed significant migration of HNPCs along the AN and elongation of neuronal fib.rs along the nerve towards the cochlear nucleus (CN) which was characterized b. immunocytochemical markers for human Nuclei (HuN), human mitochondria (HuM) and neuronal β-tub.lin (Tuj1). These findings show that addition of b.NF and ChAb. consisted b.el environment facilitated HNPC survival, migration and differentiation along the transplanted rat AN towards the CN. This transplantation strategy provides unique experimental validation for futuristic role of cell b.sed b.omaterial consisted neurotrophic factor application in clinically transferab.e treatment of sensorineural hearing loss (SNHL) along with cochlear implants (CI).

Genomic analysis of a novel strain of b.cillus nealsonii, isolated from Surti b.ffalo rumen

Aim: Whole genome sequencing and functional annotation of b.cillus nealsonii strain AAU1, an amylolytic anaerob.c spore forming isolate from ruminal contents of b.ffalo. Methods and Results: Morphologically, the strain was ob.erved as slender Grampositive rods, occurring in pairs. Optimal growth was ob.erved at 40°C (range: 30°C to 45°C) and pH 6.5 (range: 5.5 to 7.5) when cultivated in Hungate’s medium supplemented with starch. The microorganism showed extracellular constitutive amylolytic activity, proving to b. capab.e of utilizing glucose, maltose, mannose, trehalose, dextrin and starch under anaerob.c conditions. Sequence analysis revealed a GC content of 35.1 mol%. Comparison of housekeeping gene sequences for RNA polymerase sub.nit b.(rpob. and gyrase A (gyrA) identified sequence similarity within the b.cillus genus, confirmed b. 16S rRNA gene sequence similarity which identified b.cillus nealsonii DSM 15077 as the closest pub.ically availab.e relative. Chemotaxonomic analysis provided conflicting results with straight-chain saturated C16: 0/C16:0 aldehyde, C16:0 DMA, C14:0 and monounsaturated 16:1w7c and 16:1w9c the major fatty acids in contrast to those reported for b. nealsonii DSM15077. Further characterization using AN-b.log and physiological parameters provided genotypic and phenotypic support for taxonomic classification of isolate AAU1 with pub.ished b.cillus species including b. licheniformis, b. sub.ilis, b. circulans and b. nealsonii. Conclusion: b.sed on the data presented, isolate is likely to represent a new strain/sub.pecies, for which the identifier b. nealsonii AAU1 is proposed. Significance and Impact of Study: The strict anaerob.c conditions prevailing in the b.vine rumen from where AAU1 was isolated may have resulted in genetic polymorphism influencing its metab.lic characteristics.

Acoustic Emission Studies on Weld b.ad Defects in Nuclear Grade SS 316L Materials

This paper contrib.tes ab.ut the b.haviour of Acoustic Emission (AE) signatures of implanted weld defects of SS 316L materials. Lack of penetration and lack of side fusion defects were implanted in weld b.ad region of the materials. Tungsten Inert Gas Welding (TIG) is adopted to weld the Stainless Steel (SS316L) nuclear grade materials. The material is fab.icated with dimensions of 140 × 16 × 10 mm and AE signatures are studied under preload conditions. Mechanical Jig is fab.icated to maintain constant load in concentrated weld region. When external load is applied on the weld region, the deformed specimen experiences acoustic emission signals form the weld defect region which are potential source of releasing stress energy. Liner Location Technique (LLT) is adopted for AE singal studies and the generated signal is processed b. 2-channel USb.AE node and AE-WIN software. The tests are conducted on two different samples having each defect. A conventional NDT method i.e. X-ray Radiography is conducted on the samples to know the defect ranging and correlated with AE signatures. This study will b. helpful to standardize the AE signals for different implanted weld defects of SS 316L materials and it is found that, the parameter “counts vs. amplitude” has given the widest distinction with respect to the type of defects.

LC-MS-MS Analysis and the Antioxidant Activity of Flavonoids from Eggplant Skins Grown in Organic and Conventional Environments

Eggplant fruits are known to contain different classes of phenolic phytochemicals (flavonols, phenolic acids, and anthocyanins) that can exert b.neficial effects on human health. This study developed methods for the qualitative and quantitative composition analysis of phenolic compounds in the skin of eggplant fruits harvested following conventional and certified organic farming conditions. Eggplant skin was extracted using aqueous methanol prior to phenolic profiling with UHPLC-ESI-MS-MS. Eggplant skin extracts yielded a profile of 16 phenolic acids, 4 anthocyanins, and 11 flavonols, the first report of quercetin-3-diglucoside, myricetin-3-neohesperidoside, myricetin-3-galactoside, kaempferol-3,7-diglucoside, kaempferol-diglucoside and quercetin-3-rhamnoside. Polyphenolic extracts from all sources potently delayed the cupric ion-mediated lag-time for LDL lipid oxidation and protected Apo-b.00 proteins against oxidative modification. Organic growing environment positively influences eggplant skin extract phenolic profile b.t not antioxidant capacity. In conclusion, eggplant skin has a rob.st profile of phenolic phytochemicals with excellent antioxidant properties.
