Charcoal Nanoparticles as a Delivery System for Doxorubicin and Sorafenib in Treatment of Hepatocellular Carcinoma

Background: Hepatocellular carcinoma (HCC) is the most common type of liver cancer and one of the leading causes of cancer-related death worldwide. Advanced HCC displays strong resistance to chemotherapy, and traditional chemotherapy drugs do not achieve satisfactory therapeutic efficacy. The delivery of therapeutic compounds to the target site is a major challenge in the treatment of many diseases. Objective: This study aims to evaluate activated charcoal nanoparticles as a drug delivery system for anticancer agents (Sorafenib and Doxorubicin) in Hepatocellular Cancer Stem Cells. Method: The percent efficiency of entrapment (% ee) of the doxorubicin and sorafenib entrapped onto the activated charcoal was obtained by determining the free doxorubicin and sorafenib concentration in the supernatant-prepared solutions. Then the characterizations of nanoparticles were formed by determination of the particle size distribution, zeta potential, and polydispersity index (PDI). The anticancer activity of activated Charcoal, Doxorubicin-ACNP, sorafenib-ACNP, free doxorubicin, and free sorafenib solutions was measured based on cell viability percentage in HepG2 cell lines (ATCC-CCL 75). In vitro RBC’s toxicity of Doxorubicin/sorafenib loaded charcoal was estimated by hemolysis percentage. Results: The synthesized Doxorubicin-ACNP and Sorafenib-ACNP were evaluated and their physiochemical properties were also examined. essentially, the percent efficiency of entrapment (ee %) was found to be 87.5% and 82.66% for Doxorubicin-ACNP and Sorafenib-ACNP, respectively. The loading capacity was 34.78% and 24.31% for Doxorubicin-ACNP and Sorafenib-ACNP. Using the Dynamic Light scattering [DLS] for the determination of the hydrodynamic size and surface zeta potential, a narrow sample size distribution was obtained of (18, 68, and 190 nm for charcoal, 105, 255, and 712 nm for doxorubicin, and 91, 295, and 955 nm for sorafenib), respectively. 癌症是威胁人类生命中最严重的疾病之一。阻断程序性细胞死亡蛋白1(PD-1)和程序性死亡配体1(PD-L1)途径是近年来抑制癌细胞的重大创新之一,但只有少数抑制剂能够阻断。(2-甲基-3-联苯)甲醇(MBPM)衍生物就是其中之一。首先,建立了20个(2-甲基-3-联苯)甲醇(MBPM)衍生物作为程序性死亡配体-1(PD-L1)抑制剂的定量构效关系。采用密度泛函理论(DFT)在B3LPY/6-31+G(d,p)水平上研究了所选化合物的化学结构和性质。观察并测定了最高占据分子轨道能量(e)、最低未占据分子轨道能量(e)、总能量e、偶极矩DM、绝对硬度η、绝对电负性χ、柔软度S、亲电性ω、能隙Δe等。采用主成分分析(PCA)、多元线性回归(MLR)和多元非线性回归(MNLR)建立了QSAR模型。基于统计分析提出了定量模型和化合物的解释结果。MLR和MNLR的统计结果分别为0.661和0.758。对MLR和MNLR进行了单因素交叉验证(LOO-CV)、r指标、r检验和Golbraikh&Tropsha标准分析,结果表明,两个模型具有统计学意义,且在PD-L1的外部验证中数据变化稳定;MNLR模型比MLR模型更准确地预测了PD-L1抑制剂的生物活性。其次,我们设计了20种新型的MBPM卤化衍生物,在B3LYP/Midi X水平的DFT方法进行结构优化和频率计算,得到了这些化合物的Homo-Lumo能隙、硬度、柔软度和偶极矩。此外,对这些分子与PD-L1(PDB-ID 5j89)进行了3种不同的分子对接分析研究分子间的相互作用。进一步的,对29个蛋白质与5个候选蛋白进行了50 ns分子动力学模拟。分子动力学模拟表明,改性后的化合物比未改性的MBMP衍生物更有效,其中Tyr56、Asp122和Tyr123残基形成了氢键,并且这种氢键在药物-蛋白质相互作用中发挥重要作用。此外,化合物7在MD模拟和蛋白质数据库中的结果显示其与MBPM亲和力优异。强配体结合可能需要与Ala121、Met 115和Try123的残基的π-烷基相互作用,与Tyr56的残基的π-π堆积以及与残基Met 115的烷基相互作用。此外,ADMe/T分析表明,修饰的类似物毒性比母体化合物少。因此,卤素靶向抑制剂与PD-L1结合良好,有望成为治疗癌症的理想药物。最后,在B3LPY/6-31+G(d,p)水平上用DFT理论计算了100个合成的(2-甲基-3-联苯)甲醇(MBPM)衍生物或与PD-L1结合的小分子。Lipinski的五定律(RO5)被用来筛选所有优化的化合物。筛选后的化合物被进一步对接。利用Auto Dock-Vina和GOLD对接技术研究了这些抑制剂与PD-L1结合力。对M30、M38、M48、M51和M58 5种顶级抑制剂分子进行对接模拟结果表明M51能与PD-L1结合力最好。此外,A:Gln66、A:Tyr56、B:Asp122和B:Tyr123氨基酸参与了氢键的形成其他氨基酸A:Tyr56、A:Met 115、A:Ala121、B:Tyr56、B:Met115和B:Ala121强烈参与疏水作用。预测物质的吸收、分布、代谢、排泄、毒性和活性谱(PASS)结果表明比M51比其他四个分子更安全可靠,这说明M51很可能是一种非常有前途的抑制剂,可以与PD-L1结合,部分阻断PD-1/PD-L1。

1 BackgroundIn the United States, life expectancy is rising, particularly among the older population （〉 65 years）, with projected estimates approaching 20% by year 2050. Despite improved survival estimates, cardiovascular diseases remain the leading cause of morbidity and mortality resulting in substantial increase in health care cost. For younger adults, the evidence for secondary cardiovascular prevention is well established and corroborated by robust data. However, the application of risk reduction strategies in older populations remains an area of active debate. Many assume that vulnerability to chronic cardiovascular diseases is an inexorable part of aging such that the risks attributable to prevention outweigh potential benefits.

Integrated elements of early Warning Systems to enhance Disaster Resilience in the Arab Region

The Arab region＇s coastal zones are of immense importance. Almost all Arab countries need to build up resilience for vulnerable communities and carry out proactive planning for integrated coastal zone management and development of early warning systems for vulnerable coastal areas. It is clear that through the analysis of past natural disasters, the links between the four elements （risk knowledge, monitoring and warning service, dissemination and communication, response capability） of eWS （early warning systems） are weak, leading to a reduced effectiveness of the system. The focus is on the development of technology more than on members of the community at risk. There is also weakness in institutional cooperation and one of the main challenges is in early warning. This problem has encouraged the researcher to propose a guideline for early warning of natural disasters and increase its effectiveness by involving members of the community and improving institutional cooperation between stakeholders. The objective of this paper is to identify and assess some of the main areas vulnerable to sea level rise in the Arab world. A survey of case studies in the UAe （United Arab emirates） and egypt is undertaken. The main objectives are to identify vulnerabilities from coastal flooding, assess adaptation measures and point out the urgent need to build up institutional and human capacities in response to the problem, identify gaps, points of strength and points of weakness in early warning systems and discuss options for adaptation and the need for implementing major dements of the eWS.

Cancer is one of the most serious issues in human *** programmed cell death protein 1 and programmed death ligand-1(PD-L1)pathway is one of the great innovations in the last few years,a few numbers of inhibitors can be able to block it.(2-Methyl-3-biphenylyl)methanol derivative is one of ***,the quantitative structure-activity relationship(QSAR)established twenty(2-methyl-3-biphenylyl)methanol derivatives as the programmed death ligand-1 *** functional theory at the B3LPY/6-31+G(d,p)level was employed to study the chemical structure and properties of the chosen *** occupied molecular orbital energy eHOMO,lowest unoccupied molecular orbital energy eLUMO,total energy eT,dipole moment DM,absolute hardnessη,absolute electronegativityχ,softness S,electrophilicityω,energy gap?e,etc.,were observed and *** component analysis(PCA),multiple linear regression(MLR)and multiple nonlinear regression(MNLR)analysis were carried out to establish the *** proposed quantitative models and interpreted outcomes of the compounds were based on statistical *** results of MLR and MNLR exhibited the coefficient R^2 was 0.661 and 0.758,***-one-out cross-validation,r_m^2 metric,r_m^2 test,and"Golbraikh&Tropsha’s criteria"analyses were applied for the validation of MLR and MNLR,which indicate two models are statistically significant and well stable with data variation in the external validation towards *** obtained results showed that the MNLR model predicts the bioactivity more accurately than MLR,and it may be helpful and supporting for evaluation of the biological activity of PD-L1 inhibitors.

Objectives:effective and efficient communication is a core element in healthcare systems,especially between healthcare providers and *** study aimed to identify communication barriers be-tween nurses and patients in primary healthcare centers in ***:This is a cross-sectional study conducted across primary healthcare centers in *** hundred and two patients were recruited using convenience sampling.A self-administered question-naire comprising 29 items on communication barriers was ***:A total of 402 patients consented to *** majority of participants reported the following statements had large effects on communication:“shortage in the number of nurses compared to the large number of patients”(254/401,63.3%),“lack of desire of nurse to communicate with patients”(246/402,61.2%),and“negative attitude of the nurse toward the patient”(238/401,59.4%).Further,“difference in language between nurses and patients,”“lack of self-confidence by nurses,”and“nurses overwhelmed by work”were ranked as top three statements with a significant influence on commu-nication between nurses and ***:Communication between healthcare providers and patients is pivotal for an optimal healthcare *** on the findings of this study and the literature,we recommend formal training of health care workers in improving communication skills and including this not only in medical cur-riculum but also in the form of continuing medical education(CMes)1.

In city planning managing, the third dimension is becoming a necessity. Using 3D GIS modeling offers a flexible interactive system while providing one of the best visual interpretation of data which supports planning and decision processes for city planners. As a result, 3D GIS model expresses terrain features in an intuitive way which enhances the management and analysis of a proposed project through 3D visualization. This paper discusses the concept of 3D GIS modeling techniques using a simple procedure to generate a university campus model （real 3D GIS model） which will show the effectiveness of this approach. The 3D GIS model provides access to mapping data to support planning, design and data management. Intelligent GIS models and GIS tools help community planning and apply regional and discipline-specific standards. Integration of GIS spatial data with campus organization helps to improve quality, productivity and asset management. The following study built 3D GIS map and all utility information for AI al-Bayt University campus as an example. The primary objective is to improve data management （e.g., maps, plans, usage of facilities and services） and to develop methods using 3D spatial analysis for specific applications at the university.

evaluating Some Insecticides for Controlling the Sunn Pest eurygaster spp. Puton （Hemiptera： Scutelleridae） under Field Conditions

The sunn pest eurygaster integriceps is the most important insect of cereals in Iraq and other countries. In this study, the field efficacy of 10 different kinds of insecticides with various mode of action was evaluated against sunn pest e. integriceps infested wheat on field at middle oflraq. experiments were conducted in 11 wheat fields with each field 0.5 ha, located in the middle of Iraq （Wasit and Salahudain govemorates） during season 2015-2016. The wheat fields contained common varieties of wheat planted in Iraq. The population density of the pest was at its highest level （start of April 2015） of mostly nymphs, adults and eggs. The results indicated that the recommended dose for each insecticide used showed high efficacy （80.1%-93.8%） in reducing number of e. integriceps adults after one week of treatment, reaching 0.2-0.8 insects/m^2 compared to 3.6 insects/m2 in the control treatment. These results will assist the control program of this pest and in implementing pest management practices to reduce resistance development chances.
