Background Glioblastoma is characterized by highly infiltrative growth and invariably aggressive biological features. Despite combining surgery with radiotherapy and chemotherapy, the malignant nature and poor response to therapy of glioblastoma still presents a poor prognosis. Therefore, studies aimed at blocking glioblastoma infiltrative growth, promoting its drug sensitivity, and reducing its angiogenic potential are needed. A crucial form of cell–cell communication in tumors is the release and uptake of exosomes containing cellular proteins and RNAs. Cells take up exosomes delivering tumor-derived oncogenic factors and can a malignant phenotype in this *** was validated that exosomes are carriers of pro-tumorigenic factors that participate in glioblastoma progression. Fusion genes combine parts of ≥2 original genes, and can be generated from chromosomal rearrangement or abnormal transcription. Gene fusions have an important impact on the initial tumorigenesis and cancer progression *** gene fusions are strong driver mutations in neoplasia and are involved in tumorigenesis. Our RNA sequencing(RNA-seq) study of 272 gliomas revealed a novel, recurrent ZM fusion tran script in secondary *** glioblastomas or U87 cells with ZM fusion harbored more MET compared with those without ZM fusion,respectively. Endogenously expressed MET in U87 cells is not phosphorylated while exogenously expressed MET resulting from ZM fusion is phosphorylated. Clinically, survival of glioblastoma with ZM fusion is poorer than that without ZM fusion. However, whether gene fusions are transduced by exosomes is unknown. These studies reflect the need to evaluate the functional contribution of ZM fusion to the glioblastoma phenotype and its role in exosome-associated cell interaction with the tumor microenvironment. Methods We cloned a His-tagged version of CGGA475 ZM fusion into an adenovirus vector and stably expressed the fusion protein in a U87/ZM cell line.

Antimicrobial resistance(AMR) represents a substantial threat to global public health, complicating the treatment of common infections and leading to prolonged illness and escalated healthcare expenses. To effectively combat AMR, timely and accurate detection is crucial for AMR surveillance and individual-based therapy. Phenotypic antibiotic resistance testing(AST) has long been considered the gold standard in clinical applications, serving as the foundation for clinical AMR diagnosis and optimized therapy. It has significantly contributed to ensuring patients′ health and the development of novel antimicrobials. Despite advancements in automated culture-based AST technologies,inherent limitations impede the widespread use of phenotypic AST in AMR surveillance. Genotypic AST technologies offer a promising alternative option, exhibiting advantages of rapidity, high sensitivity, and specificity. With the continuous advancement and expanding applications of genotypic AST technologies, such as microfluidics, mass spectrometry, and high-resolution melting curve analysis, new vigor has been injected into the development and clinical implementation of genotypic AST technologies. In this narrative review, we discuss the principles, applications, and advancements of emerging genotypic AST methods in clinical settings. The comprehensive review aims to highlight the significant scientific potential of emerging genotypic AST technologies in clinical AMR diagnosis, providing insights to enhance existing methods and explore novel approaches.

Experimental Study on Geometry Characteristics of Turbulent Premixed Flames for Natural Gas/Air Mixtures

In this study,focusing on the geometry characteristics of spherical expanding flame,the turbulent premixed flames of natural gas/air mixtures were investigated in a fan-stirred turbulent *** effects of initial temperature（T=300-400 K）,initial pressure（P=0.1-0.3 MPa）,turbulence intensity（u'=1.0-2.7 m/s）,oxygen volumetric percentage(φ（O2）=15%-21%) and carbon dioxide volumetric percentage(φ（CO2）=0-20%)were delved *** flame profile under the Cartesian coordinate system was derived from the schlieren images taken by the high-speed ***,from both macroscopic and microscopic perspectives,the influence of experimental conditions on the flame geometry characteristics was explored through flame front extraction,wavelet decomposition and network *** results demonstrate that for significant flame wrinkling,changes in species concentrations and turbulence intensity have more pronounced effects on the flame wrinkling *** wrinkling of the flame front maintains a certain degree of similarity,as evidenced by the locally concentrated distribution of the angles of the maximum fluctuation *** disturbance energy under large-scale（D6-D8） disturbances exhibits relatively high values with a similar trend,exerting a significant impact on the geometry characteristics of the flame *** peaks of correlation degree are scattered either with the decomposition scale or the development of flame radius,indicating no linear correlation between different detail ***,the probability distribution of node degrees in key wrinkled regions exhibits different trends with that of large-scale wrinkling and disturbance energy,especially with changes in initial *** occurs because the number of key wrinkles varies based on the perturbation's strength or the region's ***,an increase in the fluctuation frequency of the flame's local radius may not necessarily lead to an increase in the number of key folded regions.

Inexact proximal gradient algorithm with random reshuffling for nonsmooth optimization

Proximal gradient algorithms are popularly implemented to achieve convex optimization with nonsmooth regularization. Obtaining the exact solution of the proximal operator for nonsmooth regularization is challenging because errors exist in the computation of the gradient; consequently, the design and application of inexact proximal gradient algorithms have attracted considerable attention from researchers. This paper proposes computationally efficient basic and inexact proximal gradient descent algorithms with random reshuffling. The proposed stochastic algorithms take randomly reshuffled data to perform successive gradient descents and implement only one proximal operator after all data pass through. We prove the convergence results of the proposed proximal gradient algorithms under the sampling-without-replacement reshuffling *** computational errors exist in gradients and proximal operations, the proposed inexact proximal gradient algorithms can converge to an optimal solution neighborhood. Finally, we apply the proposed algorithms to compressed sensing and compare their efficiency with some popular algorithms.

Brain-derived neurotrophic factor is a key factor in stress adaptation and avoidance of a social stress behavioral *** studies have shown that brain-derived neurotrophic factor expression in stressed mice is brain region–specific,particularly involving the corticolimbic system,including the ventral tegmental area,nucleus accumbens,prefrontal cortex,amygdala,and *** how brain-derived neurotrophic factor participates in stress processing in different brain regions will deepen our understanding of social stress *** this review,we discuss the expression and regulation of brain-derived neurotrophic factor in stress-sensitive brain regions closely related to the pathophysiology of *** focused on associated molecular pathways and neural circuits,with special attention to the brain-derived neurotrophic factor–tropomyosin receptor kinase B signaling pathway and the ventral tegmental area–nucleus accumbens dopamine *** determined that stress-induced alterations in brain-derived neurotrophic factor levels are likely related to the nature,severity,and duration of stress,especially in the above-mentioned brain regions of the corticolimbic ***,BDNF might be a biological indicator regulating stress-related processes in various brain regions.

The influence of partitioned profiling design based on a large-pitch highly loaded cascade is studied by numerical *** partitioned profile is mainly composed of a pressure-side convex structure near the leading edge and a suction-side convex structure at the midstream and downstream sides of the *** influence of the change in the vertex axial position and peak value of the B-line on the secondary flow control is *** this paper,air(ideal gas) is selected as the flow *** average static pressure at the outlet and the average total temperature at the inlet are kept *** γ-θ is used as the turbulence *** results show that the pressure-side convex structure suppresses the spanwise and pitchwise migration of the inlet flow by adjusting the static pressure distribution of the flow field,so the development of the pressure-side leg of the horseshoe vortex is effectively *** suction-side convex structure adjusts the static pressure distribution of the flow field and increases the included angle between the cross-flow and suction surface,so the accumulation of low-momentum fluid,the development of a corner vortex and the flow separation at the trailing edge of the suction-side surface are all suppressed near the endwall-suction ***,the energy loss coefficient of the large-pitch highly loaded cascade is decreased from 0.0564 to 0.0485,representing a 25% reduction in secondary flow losses.

Multi-Stage Voltage Control Optimization Strategy for Distribution Networks Considering Active-Reactive Co-Regulation of Electric Vehicles

The high proportion of uncertain distributed power sources and the access to large-scale random electric vehicle(EV)charging resources further aggravate the voltage fluctuation of the distribution network,and the existing research has not deeply explored the EV active-reactive synergistic regulating characteristics,and failed to realize themulti-timescale synergistic control with other regulatingmeans,For this reason,this paper proposes amultilevel linkage coordinated optimization strategy to reduce the voltage deviation of the distribution ***,a capacitor bank reactive power compensation voltage control model and a distributed photovoltaic(PV)activereactive power regulationmodel are ***,an external characteristicmodel of EVactive-reactive power regulation is developed considering the four-quadrant operational characteristics of the *** optimization model of the distribution network is then constructed considering the time-series coupling constraints of multiple types of voltage regulators.A multi-timescale control strategy is proposed by considering the impact of voltage regulators on active-reactive EV energy consumption and PV energy ***,a four-stage voltage control optimization strategy is proposed for various types of voltage regulators with multiple time ***-objective optimization is solved with the improvedDrosophila algorithmto realize the power fluctuation control of the distribution network and themulti-stage voltage control *** results validate that the proposed voltage control optimization strategy achieves the coordinated control of decentralized voltage control resources in the distribution *** effectively reduces the voltage deviation of the distribution network while ensuring the energy demand of EV users and enhancing the stability and economic efficiency of the distribution network.
