Oxidative stress(OS) plays a major role in the gastrointestinal *** probiotics were reported to repress OS,few researches compared the antioxidant ability of different Bacillus strains and decipher the *** select a Bacillus strain with higher antioxidant capacity,we used HO to induce intestinal porcine epithelial cell 1(IPEC-1) OS *** most suitable HO concentration and incubation time were determined by the half lethal dose and Methyl Thiazolyl *** analysis was performed to choose a sensitive indicator for *** for the comparison of Bacillus,cells were divided into:control,Bacillus treatment,HO treatment and Bacillus pre-protection+HO *** were co-cultured with IPEC-1 for 3h in Bacillus and Bacillus pre-protection+HO ***,based on OS model,300 μmol/L HO were added into medium of HO and Bacillus pre-protection+HO treatments for another 12 *** and apoptosis gene expressions were detected to screen the target *** factor erythroid-derived 2 related factor 2(Nrf2)/Kelch-like ECH-associated protein 1(Keap1) pathway,reactive oxygen species(ROS) production,mitochondrial membrane potential(△ψm),apoptosis and necrosis were *** revealed that heme oxygenase-1(HO-1) gene expression had a positive correlation with HO ***,Bacillus amyloliquefaciens SC06(SC06)-meditated IPEC-1 showed the best antioxidant capacity though modulating Nrf2 phosphorylation.△ψm was elevated while ROS generation was reduced with SC06 pre-protection,resulting in decreased apoptosis and ***,HO-1 expression could be regarded as an OS *** regulation of Nrf2/Keap1 pathway and ROS production by SC06 are involved in alleviating OS of IPEC-1.

Echinacea purpurea is an indigenous North American purple cone flower used by North Americans for treatment of various infectious diseases and *** study investigated the effect of polysaccharide enriched extract of Echinacea purpurea(EE) on the polarization of *** results showed that 100 μg/mL of EE could markedly activate the macrophage by increasing the expression of CD80,CD86,and MHCII ***,EE upregulated the markers of classically activated macrophages(Ml) such as CCR7 and the production of IL-1β,IL-6,IL-12 p70,TNF-αand *** functional tests showed that EE enhanced the phagocytic and intracellular bactericidal activity of macrophage against ***,we demonstrated that JNK are required for EE-induced NO and M1-related cytokines ***,these results demonstrated that Echinacea purpurea can polarize macrophages towards Ml phenotype,which is dependent on the JNK signaling pathways.

Probiotics are increasingly applied in popularity in both humans and *** of research has revealed their beneficial effects,including the immune modulation in intestinal pathogens ***,cellular process that involves the delivery of cytoplasmic proteins and organelles to the lysosome for degradation and recirculation,is essential to protect cells against bacterial ***,the mechanism of probiotics-mediated autophagy and its role in the elimination of pathogens are still ***,we evaluated Bacillus amyloliquefaciens SC06(Ba)-induced autophagy and its antibacterial activity against Escherichia coli in RAW264.7 *** blotting and confocal laser scanning analysis showed that Ba stimulated autophagy in a dose-and time-dependent ***-induced autophagy was found to play a role in the elimination of intracellular bacteria when RAW264.7 cells were challenged with Escherichia coli(***).Ba induced autophagy by increasing the expression of Beclin1 and Atg5-Atg12-Atg16 complex,but not the AKT/mTOR signaling ***,Ba pretreatment attenuated the activation of JNK in RAW264.7 cells during *** infection,further indicating a protective role for probiotics via modulating macrophage *** above findings highlight a novel mechanism underlying the antibacterial activity of *** study enriches the current knowledge on probiotic-mediated autophagy,and provides a new perspective on the prevention of bacterial infection in intestine,which further the application of probiotics in food products.

解淀粉芽孢杆菌SC06对免疫抑制小鼠肠道细菌酶活性和肠黏膜屏障功能的影响

本试验通过连续3 d腹腔注射免疫抑制剂环磷酰胺（ cyclophosphamide,CTX）建立小鼠免疫抑制模型,研究解淀粉芽孢杆菌SC06对免疫抑制小鼠肠道细菌酶活性和肠黏膜屏障功能的影响。选取120只BALB/c小鼠（4周龄,雌性,体重10~16 g）,随机分成3组,每组4个重复,每个重复10只。对照组饲喂基础饲粮,免疫抑制组和芽孢杆菌组分别饲喂基础饲粮和添加解淀粉芽孢杆菌SC06的试验饲粮（活菌数为1＆#215;105 Cfu/g）,并均在试验第1~3天连续3 d腹腔注射CTX 80μg/g BW。试验期为60 d。结果表明：与对照组相比,免疫抑制组小鼠盲肠和小肠内容物中各细菌酶（α-葡萄糖苷酶、β-葡萄糖苷酶、β-葡萄糖苷酸酶、α-半乳糖苷酶和β-半乳糖苷酶）以及小肠灌洗液中二胺氧化物酶（DAO）活性极显著降低（P〈0.01）,小肠灌洗液中一氧化氮（NO）含量及诱导型一氧化氮合成酶（iNOS）活性极显著升高（P〈0.01）,小肠灌洗液中促炎细胞因子增殖诱导配体（APRIL）、干扰素-γ（IFN-γ）、白细胞介素-6（IL-6）和白细胞介素-12（IL-12）及抗炎细胞因子白细胞介素-10（IL-10）含量极显著升高（P〈0.01）,说明腹腔注射CTX后小鼠肠道菌群发生紊乱,肠上皮细胞遭到破坏,肠黏膜屏障功能受损。解淀粉芽孢杆菌SC06可极显著提高免疫抑制小鼠盲肠内容物中β-葡萄糖苷酸酶和α-葡萄糖苷酶及小肠内容物中β-葡萄糖苷酸酶活性（P〈0.01）,分别极显著和显著提高小肠灌洗液中髓过氧化物酶（MPO）和分泌型磷脂酶A2（sPLA2）活性（P〈0.01和P〈0.05）。以上结果表明解淀粉芽孢杆菌SC06可在一定程度上提高免疫抑制小鼠的肠道益生菌数量,但仅能部分恢复免疫抑制小鼠的肠黏膜屏障功能。
