Identification of an immune-related gene signature for predicting prognosis and immunotherapy efficacy in liver cancer via cell-cell communication

BACKGROUND Liver cancer is one of the deadliest malignant tumors *** has provided hope to patients with advanced liver cancer,but only a small fraction of patients benefit from this treatment due to individual *** immune-related gene signatures in liver cancer patients not only aids physicians in cancer diagnosis but also offers personalized treatment strategies,thereby improving patient survival *** several methods have been developed to predict the prognosis and immunotherapeutic efficacy in patients with liver cancer,the impact of cell-cell interactions in the tumor microenvir-onment has not been adequately *** To identify immune-related gene signals for predicting liver cancer prognosis and immunotherapy *** Cell grouping and cell-cell communication analysis were performed on single-cell RNA-sequencing data to identify highly active cell groups in immune-related *** active immune cells were identified by intersecting the highly active cell groups with B cells and T *** significantly differentially expressed genes between highly active immune cells and other cells were subsequently selected as features,and a least absolute shrinkage and selection operator(LASSO)regression model was constructed to screen for diagnostic-related *** genes that were selected more than 5 times in 10 LASSO regression experiments were included in a multivariable Cox regression ***,3 genes(stathmin 1,cofilin 1,and C-C chemokine ligand 5)significantly associated with survival were identified and used to construct an immune-related gene *** The immune-related gene signature composed of stathmin 1,cofilin 1,and C-C chemokine ligand 5 was identified through cell-cell *** effectiveness of the identified gene signature was validated based on experi-mental results of predictive immunotherapy response,tumor mutation burden analysis,immune cell infiltrat

The cohesive clustered satellite(CCS) concept integrates diverse satellite payloads from different orbits, such as communication, remote sensing, and navigation satellites, into mobile, geometrically stable clusters for efficient collaborative networking [1]. By aggregating, reconstructing, and orchestrating heterogeneous satellite resources, such as computation, communication, and storage [2], the CCS enables resource ultra-sharing and provides information-centric and goal-oriented intelligent on-demand services [3] for a range of emerging Earth observation and deep space exploration missions [4].

In recent years, tungsten disulfide(WS_(2)) and tungsten selenide(WSe_(2)) have emerged as favorable electrode materials because of their high theoretical capacity, large interlayer spacing, and high chemical activity;nevertheless, they have relatively low electronic conductivity and undergo large volume expansion during cycling, which greatly hinder them in practical applications. These drawbacks are addressed by combining a superior type of carbon material, graphene, with WS_(2) and WSe_(2) to form a WS_(2)/WSe_(2)@graphene *** materials have received considerable attention in electro-chemical energy storage applications such as lithium-ion batteries(LIBs), sodium-ion batteries(SIBs),and supercapacitors. Considering the rapidly growing research enthusiasm on this topic over the past several years, here the recent progress of WS_(2)/WSe_(2)@graphene nanocomposites in electrochemical energy storage applications is summarized. Furthermore, various methods for the synthesis of WS_(2)/WSe_(2)@graphene nanocomposites are reported and the relationships among these methods, nano/microstructures, and electrochemical performance are systematically summarized and discussed. In addition, the challenges and prospects for the future study and application of WS_(2)/WSe_(2)@graphene nanocomposites in electrochemical energy storage applications are proposed.

A distributed fiber sensor was fabricated by splicing two single-mode fibers(SMFs)using the few-mode fiber(FMF)technique.A Brillouin optical time domain analysis(BOTDA)system was developed to measure the sensor’s temperature and bending ***-mode and four-mode step FMFs were combined to splice the few-mode *** results indicate that the temperature response coefficients of the few-mode segment are only slightly higher than those of the connected single-mode segment,measuring at 1.13 MHz/℃and 1.12 MHz/℃,*** minimum bending radius for the sensor is 0.9 cm,and the four-mode bending response curve is superior to that of the two-mode one,proving that 4-SI-FMF offers better bending sensitivity.

Insights into the mechanism of L-malic acid on drip loss of chicken meat under commercial conditions

Background A deterioration in the meat quality of broilers has attracted much more attention in recent years.L-malic acid(MA)is evidenced to decrease meat drip loss in broilers,but the underlying molecular mechanisms are still ***’s also not sure whether the outputs obtained under experimental conditions can be obtained in a com-mercial ***,we investigated the effects and mechanisms of dietary MA supplementation on chicken meat drip loss at large-scale *** Results showed that the growth performance and drip loss were improved by MA *** metabolome revealed that L-2-aminoadipic acid,β-aminoisobutyric acid,eicosapentaenoic acid,and nicotinamide,as well as amino acid metabolism pathways connected to the improvements of meat quality by MA *** transcriptome analysis further indicated that the effect of MA on drip loss was also related to the proper immune response,evidenced by the enhanced B cell receptor signaling pathway,NF-κB signaling pathway,TNF signaling pathway,and IL-17 signaling *** We provided evidence that MA decreased chicken meat drip loss under commercial *** and transcriptome revealed a comprehensive understanding of the underlying ***,MA could be used as a promising dietary supplement for enhancing the water-holding capacity of chicken meat.

Effect of high-multiple water injection on rock pore structure and oil displacement efficiency

Experimental methods,including mercury pressure,nuclear magnetic resonance(NMR)and core(wateroil)displacement,are used to examine the effects of high-multiple water injection(*** injection with high injected pore volume)on rock properties,pore structure and oil displacement efficiency of an oilfield in the western South China *** results show an increase in the permeability of rocks along with particle migration,an increase in the pore volume and the average pore throat radius,and enhanced heterogeneity after high-multiple water *** with normal water injection methods,a high-multiple water injection is more effective in improving the oil displacement *** degree of recovery increases faster in the early stage due to the expansion of the swept area,and the transition from oil-wet to *** degree of recovery increases less in the late stage due to various factors,including the enhancement of heterogeneity in the *** both the economic aspect and the production limit of water flooding,it is recommended to adopt other technologies to further enhance oil recovery after 300 PV water injection.

ATAT1 deficiency enhances microglia/macrophage-mediated erythrophagocytosis and hematoma absorption following intracerebral hemorrhage

MIcroglia/macrophage-mediated erythrophagocytosis plays a crucial role in hematoma clearance after intracerebral *** cytoskeletal changes accompany ***,whether and how these changes are associated with microglia/macrophage-mediated erythrophagocytosis remain *** this study,we investigated the function of acetylatedα-tubulin,a stabilized microtubule form,in microglia/macrophage erythrophagocytosis after intracerebral hemorrhage both in vitro and in *** first assessed the function of acetylatedα-tubulin in erythrophagocytosis using primary DiO GFP-labeled red blood cells co-cultured with the BV2 microglia or RAW264.7 macrophage cell ***α-tubulin expression was significantly decreased in BV2 and RAW264.7 cells during ***,silencingα-tubulin acetyltransferase 1(ATAT1),a newly discoveredα-tubulin acetyltransferase,decreased Ac-α-tub levels and enhanced the erythrophagocytosis by BV2 and RAW264.7 *** with these findings,in ATAT1-/-mice,we observed increased ionized calcium binding adapter molecule 1(Iba1)and Perls-positive microglia/macrophage phagocytes of red blood cells in peri-hematoma and reduced hematoma volume in mice with intracerebral ***,knocking out ATAT1 alleviated neuronal apoptosis and pro-inflammatory cytokines and increased anti-inflammatory cytokines around the hematoma,ultimately improving neurological recovery of mice after intracerebral *** findings suggest that ATAT1 deficiency accelerates erythrophagocytosis by microglia/macrophages and hematoma absorption after intracerebral *** results provide novel insights into the mechanisms of hematoma clearance and suggest ATAT1 as a potential target for the treatment of intracerebral hemorrhage.

Using Machine Learning to Identify and Optimize Sensitive Parameters in Urban Flood Model Considering Subsurface Characteristics

This study presents a novel method for optimizing parameters in urban flood models,aiming to address the tedious and complex issues associated with parameter ***,a coupled one-dimensional pipe network runoff model and a two-dimensional surface runoff model were integrated to construct an interpretable urban flood ***,a principle for dividing urban hydrological response units was introduced,incorporating surface attribute *** K-means algorithm was used to explore the clustering patterns of the uncertain parameters in the model,and an artificial neural network(ANN)was employed to identify the sensitive ***,a genetic algorithm(GA) was used to calibrate the parameter thresholds of the sub-catchment units in different urban land-use zones within the flood *** results demonstrate that the parameter optimization method based on K-means-ANN-GA achieved an average Nash-Sutcliffe efficiency coefficient(NSE) of *** to the ANN-GA and K-means-deep neural networks(DNN) methods,the proposed method better characterizes the runoff generation and flow *** study demonstrates the significant potential of combining machine learning techniques with physical knowledge in parameter optimization research for flood models.

Protective effects of CY-09 and astaxanthin on NaIO_(3)-induced photoreceptor inflammation via the NLRP3/autophagy pathway

AIM:To study the effect of the NLRP3/autophagy pathway on the photoreceptor inflammatory response and the protective mechanism of CY-09 and astaxanthin(AST).METHODS:ICR mice were intraperitoneally injected NaIO_(3),CY-09,AST successively and divided into 5 groups,including the control,NaIO_(3),NaIO_(3)+CY-09,NaIO_(3)+AST,and NaIO_(3)+CY-09+AST *** domain optical coherence tomography and flash electroretinogram were examined and the retina tissues were harvested for immunohistochemistry,enzyme linked immunosorbent assay(ELISA),and Western *** pigment epithelium cell line(ARPE-19 cells)and mouse photoreceptor cells line(661W cells)were also treated with NaIO_(3),CY-09,and AST *** proliferation was assessed by cell counting kit-8(CCK-8)*** was analyzed by flow *** in autophagosome morphology were observed by transmission electron *** polymerase chain reaction(qPCR)was used to detect NLRP3 and ***3,caspase-1,cleaved caspase-1,p62,Beclin-1,and LC3 protein levels were measured by Western ***-1βand IL-18 were measured by ***:Compared with the control group,the activity of NaIO_(3)-treated 661W cells decreased within 24 and 48h,apoptosis increased,NLRP3,caspase-1,IL-1βand IL-18 levels increased,and autophagy-related protein levels increased(P<0.05).Compared with NaIO_(3) group,CY-09 and AST inhibited apoptosis(P<0.05),reduced NLRP3,caspase-1,IL-1βand IL-18 expression(P<0.05),and inhibited *** with the other groups,CY-09 combined with AST significantly decreased NLRP3 expression and inhibited the expression of the autophagy-related proteins p62,Beclin-1,and LC3 in vitro and in vivo(P<0.05).CONCLUSION:CY-09 and AST inhibit NaIO_(3)-induced inflammatory damage through the NLRP3/autophagy pathway in vitro and in ***-09 and AST may protect retina from inflammatory injury.
