BACKGROUND leukemia stem cells(lSCs)are found to be one of the main factors contributing to poor therapeutic effects in acute myeloid leukemia(AMl),as they are protected by the bone marrow microenvironment(BMM)against conventional therapies.Gossypol acetic acid(GAA),which is extracted from the seeds of cotton plants,exerts anti-tumor roles in several types of cancer and has been reported to induce apoptosis of lSCs by inhibiting Bcl2.aiM To investigate the exact roles of GAA in regulating lSCs under different microenvironments and the exact mechanism.METHODS In this study,lSCs were magnetically sorted from AMl cell lines and the CD34+CD38-population was obtained.The expression of leucine-rich pentatricopeptide repeat-containing protein(lRPPRC)and forkhead box M1(FOXM1)was evaluated in lSCs,and the effects of GAA on malignancies and mitochondrial RESUlTS lRPPRC was found to be upregulated,and GAA inhibited cell proliferation by degrading lRPPRC.GAA induced lRPPRC degradation and inhibited the activation of interleukin 6(Il-6)/janus kinase(JAK)1/signal transducer and activator of transcription(STAT)3 signaling,enhancing chemosensitivity in lSCs against conventional chemotherapies,including l-Asparaginase,Dexamethasone,and cytarabine.GAA was also found to downregulate FOXM1 indirectly by regulating lRPPRC.Furthermore,GAA induced reactive oxygen species accumulation,disturbed mitochondrial homeostasis,and caused mitochondrial dysfunction.By inhibiting Il-6/JAK1/STAT3 signaling via degrading lRPPRC,GAA resulted in the elimination of lSCs.Meanwhile,GAA induced oxidative stress and subsequent cell damage by causing mitochondrial damage.CONClUSION Taken together,the results indicate that GAA might overcome the BMM protective effect and be considered as a novel and effective combination therapy for AMl.

Objective:Appraisal of treatment outcomes in integrative medicine is a challenge due to a gap between the concepts of Western medicine(WM)disease and traditional Chinese medicine(TCM)syndrome.This study presents an approach for the appraisal of integrative medicine that is based on targeted metabolomics.We use non-alcoholic fatty liver disease with spleen deficiency syndrome as a test case.Methods:A patient-reported outcome(PRO)scale was developed based on literature review,Delphi consensus survey,and reliability and validity test,to quantitatively evaluate spleen deficiency syndrome.Then,a metabonomic foundation for the treatment of non-alcoholic fatty liver disease with spleen deficiency syndrome was identified via a longitudinal interventional trial and targeted metabolomics.Finally,an integrated appraisal model was established by identifying metabolites that responded in the treatment of WM disease and TCM syndrome as positive outcomes and using other aspects of the metabonomic foundation as independent variables.Results:Ten symptoms and signs were included in the spleen deficiency PRO scale.The internal reliability,content validity,discriminative validity and structural validity of the scale were all qualified.Based on treatment responses to treatments for WM disease(homeostasis model assessment of insulin resistance)or TCM syndrome(spleen deficiency PRO scale score)from a previous randomized controlled trial,two cohorts comprised of 30 participants each were established for targeted metabolomics detection.Twenty-five metabolites were found to be involved in successful treatment outcomes to both WM and TCM,following quantitative comparison and multivariate analysis.Finally,the model of the integrated appraisal system was exploratively established using binary logistic regression;it included 9 core metabolites and had the prediction probability of 83.3%.Conclusion:This study presented a new and comprehensive research route for integrative appraisal of treatment outcomes for WM disease and TCM syndrome.Critica

Preparation of Polyurea Elastomer with Flame Retardant, Insulation and Thermal Conductivity Properties

By using 6,6-((sulfonylbis(4,1-phenylene)bis(azanediyl))bis(thiophen-2-ylm-ethylene))bis6H-di-benzo[c,e][1,2]oxaphosphinine 6-oxide(DOPO-N)as phosphorus-nitrogen flame retardant,the polyurea(PUA)with flame retardant properties(PUA/DOPO-N)was prepared.In addition,organically modified montmorillonite(OMMT)and magnesium hydroxide(MH)were used as co-effectors respectively,and the flame retardant PUA(PUA/DOPO-N/OMMT and PUA/DOPO-N/MH)were also prepared.Thermal properties,flame retardant properties,flame retardant mechanism and mechanical properties of PUA/DOPO-N,PUA/DOPO-N/OMMT and PUA/DOPO-N/MH were investigated by thermogravimetric(TG)analysis,limiting oxygen index(lOI),Ul 94,cone calorimeter test,scanning electron microscopy(SEM),and tensile test.The results show that the lOI value of PUA/20%DOPO-N,PUA/18%DOPO-N/2%OMMT and PUA/15%DOPO-N/5%MH are 27.1%,27.7%,and 28.3%,respectively,and Ul 94 V-0 rating is attained.Compared with PUA,the peak heat release rate(pk-HRR),total heat release(THR)and average effective heat combustion(av-EHC)of PUA/20%DOPO-N,PUA/18%DOPO-N/2%OMMT and PUA/15%DOPO-N/5%MH decrease significantly.SEM results indicate that the residual chars of PUA/20%DOPO-N,PUA/18%DOPO-N/2%OMMT and PUA/15%DOPO-N/5%MH are completer and more compact.The complex of DOPO-N/OMMT and DOPO-N/MH have synergistic flame retardancy.The mechanical properties of PUA can be improved by the addition of DOPO-N,DOPO-N/OMMT and DOPO-N/MH,respectively.The insulation performance test shows that the volume resistivity of PUA/20%DOPO-N is 6.25×10^(16)Ω.cm.Furthermore,by using modified boron nitride(MBN)as heat dissipating material,the complex of PUA/MBN was prepared,and the thermal conductivity of PUA/MBN was investigated.The thermal conductivity of PUA/8%MBN complex coating at room temperature is 0.166 W/(M·K),which is a 163%improvement over pure PUA.

Changes in posterior corneal elevation after small incision lenticule extraction for different myopic diopters

●aiM:To study the changes and effect factors of posterior corneal surface after small incision lenticule extraction(SMIlE)with different myopic diopters.●METHODS:Ninety eyes of 90 patients who underwent SMIlE were included in this retrospective study.Patients were allocated into three groups based on the preoperative spherical equivalent(SE):low myopia(SE≥-3.00 D),moderate myopia(-3.00 D>SE>-6.00 D)and high myopia(SE≤-6.00 D).Posterior corneal surfaces were measured by a Scheimpflug camera preoperatively and different postoperative times(1wk,1,3,6mo,and 1y).Posterior mean elevation(PME)at 25 predetermined points of 3 concentric circles(2-,4-,and 6-mm diameter)above the best fit sphere was analyzed.●RESUlTS:All surgeries were completed uneventfully and no ectasia was found through the observation.The difference of myopia group was significant at the 2-mm ring at 1 and 3mo postoperatively(1mo:P=0.017;3mo:P=0.018).The effect of time onΔPME was statistically significant(2-mm ring:P=0.001;4-mm ring:Pl locations onΔPME was significant except 1wk postoperatively(1mo:P=0.000;3mo:P=0.000;6mo:P=0.001;1y:P=0.001).Posterior corneal stability was linearly correlated with SE,central corneal thickness,ablation depth,residual bed thickness,percent ablation depth and percent stromal bed thickness.●CONClUSION:The posterior corneal surface changes dynamically after SMIlE.No protrusion is observed on the posterior corneal surface in patients with different degrees of myopia within one year after surgery.SMIlE has good stability,accuracy,safety and predictability.

Endovascular treatment of ruptured lobulated anterior communicating artery aneurysms:A retrospective study of 24 patients

BACKGROUND lobulated intracranial aneurysm is a special type of aneurysm with at least one additional cyst in the neck or body of the aneurysm.lobulated intracranial aneurysm is a complex aneurysm with complex morphology and structure and weak tumor wall,which is an independent risk factor for rupture and hemorrhage.lobular aneurysms located in the anterior communicating artery complex account for 36.9%of all intracranial lobular aneurysms.Due to its special anatomical structure,both craniotomy and endovascular treatment are more difficult.Compared with single-capsule aneurysms,craniotomy for lobular intracranial aneurysms has a higher risk and complication rate.aiM To investigate the efficacy and safety of endovascular treatment for ruptured lobulated anterior communicating artery aneurysm(ACoAA).METHODS Patients with ruptured lobulated ACoAA received endovascular treatment in Sanming First Hospital Affiliated to Fujian Medical University from June 2020 to June 2022 were retrospectively included.Their demographic,clinical and imaging characteristics,endovascular treatment methods and follow-up results were collected.RESUlTS A total of 24 patients with ruptured lobulated ACoAA were included,including 9 males(37.5%)and 15 females(62.5%).Their age was 56.2±8.9 years old(range 39-74).The time from rupture to endovascular treatment was 10.9±12.5 h.The maximum diameter of the aneurysms was 5.1±1.0 mm and neck width were 3.0±0.7 mm.Nineteen patients(79.2%)were double-lobed and 5(20.8%)were multilobed.Fisher's grade:Grade 2 in 16 cases(66.7%),grade 3 in 6 cases(25%),and grade 4 in 2 cases(8.3%).Hunt-Hess grade:Grade 0-2 in 5 cases(20.8%),grade 3-5 in 19 cases(79.2%).Glasgow Coma Scale score:9-12 in 14 cases(58.3%),13-15 in 10 cases(41.7%).Immediately postprocedural Raymond-Roy grade:grade 1 in 23 cases(95.8%),grade 2 in 1 case(4.2%).Raymond-Roy grade in imaging follow-up for 2 wk to 3 months:grade 1 in 23 cases(95.8%),grade 2 in 1 case(4.2%).Followup for 2 to 12 months showed that 21 patients(87.5%)had good functional o

Design and test of the key components for a combing-type tobacco harvester

In response to the problems of low efficiency,high labor intensity,and low mechanization in manual tobacco harvesting,a comb-off tobacco picking device for southern hilly tobacco areas was designed following the agronomic requirements and the principle of manual picking of tobacco harvesting in southern China.The device was composed of the power chassis,picking mechanism,and storage mechanism.This study involved the theoretical analysis,structural design,and modeling of the key components,such as chassis structure,combing-type picking mechanism,and power synchronization mechanism.The results of the motion analysis and calculation of the picking process demonstrated that the adjustment range of the comb chain elevation angles was from 12.4°to 20.9°,the comb rod installation distance was 76.2 mm,and the synchronizing mechanism transmission ratio was 3:10.One-factor test and three-factor three-level orthogonal test was performed with the forward speed of the chassis,the distance between the picking device baffles and the elevation angle of the chain with the combing bar as test factors,and the rate of broken and missed tobacco picking as evaluation indicators.It was revealed that the optimal combination of the forward speed of the chassis,the distance between the baffles,and the chain elevation angle were 1.5 km/h,75 mm,and 12.4°,respectively.Moreover,verification tests suggested that the breakage rate of tobacco leaves was 9.99%,and the probability of missed tobacco picking was 7.31%,both of which satisfy the agriculture requirements and the operational requirements in the agricultural machinery certification syllabus.

Genetic diversity of the S-type small subunit ribosomal RNA gene of Plasmodium knowlesi isolates from Sabah,Malaysian Borneo and Peninsular Malaysia

Objective:To determine the genetic diversity of Plasmodium(P.)knowlesi isolates from Sabah,Malaysian Borneo and Peninsular Malaysia,targeting the S-type SSU rRNA gene and including aspects of natural selection and haplotype.Methods:Thirty-nine blood samples infected with P.knowlesi were collected in Sabah,Malaysian Borneo and Peninsular Malaysia.The S-type SSU rRNA gene was amplified using polymerase chain reaction,cloned into a vector,and sequenced.The natural selection and haplotype of the S-type SSU rRNA gene sequences were determined using DnaSP v6 and illustrated using NETWORK v10.This study's 39 S-type SSU rRNA sequences and eight sequences from the Genbank database were subjected to phylogenetic analysis using MEGA 11.Results:Overall,the phylogenetic analysis showed no evidence of a geographical cluster of P.knowlesi isolates from different areas in Malaysia based on the S-type SSU rRNA gene sequences.The S-type SSU rRNA gene sequences were relatively conserved and with a purifying effect.Haplotype sharing of the S-type SSU rRNA gene was observed between the P.knowlesi isolates in Sabah,Malaysian Borneo,but not between Sabah,Malaysian Borneo and Peninsular Malaysia.Conclusions:This study suggests that the S-type SSU rRNA gene of P.knowlesi isolates in Sabah,Malaysian Borneo,and Peninsular Malaysia has fewer polymorphic sites,representing the conservation of the gene.These features make the S-type SSU rRNA gene suitable for comparative studies,such as determining the evolutionary relationships and common ancestry among P.knowlesi species.

Amorphous Sb/C composite with isotropic expansion property as an ultra-stable and high-rate anode for lithium-ion batteries

Antimony （Sb） is an intriguing anode material for li-ion batteries （lIBs） owing to its high theoretical capacity of 660 m Ah·g-1and appropriate working potential of～0.8 V （vs.li+/li）.However,just like all alloying materials,the Sb anode suffers from huge volume expansion （230%） during repeated insertion/extraction of li+ions,resulting in structural deterioration and rapid capacity decay.In this work,a novel amorphous Sb/C composite with atomically dispersed Sb particles in carbon matrix is prepared via a straightforward high-energy ball milling approach.The intimate intermixing of amorphous Sb with C provides homogeneous element distribution and isotropic volume expansion during cycling,resulting in persistent structural stability.Meanwhile,the disordered structure of amorphous material shortens the diffusion distance of lithium ions/electrons,promoting fast reaction kinetics and rate capability.Benefiting from the aforementioned effects,the amorphous Sb/C exhibits a high reversible capacity of537.4 m Ah·g-1at 0.1 A·g-1and retains 201.0 m Ah·g-1at an ultrahigh current rate of 10.0 A·g-1.Even after 1500deep cycles at 2.0 A·g-1,the amorphous Sb/C electrode still maintains 86.3%of its initial capacity,which outperforms all existing Sb-based anodes reported so far.Postmortem analysis further reveals a greatly reduced volume variation of merely 34.6%for the amorphous Sb/C electrode,much lower than that of 223.1%for crystalline Sb materials.This study presents a new approach to stabilizing Sb-based alloy anodes and contributes to the construction of high-performance amorphous anode materials for lIBs,enabling advanced energy storage.

Artificial Intelligence and Network Pharmacology Reveal the Medication Rules of Professor Wang Yu-Ying in the Treatment of Climacteric Syndrome

Objective: The objective of this study was to explore the medication rules and academic ideas of Professor Wang Yu-Ying in the treatment of climacteric syndrome(ClS) and to predict new prescriptions. Materials and Methods: The characteristics of frequency, clustering, four properties, and five flavors were analyzed, and new prescriptions were predicted through an artificial intelligence(ai)-based method. The potential pathways of new prescriptions were explored through network pharmacology-based analysis. Results: The top 16 medicinals used by Professor Wang Yu-Ying in the treatment of ClS included Danggui, longgu, Muli, Fuling, Chuanxiong, Gancao, Xiangfu, and Tusizi. The ai method was applied to predict the basic prescription for treating ClS: Danggui 15 g, Duanlonggu 30 g, Duanmuli 30 g, Fuling 28 g, Chuanxiong 10 g, Gancao 6 g, Xiangfu 12 g, Tusizi 14 g, etc., Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses showed that the pathogenesis of ClS might be related to the estrogen pathway, involving typical steroid responses. Conclusions: This study summarized Professor Wang's medication experience in the treatment of ClS based on the data mining of clinical diagnoses and treatment cases. The ai method was used to predict the new prescription of ClS treatment, which was found to be reasonable by network pharmacology studies on its multi-target and multi-pathway mechanisms.

Understanding Human and Machine Interaction from Decision Perspective: An Empirical Study Based on the Game of Go

The authors aim to interpret human and ai interactions from the decision perspective.The authors decompose the interaction analysis into the following main components in the context of interactions:Individual behavior patterns,interaction relationships,and comprehensive analysis.The authors interpret intertemporal decisions from a physical perspective and employ cross-discipline concepts and methodologies to extract the behavior characteristics of players in the empirical case study.About the individual behavior patterns,the authors find that human players prefer short-term periods to ai in decision-making.The interaction relationship analysis reveals a dynamic relationship between possible short-term co-movement and nearly counter-movement in the long run.The authors apply principal component analysis to descriptive indicators and discover a regular decision hierarchy.The main behavior pattern of players in the game of Go is switching between careful and daring behaviors.The differences in the decision hierarchies imply a discrepancy of patience between humans and ai.
