Ommochrome is the major source for eye coloration of all insect species so far examined.Phenoxazinone synthetase (PHS)has always been regarded as the terminal step enzyme for ommochrome formation,which is encoded by cardinal or karmoisin genes. Our previous study indicated that the karmoisin ortholog gene (Nl-karmoisin)product in the brown planthopper (BPH)was a monocarboxylate transporter,while not a PHS. Here,based on full-length complementary DNA,the cardinal ortholog gene in BPH (Nl-cardinal)product was predicted to be a haem peroxidase rather than a PHS.We suggest for the first time that neither karmoisin nor cardinal encodes the PHS,but whether PHS participates in BPH eye pigmentation needs further research.Nymphal RNA interference (RNai)experiments showed that knockdown Nl-cardinal transcript led the BPH ocelli and compound eye to color change from brown to red,while knockdown Nl-karmoisin only made the ocelli present the red phenotype.Notably,not only the Nl-cardinal transcript, dscd injection (Nl-cardinal targeting double-stranded DNA (dsRNA))also significantly reduced the Nl-karmoisin transcript by 33.7%,while dska (Nl-karmoisin targeting dsRNA) injection did not significantly change the Nl-cardinal transcript.Considering the above RNai and quantitative real-time polymerase chain reaction results,we propose that Nl-cardinal plays a more important role in ommochrome synthesis than Nl-karmoisin,and it may be an upstream gene of Nl-karmoisin.The present study suggested that both karmoisin and cardinal ortholog genes play a role in ommochrome synthesis in a hemimetabolous insect.

This research demonstrates the production of mesoporous activated carbon from sargassum fusiforme via physical activation with carbon dioxide.Central composite design was applied to conduct the experiments at different levels by altering three operating parameters.Activation temperature(766-934℃),CO2 flow rate(0.8-2.8 L·min^-1)and activation time(5-55 min)were the variables examined in this study.The effect of parameters on the specific surface area,total pore volume and burn-out rate of activated carbon was studied,and the influential parameters of methylene blue adsorption value were identified employing analysis of variance.The optimum conditions for maximum methylene blue adsorption value were:activation temperature=900℃,activation time=29.05 min and CO2 flow rate=1.8 L·min(-1).The activated carbon produced under optimum conditions was characterized by BET,FTIR and SEM.The adsorption behavior on congo red was studied.The effect of parameters on the adsorbent dosage,temperature,PH and initial congo red concentration was investigated.The adsorption properties of the activated carbon were investigated by kinetics.The equilibrium removal rate and maximum adsorption capacity reaches up to 94.72%,234 mg·g^-1,respectively when initial congo red concentration is 200 mg·L^-1 under adsorbent dosage(0.8 g·L^-1),temperature(30℃),PH7.

In August 2021,three students with diarrhea from the same school visited a local hospital in the S district of Beijing.An epidemic investigation showed that there were more students with diarrhea in the same school and they had one meal together.Campylobacter jejuni was isolated from both patients with diarrhea and asymptomatic food handlers;however,the latter also carried Campylobacter coli.Phylogenomic analysis showed that there was a campylobacteriosis outbreak among the students,and the asymptomatic food handler may have been the source of the infection.Routine inspection and surveillance for Campylobacter is needed for the food producing staff,particularly those cooking in the cafeteria in schools or other public food services.

Previous studies have shown that vagus nerve stimulation can improve patients' locomotor function.The stimulation of the auricular vagus nerve,which is the only superficial branch of the vagus nerve,may have similar effects to vagus nerve stimulation.However,the precise mechanisms remain poorly understood.In this study,rat models of cerebral ischemia/reperfusion injury were established by modified Longa ligation.Twenty-four hours later,7-day auricular vagus nerve stimulation was performed.The results showed that auricular vagus nerve stimulation promoted the secretion of acetylcholine,inhibited the secretion of interleukin-1β,interleukin-6,and tumor necrosis factor-α,and reduced connexin 43 phosphorylation in the ischemic penumbra and motor cortex,promoting locomotor function recovery in rats with cerebral ischemia/reperfusion injury.These findings suggested that auricular vagus nerve stimulation promotes the recovery of locomotor function in rats with cerebral ischemia/reperfusion injury by altering the secretion of acetylcholine and inflammatory factors and the phosphorylation of connexin 43.This study was approved by the Animal Use and Management Committee of Shanghai University of Traditional Chinese Medicine on November 8,2019(approval No.PZSHUTCM191108014).

Objective: To evaluate the effect of salidroside on oxygen and glucose deprivation(OGD)-treated NT2 cells and its underlying mechanisms of action.Methods: Retinoic acid was used to induce the differentiation of NT2 cells into neurons. The effects of salidroside on survival, apoptosis, inflammatory response, and oxidative stress of neurons undergoing OGD were evaluated. Using precursor cells as controls, the effect of salidroside on the differentiation progression of OGDtreated cells was evaluated. In addition, the effect of erastin, a ferroptosis inducer, on NT2 cells was examined to investigate the underlying mechanisms of neuroprotective action of salidroside.Results: Salidroside alleviated the effects of OGD on neuronal survival, apoptosis, inflammation, and oxidative stress, and promoted NT2 cell differentiation. Moreover, salidroside prevented ferroptosis of OGD-treated cells, which was abolished following erastin treatment, indicating that ferroptosis mediated the regulatory pathway of salidroside.Conclusions: Salidroside attenuates OGD-induced neuronal injury by inhibiting ferroptosis and promotes neuronal differentiation.

The onset of retinal degenerative disease is often associated with neuronal loss. Therefore, how to regenerate new neurons to restore vision is an important issue. NeuroD1 is a neural transcription factor with the ability to reprogram brain astrocytes into neurons in vivo. Here, we demonstrate that in adult mice, NeuroD1 can reprogram Müller cells, the principal glial cell type in the retina, to become retinal neurons. Most strikingly, ectopic expression of NeuroD1 using two different viral vectors converted Müller cells into different cell types. Specifically, AAV7 m8 GFAP681::GFP-ND1 converted Müller cells into inner retinal neurons, including amacrine cells and ganglion cells. In contrast, AAV9 GFAP104::ND1-GFP converted Müller cells into outer retinal neurons such as photoreceptors and horizontal cells, with higher conversion efficiency. Furthermore, we demonstrate that Müller cell conversion induced by AAV9 GFAP104::ND1-GFP displayed clear dose-and time-dependence. These results indicate that Müller cells in adult mice are highly plastic and can be reprogrammed into various subtypes of retinal neurons.

Antibiotic resistance genes(ARGs)as new pollutants have become a global environmental pollution problem in recent years.Elevated atmospheric CO_(2) is one of the major factors affecting global climate change.But,the impacts of elevated CO_(2) on soil ARGs in multiple antibiotics-contaminated paddy soils are largely unknown.In this study,six antibiotics including sulfadiazine(SDZ),sulfamethoxazole(SMZ),tetracycline(TC),oxytetracycline(OTC),enrofloxacin(ENR),and ciprofloxacin(CIP)were selected to investigate their combined effects on rice biomass,antibiotics accumulation,soil bacterial community and ARGs under elevated CO_(2) levels.Results showed that elevated CO_(2) significantly reduced the accumulation of SMZ,OTC,ENR,and CIP in rice grains by 18.98%,20.07%,41.73%,and 44.25%,respectively.Elevated CO_(2) could affect soil microbialβ-diversity,and tend to reduce the microbial functions of human diseases,organismal systems,and genetic information processing.In addition,elevated CO_(2) significantly decreased the abundance of sulfonamide ARGs,tetracycline ARGs,and quinolone ARGs by 19.59%,18.58%,and 28.96%,respectively,while increased that of multidrug ARGs by 11.54%.Overall,this study emphasized that elevated CO_(2) may mitigate the threat of antibiotics contamination to rice food security but aggravate the environmental risk of multidrug ARGs in soil,contributing to a better understanding of the consequences of elevated CO_(2) levels on food security and soil ecological health in multiple antibiotics-contaminated paddy fields.

Major depressive disorder(MDD)is a prevalent,often chronic,and highly disabling multidimensional psychiatric illness.[1] Moreover,co-occurring anxiety symptoms are extremely common among patients with MDD;up to 90%of patients present with anxiety symptoms.[2]Notably,high levels of anxiety symptoms may predict worse clinical outcomes because of poor response to pharmacotherapy for MDD.[3]So use of augmentation or combination strategies during early course of treatment could be necessary,but ensuring the accurate and timely change is difficult because of the lack of consensus to assess the early improvement of initial treatment.To date,replicated evidence indicates that the lack of early improvement(eg,aimed to evaluate the early onset of antidepressant action and clinical outcomes in patients with MDD and high anxiety,and to explore the potential influencing factors of early onset improvement.

Objective: To investigate the mechanism by which Chinese medicine Shengmai Yin(SMY)reverses epithelial-mesenchymal transition(EMT) through lipocalin-2(LCN2) in nasopharyngeal carcinoma(NPC)cells CNE-2R. Methods: Morphological changes in EMT in CNE-2R cells were observed under a microscope,and the expressions of EMT markers were detected using quantitative real-time PCR(RT-q PCR) and Western blot assays. Through the Gene Expression Omnibus dataset and text mining, LCN2 was found to be highly related to radiation resistance and EMT in NPC. The expressions of LCN2 and EMT markers following SMY treatment(50 and 100 μg/m L) were detected by RT-q PCR and Western blot assays in vitro. Cell proliferation,migration, and invasion abilities were measured using colony formation, wound healing, and transwell invasion assays, respectively. The inhibitory effect of SMY in vivo was determined by observing a zebrafish xenograft model with a fluorescent label. Results: The CNE-2R cells showed EMT transition and high expression of LCN2,and the use of SMY(5, 10 and 20 μg/mL) reduced the expression of LCN2 and reversed the EMT in the CNE-2R cells. Compared to that of the CNE-2R group, the proliferation, migration, and invasion abilities of SMY high-concentration group were weakened(P<0.05). Moreover, SMY mediated tumor growth and metastasis in a dose-dependent manner in a zebrafish xenograft model, which was consistent with the in vitro results.Conclusions: SMY can reverse the EMT process of CNE-2R cells, which may be related to its inhibition of LCN2expression. Therefore, LCN2 may be a potential diagnostic marker and therapeutic target in patients with NPC.
