Hemophagocytic lymphohistiocytosis after autologous stem cell transplantation in angioimmunoblastic T-cell lymphoma:A case report

BACKGROUND Angioimmunoblastic T-cell lymphoma(aiTL), a unique subtype of peripheral Tcell lymphoma, has relatively poor outcomes. High-dose chemotherapy with autologous stem cell transplantation(ASCT) can achieve complete remission and improve outcomes. Unfortunately, subsequent T-cell lymphoma-triggered hemophagocytic lymphohistiocytosis(HLH) has a worse prognosis than B-cell lymphoma-triggered *** SUMMARY We here report a 50-year-old woman with aiTL who achieved a favorable outcome after developing HLH 2 mo after receiving high-dose chemotherapy/ASCT. The patient was initially admitted to our hospital because of multiple enlarged lymph nodes. The final pathologic diagnosis, made on biopsy of a left axillary lymph node was aiTL(Stage Ⅳ, Group A). Four cycles of the following chemotherapy regimen were administered: Cyclophosphamide 1.3 g, doxorubicin 86 mg, and vincristine 2 mg on day 1;prednisone 100 mg on days 1-5;and lenalidomide 25 mg on days 1-14. The interval between each cycle was 21 d. The patient received a conditioning regimen(busulfan, cyclophosphamide, and etoposide) followed by peripheral blood stem cell infusion. Unfortunately, she developed sustained fever and a low platelet count 17 d after ACST, leading to a diagnosis of HLH after ASCT. During treatment, she experienced thrombocytopenia and Pneumocystis carinii pneumonia. The patient was successfully treated with etoposide and *** It is possible that development of HLH is related to immune reconstitution after ASCT.

Cyclic adenosine monophosphate involvement in low-dose cyclophosphamide-reversed immune evasion in a mouse lymphoma model

Lymphoma cells mobilize many mechanisms to evade the immune system. There is substantial evidence that CD4＋CD25＋ regulatory T cells （Tregs） play a key role in the control of immune evasion. Tregs can transfer cyclic adenosine monophosphate （cAMP） to effector T cells, suggesting an association between Tregs＇ immune-evasion role and the intracellular cAMP pathway. In this study, we used A20 B-cell lymphoma mice as aggressive tumor models to investigate the mechanism of the depletion of Tregs by low-dose cyclophosphamide （CY, 20 mg/kg）. The tumor-bearing mice had longer survival times and slower tumor growth rates following treatment with CY, but its effects were temporary. Along with the depletion of Tregs by low-dose CY treatment, the expression of interleukin-2 （IL-2） in T effector cells increased, and intracellular cAMP concentrations in immune cells decreased. Our study demonstrates the ability of low-dose CY to reverse Tregs-mediated immune evasion in a mouse model. The changes in intracellular cAMP concentrations correlated with the upregulation of effector T cells and the downregulation of Tregs, indicating the close association of cAMP analogs and low-dose CY in the immune therapy of B-cell lymphoma.

XRL-SHAP-Cache:an explainable reinforcement learning approach for intelligent edge service caching in content delivery networks

Content delivery networks(CDNs) play a pivotal role in the modern internet infrastructure by enabling efficient content delivery across diverse geographical regions. As an essential component of CDNs, the edge caching scheme directly influences the user experience by determining the caching and eviction of content on edge servers. With the emergence of 5G technology, traditional caching schemes have faced challenges in adapting to increasingly complex and dynamic network environments. Consequently, deep reinforcement learning(DRL) offers a promising solution for intelligent zero-touch network governance. However, the blackbox nature of DRL models poses challenges in understanding and making trusting decisions. In this paper,we propose an explainable reinforcement learning(XRL)-based intelligent edge service caching approach,namely XRL-SHAP-Cache, which combines DRL with an explainable artificial intelligence(Xai) technique for cache management in CDNs. Instead of focusing solely on achieving performance gains, this study introduces a novel paradigm for providing interpretable caching strategies, thereby establishing a foundation for future transparent and trustworthy edge caching solutions. Specifically, a multi-level cache scheduling framework for CDNs was formulated theoretically, with the D3QN-based caching scheme serving as the targeted interpretable model. Subsequently, by integrating Deep-SHAP into our framework, the contribution of each state input feature to the agent's Q-value output was calculated, thereby providing valuable insights into the decision-making process. Rheumatoid arthritis(RA)is a chronic autoimmune disorder marked by persistent synovial inflammation and joint degradation,posing challenges in the development of effective ***,an alkaloid found in lotus leaf,has shown promising anti-inflammatory and anti-tumor effects,yet its efficacy in RA treatment remains *** study investigated the antiproliferative effects of nuciferine on the MH7A cell line,a human RA-derived fibroblast-like synoviocyte,revealing its ability to inhibit cell proliferation,promote apoptosis,induce apoptosis,and cause G1/S phase ***,nuciferine significantly reduced the migration and invasion capabilities of MH7A *** therapeutic potential of nuciferine was further evaluated in a collagen-induced arthritis(CIA)rat model,where it markedly alleviated joint swelling,synovial hyperplasia,cartilage injury,and inflammatory *** also improved collagen-induced bone erosion,decreased pro-inflammatory cytokines and serum immunoglobulins(IgG,IgG1,IgG2a),and restored the balance between T helper(Th)17 and regulatory T cells in the spleen of CIA *** results indicate that nuciferine may offer therapeutic advantages for RA by decreasing the proliferation and invasiveness of FLS cells and correcting the Th17/Treg cell imbalance in CIA rats.

Since the industrial revolution,enhancement of atmospheric greenhouse gas concentrations as a result of human activities has been the primary cause of global *** monitoring and evaluation of greenhouse gases are significant prerequisites for carbon emission *** monthly data of global atmospheric carbon dioxide(CO_(2))and methane(CH4)column concentrations(hereinafter XCO_(2) and XCH_(4),respectively)retrieved by the Greenhouse Gas Observation Satellite(GOSAT),we analyzed the variations in XCO_(2)and XCH_(4)in China during 2010-2022 after confirming the reliability of the ***,the influence of a strong El Niño event in 2015-2016 on XCO_(2) and XCH_(4) variations in China was further *** results show that the retrieved XCO_(2) and XCH_(4) from GOSAT have similar temporal variation trends and significant correlations with the ground observation and emission inventory data of an atmospheric background station,which could be used to assess the variations in XCO_(2) and XCH_(4) in ***_(2) is high in spring and winter while XCH_(4) is high in *** XCO_(2) and XCH_(4) gradually declined from Southeast China to Northwest and Northeast China,with variation ranges of 401-406 and 1.81-1.88 ppmv,respectively;and the high value areas are located in the middle-lower Yangtze River ***_(2) and XCH_(4) in China increased as a whole during 2010-2022,with rapid enhancement and high levels of XCO_(2) and XCH_(4) in several *** significant increases in XCO_(2) and XCH_(4) over China in 2016 might be closely related to the strong El Niño-Southern Oscillation(ENSO)event during *** a global warming background in 2015,XCO_(2) and XCH_(4) increased by 0.768%and 0.657%in 2016 in *** analysis reveals that both the XCO_(2) and XCH_(4) variations might reflect the significant impact of the ENSO event on glacier melting in the Tibetan Plateau.

Metabolomic Analysis of the Anthocyanins Associated with Different Colors of Cymbidium goeringii in Guizhou, China

Cymbidium goeringii is an economically important ornamental plant,and flower color is one of the main features of *** that contributes to its high economic *** clarify the molecular mechanisms underlying the role of anthocyanins in mediating differences in color among varieties,liquid chromatography–tandem mass spectrometry was used to perform anthocyanin-targeted metabolomics of seven *** varieties,including‘Jin Qian Yuan’(JQY),‘Jin Xiu Qian Yuan’(JXQY),‘Miao Jiang Su Die’(MJSD),‘Qian Ming Su’(QMS),‘Shi Chan’(SC),and‘Yang Ming Su’(YMS),as well as the *** detected 64 anthocyanins,including cyanidins,delphinidins,malvidins,pelargonidins,peonidins,petunidins,procyanidins,and *** identified six shared differentially accumulated metabolites(DAMs),including cyanidin-3-O-rutinoside,delphinidin-3-Osophoroside,pelargonidin-3-O-rutinoside,peonidin-3-O-(6-O-malonyl-beta-D-glucoside),peonidin-3-Osophoroside,and *** DAMs were enriched in the anthocyanin biosynthesis *** Encyclopedia of Genes and Genomes pathway analysis revealed that the differentially expressed metabolites were significantly enriched in the anthocyanin biosynthesis *** of the content of differentially expressed metabolites indicated that peonidin-3-O-(6-O-malonyl-beta-D-glucoside)was the key metabolite underlying color differences among *** *** B2,pelargonidin-3-O-galactoside,and naringenin might also affect the color formation of JQY and QMS,SC,and MJSD,*** results of this study shed light on the metabolic mechanism underlying flower color differences in *** at the molecular *** findings will aid future studies of the mechanism of flower color regulation in *** and have implications for the breeding of new varieties.
