＜正＞Objective:To study the effects of Ginkgo biloba extract 50（GBE50） on myocardial function,area at risk and myocardial ultra-structure in myocardium ischemic ***:Myocardium ischemia was induced by the left anterior descending（LAD） coronary artery occlusion and myocardial function was *** vitro myocardial infarction was measured by 2.3,5-triphenyltetrazolium chloride（TTC） and Evans blue staining of heart *** hematoxylin and eosin（HE） staining was used for morphological evaluation by light microscopy. Results:Hemodynamic experiments in vivo showed that postischemic cardiac contractile function was reduced in ischemic ***（7.5 g/kg/d×7） and GBE50（100 mg/kg/d×7） improved postischemic cardiac diastolic dysfunction. The area at risk was significantly reduced and myocardial structure was better-preserved in hearts of GBE50 group and SM ***:GBE50 could attenuate myocardium damage induced by ischemia and provide potent myocardial protection.

目的化学交换饱和转移(CEST)成像技术已成为诊断与脑部和全身疾病代谢相关变化的有用工具,通过计算与水分子相邻化合物的可交换质子的含量进行定量分析。具体而言,酰胺质子转移(APT)CEST技术通过比较可交换的内源性蛋白质或肽的含量变化来区分正常组织与脑卒中和脑肿瘤组织。在体内的小器官病变诊断中,小视野(rFOV)成像技术已被广泛应用,本研究旨在应用rFOV成像技术来识别直肠中的CEST信号,探讨rFOV成像技术在直肠疾病临床诊断中的潜在效用,并为直肠疾病的放化疗提供代谢影像信息。方法使用3.0T磁共振成像扫描仪对11名健康志愿者进行了横断面全视野(Full_FOV)和rFOV CEST成像。设置的分辨率分别为2.5×2.5×6 mm3和1.5×1.5×6 mm3。采用了0.7μT和2.T两种预饱和脉冲。rFOV成像采用了ZOOM成像方法。对于2.T的饱和脉冲,采用了±3.5 ppm的MTRasym方法进行定量分析,而对于0.7μT的饱和脉冲,则采用Lorentzian Difference的方法来量化CEST的对比度图和曲线。结果相较于Full_FOV成像,rFOV方法可以在保持较好对比度的同时将扫描时间减半。与Full_FOV方法相比,rFOV成像方法可以得出与Full_FOV方法几乎相同的Z谱和MTRasym曲线。此外,以大约3 min的时间可以实现1.5 mm×1.5 mm分辨率的rFOV成像。这种r FOV成像方法可以更好地显示出整个直肠的解剖细节,包括CEST成像对比图及定量分析曲线。结论CEST MRI在直肠疾病诊断方面具有较高价值,采用rFOV技术可以提供更高的空间和时间分辨率。由于其在直肠疾病诊断方面的潜力,CEST MRI可以作为临床诊断直肠疾病的首选。

＜正＞Objective:To observe the effect of Ginkgo biloba extract 50（GBE50） on NF-kB,p38MAPK signaling pathways and related genes in rats with myocardial ischemia-reperfusion（I/R）,and investigate its mechanism against inflammatory reaction after I/***:Twenty-four SD rats were divided into sham-operated group, model group,Salviae miltiorrhizae（SM） injection group and GBE50 group *** drug administration by intragastric route for 7 days,the left anterior descending（LAD） coronary artery was occluded for 30 minutes followed by 60 minutes of reperfusion to induce ischemia-reperfusion *** protein expression of nuclear factor-kappa B（NF-kB）,p38 mitogen-activated protein kinase（p38MAPK） in myocardium were measured by immunohistochemical method;the gene expression of tumor necrosis factor-alpha（TNF-α）,inducible nitric oxide synthase（iNOS）,cycloxygenase-2.COX-2. were detected by Real-time ***:In model group,the protein expression of NF-kB in myocardium was obviously higher than those in sham-operated group（P＜0.001）, the protein expression of p38MAPK was higher than sham-operated group too,but without significant difference. Compared with model group,rats in GBE50 group had a lower expression of NF-kB（OD value P＜0.05,positive area P＜0.001）,lower expression of p38MAPK（OD value P＜0.05,positive area P＜0.01）.The gene expression of COX-2.iNOS and TNF-αin myocardium were significantly up-regulated in model group than those in sham-operated group（P＜0.05-0.001）.Compared with model group,GBE50 could down-regulate the gene expression of COX-2.and TNF-αin myocardium after I/R（P＜0.01）,and reduce the gene expression of iNOS partly（P＞0.05）. Conclusion:GBE50 can down-regulate gene expression of inflammatory reaction key enzyme COX-2.and proinflammatory cytokine TNF-α,partly down-regulate gene expression of oxidative stress key enzyme iNOS via inhibition of NF-kB and p38MAPK signaling *** suggests that GBE50 could regulate the inflammatory reaction after I/R.

目的 :研究黄芪甲苷在家兔体内的药动学和在大鼠的排泄。方法 :健康家兔和大鼠一次静脉注射 (静注 )给予黄芪甲苷 4mg·kg- 1,高效液相色谱蒸发光散射检测器法检测兔血浆和大鼠尿及粪黄芪甲苷浓度 ,用 3P97药动学软件对兔血浆浓度时间数据进行动力学分析和计算药动学参数 ,并估算大鼠体内的排泄情况。结果 :黄芪甲苷静注给药后 ,T12.α 为 0 .10h ,T12.β 为 1.4h ,Vc 为 0 .15L·kg- 1,VD 为 0 .6L·kg- 1,Cl为 0 .32.·h- 1·kg- 1,AUC为 15mg·L- 1·h。大鼠静注给药后 ,原形从尿和粪排出量分别为给药量的 16 %和 3.2.%。结论 :家兔体内黄芪甲苷的动力学过程符合二室模型 ,大鼠仅有少量原形药物从尿和粪排泄。

Bufalin is one of the main pharmacological and toxicological components of Venenum Bufonis and many traditional {2. medicine *** cardiotoxicity clearly limits its application to patients living in ***,an investigation of its toxicological mechanism is helpful for new drug development and treatment of the related clinical adverse *** investigate the cardiotoxicity of bufalin using human induced pluripotent stem cells-derived cardiomyocytes(hiPSC-CMs)(0.003–0.1μmol·L–1),human induced pluripotent stem cells-derived cardiomyocytes(hiPSC-CMs)(0.03–0.3μmol·L–1)and eight human cardiac ion channel currents(0.01–100μmol·L–1)combined with an impedance-based bioanalytical and patch clamp *** effect of bufalin on the contractility in hiPSC-CMs,which has been shown to strengthen myocardial contractility,accelerate conduction,and increase beating rate at the earlier stage of administration,whereas weakened myocardial contractility,abolished conduction,and ceased beating rate at the later stage of *** decreased the action potential duration(Action potential duration at 30%,50%and 90%repolarization),cardiac action potential amplitude,and maximal depolarization rate and depolarized the resting membrane potential of *** beating rates of hiPSC-CMs were markedly increased at 0.03μmol·L–1,while were weakened at 0.3μmol·L–1 after *** blocks INav1.5 in a concentration-dependent manner with half maximal inhibitory concentration of 74.5μmol·L–*** respectively increased the late sodium current and Na+-Ca2.exchange current with a concentration for 50%of maximal effect of 2.48 and 66.06μmol·L–1 in ***,bufalin showed no significant effects on other cardiac ion channel *** enhancement of the late sodium current is one of the main mechanism for cardiotoxicity of bufalin.
