RIG-I is a pivotal cytoplasmic sensor that recognizes different species of viral RNAs. This recognition leads to activation of the transcription factors NF-κB and IRF3, which collaborate to induce type I interferons （IFNs） and innate antiviral response. In this study, we identified the TRIM family protein TRIM4 as a positive regulator of RIG-I-mediated IFN induction. Overexpression of TRIM4 potentiated virus-triggered activation of IRF3 and NF-κB, as well as IFN-13 induction, whereas knockdown of TRIM4 had opposite effects. Mechanistically, TRIM4 associates with RIG-I and targets it for K63-linked poiyubiquitination. Our findings demonstrate that TRIM4 is an important regulator of the virus-induced IFN induction pathways by mediating RIG-I for K63-Unked ubiquitination.

Physical simulation of rock burst induced by stress waves

The behavior of stress wave propagation in rock walls and the process of rock bursts were simulated by application tests of material similar to rock. Results show that 1) the attenuation characteristics of stress waves were related to the material proper-ties, stress waves attenuate more quickly in soft material and 2) when the explosion load was applied at the top of the roadway, the number and the length of the cracks increased with a decrease in the distance between the explosive point and roof of the roadway. When the distance was 280 mm, only some chips appeared near the source, when the distance was 210 mm, some small cracks started to appear near the road-rib and when the distance was reduced to 140 mm, larger cracks appeared at the road-rib. It can be concluded that, under a given stress the number of cracks is closely related to the intensity of stress waves. The cracks in the sur-rounding rock can be reduced by controlling the intensity of the stress waves and rock bursts can be avoided to some extent by pre-venting the formation of layered crack structures. A new experimental approach has been provided for studying rock bursts by using physical simulation.

Carbon ion radiotherapy has the advantages of better therapeutic effect and fewer side effects compared with those of X-rays in many kinds of tumors,including prostate cancer,and thus is an attractive treatment approach for prostate ***,the biological effects and underlying mechanisms of carbon ion irradiation in prostate cancer are not yet fully ***,this study systematically compared the effects of carbon ion irradiation with those of X-ray irradiation on DNA damage response and found that carbon ion irradiation was more effective than X-ray *** ion irradiation can induce a high level of DNA double-strand break damage,reflected by the number of y-H2 A histone family member X foci,as well as by the foci lasting time and ***,carbon ion irradiation exhibited strong and long-lasting inhibitory effect on cell survival capability,induced prolonged cell cycle arrest,and increased apoptosis in PC-3 *** an underlying mechanism,we speculated that carbon ion irradiation-induced DNA damage evokes cell cycle arrest and apoptosis via the pRb/E2 F1/c-Myc signaling pathway to enhance the radiosensitivity of p53-deficient prostate cancer PC-3 ***,the present study suggests that carbon ion irradiation is more efficient than X-ray irradiation and may help to understand the effects of different radiation qualities on the survival potential of p53-deficient prostate cancer cells.

As a renewable,biocompatible,biodegradable soft material,chitin hydrogels have better advantages in stability,antibacterial activity,antifouling,cost,immunogenicity,and so on than most polymer ***,compared with other widely used polymer hydrogels with high strength and toughness,the practical applications of chitin-based hydrogels have been limited by their weak mechanical properties,such as cartilage repair and meniscus ***,we present the design and fabrication of chitin hydrogels with excellent mechanical strength and toughness by a dehydration and rehydration *** sequential dehydration and rehydration processes,the crystalline domains in the chitin hydrogels can be properly *** optimized crystallinity,the elastic modulus of the chitin hydrogels exceeds all previously reported values,and the fracture toughness is even comparable to some synthetic polymer hydrogels,while maintaining a high-water-content of about 80 wt.%.At the same water content,the mechanical properties of the chitin hydrogels are positively correlated with the hydrogel crystallinity,which proves that the change of mechanical properties of hydrogels is not simply dependent on weight *** hydrogels can be further strengthened by incorporating other biopolymers that are intrinsically weak,which makes the hydrogels promising for applications in fields such as cartilage repair and meniscus ***,the hydrogels enable loading and release of water-soluble and poorly water-soluble *** highly extendable strengthening and toughening strategy of chitin and chitin-based biopolymer hydrogels paves the way for their widely applications.

A Data-Oriented Method to Optimize Hydraulic Fracturing Parameters of Tight Sandstone Reservoirs

Based on the actual data collected from the tight sandstone development zone, correlation analysis using theSpearman method was conducted to determine the main factors influencing the gas production rate of tightsandstone fracturing. An integrated model combining geological engineering and numerical simulation of fracturepropagation and production was completed. Based on data analysis, the hydraulic fracture parameters wereoptimized to develop a differentiated fracturing treatment adjustment plan. The results indicate that the influenceof geological and engineering factors in the X1 and X2 development zones in the study area differs ***, it is challenging to adopt a uniform development strategy to achieve rapid production increase. Thedata analysis reveals that the variation in gas production rate is primarily affected by the reservoir thickness andpermeability parameters as geological factors. On the other hand, the amount of treatment fluid and proppantaddition significantly impact the gas production rate as engineering factors. Among these factors, the influence ofgeological factors is more pronounced in block X1. Therefore, the main focus should be on further optimizing thefracturing interval and adjusting the geological development well location. Given the existing well location, thereis limited potential for further optimizing fracture parameters to increase production. For block X2, the fracturingparameters should be optimized. Data screening was conducted to identify outliers in the entire dataset, and adata-driven fracturing parameter optimization method was employed to determine the basic adjustment directionfor reservoir stimulation in the target block. This approach provides insights into the influence of geological,stimulation, and completion parameters on gas production rate. RNA modifications have been involved in numerous biological processes, and aberrations of these modifications are tightly associated with various diseases including cancer. Herein, we developed graphenebased solid-phase extraction and robust ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) combined with stable isotope-dilution for simultaneous enrichment and accurate determination of 17 modified nucleosides in human urine. We found graphene could effectively adsorb various modified nucleosides in human urine samples. With this method, we identified and quantified these modified nucleosides in urine samples collected from lung cancer patients and healthy *** revealed that the levels of 12 modified nucleosides were all diminished in urine from lung cancer patients, compared with healthy controls. It is worth noting that we demonstrated, for the first time, the presence of 5,2-O-dimethyluridine(m~5U_m) in human urine. Together, we established a robust analytical method for simultaneous determinations of 17 modified nucleosides in human urine, and our results revealed a close correlation between the concentrations of urinary modified nucleosides and the occurrence of lung cancer, implying the potential applications of these modified nucleosides as noninvasive biomarkers for the early detection of lung cancer. Moreover, this study will stimulate future investigations on the regulatory roles of RNA modifications in the initiation and progression of lung cancer.

Reliability of DC-link capacitor in pulsed power supply for accelerator magnet

Capacitors are widely used in pulsed magnet power supplies to reduce ripple voltage, store energy, and decrease power variation. In this study, DC-link capacitors in pulsed power supplies were investigated. By deriving an analytical method for the capacitor current on the H-bridge topology side, the root-mean-square value of the capacitor current was calculated, which helps in selecting the DC-link capacitors. The proposed method solves this problem quickly and with high accuracy. The current reconstruction of the DC-link capacitor is proposed to avoid structural damage in the capacitor's current measurement, and the capacitor's hotspot temperature and temperature rise are calculated using the FFT transform. The test results showed that the error between the calculated and measured temperature increases was within 1.5 degrees C\documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\circ \hbox {C}$$\end{document}. Finally, the lifetime of DC-link capacitors was predicted based on Monte Carlo analysis. The proposed method can evaluate the reliability of DC-link capacitors in a non-isolated switching pulsed power supply for accelerators and is also applicable to film capacitors.
