Early Identification of Stable Transformation Events by Combined Use of Antibiotic Selection and Vital Detection of Green Fluorescent Protein (GFP) in Carrot (Daucus carota L.) Callus

Genetic transformation is a useful technique to complement conventional breeding in crop improvement. Although carrot has been a model organism for in vitro embryogenesis study, genetic transformation of carrot is still lengthy and labor intensive. An efficient transformation and detection system is desirable. Direct infection of Agrobacterium to carrot calli has provided an easy way for carrot genetic transformation. To improve the efficiency of antibiotic selection in this method, we report the combined use of an improved green-fluorescent protein, referred to as smGFP, to establish a versatile selection method for carrot callus transformation system. By combining antibiotic selection with the bright fluorescence observed in the callus tissue, we were able to easily identify stable transformants in early stage of the transformation process. In addition to the GFP expression of the callus cells, the transgenic nature of callus cells was confirmed with Southern and Western analysis. We found we can link the simplicity of carrot-callus-cell transformation, early detection of stable transformants with antibiotic selection, visualization of GFP fluorescence, and molecular analysis （Southern and Western） of callus tissue （non-photosynthetic tissue） to provide a more efficient way in identifying stable transformants at early stage of carrot transformation.

Relationship between indoleamine 2,3-dioxygenase activity and lymphatic invasion propensity of colorectal carcinoma

AIM: To evaluate whether serum and tumor indoleamine 2,3-dioxygenase activities can predict lymphatic invasion (LI) or lymph node metastasis in colorectal ***: The study group consisted of 44 colorectal carcinoma patients. The patients were re-grouped according to the presence or absence of LI and lymph node metastasis. Forty-three cancer-free subjects without any metabolic disturbances were included into the control group. Serum neopterin was measured by enzyme linked immunosorbent assay. Urinary neopterin and biopterin, serum tryptophan (Trp) and kynurenine (Kyn) concentrations of all patients were determined by high performance liquid chromatography. Kyn/Trp was calculated and its correlation with serum neopterin was determined to estimate the serum indoleamine 2,3-dioxygenase activity. Tissue sections from the studied tumors were re-examined histopathologically and were stained by immunohistochemistry with indoleamine-2,3-dioxygenase ***: Neither serum nor urinary neopterin was significantly different between the patient and control groups (both P > 0.05). However, colorectal carcinoma patients showed a significant positive correlation between the serum neopterin levels and Kyn/Trp (r = 0.450, P < 0.01). Urinary biopterin was significantly higher in cancer cases (P < 0.05). Serum Kyn/Trp was significantly higher in colorectal carcinoma patients (P < 0.01). Lymphatic invasion was present in 23 of 44 patients, of which only 12 patients had lymph node metastasis. Eleven patients with LI had no lymph node metastasis. Indoleamine-2,3-dioxygenase intensity score was significantly higher in LI positive cancer group (44.56% ± 6.11%) than negative colorectal cancer patients (24.04% ± 6.90%), (P < 0.05). Statistical Analysis of Bursa Nilufer Creek’s Water Quality Parameters for Period of 2002-2010

Increasing contamination of water resources in the world and our country and decreasing water quality over time, not having met the objectives of utilization of water resources;it has increased the importance of water management. The monitoring of the water resources and evaluation of these monitoring results have given direction to the studies’ outcome in order to control factors that pollute water resources and reduce water quality. Nilüfer Creek is very important for both being a source of drinking and potable water and a discharge area for wastewaters for the city of Bursa. In this study, the results of the analysis belonging to the period between 2002-2010 which are taken from 15 points by General Directorate of Bursa Water and Sewerage Administration (BUWSA) were evaluated in relation to water quality of the Nilüfer Creek. Non-parametric methods were used in the evaluation of the water quality data due to the lack of normally distributed data. The identification of the best represented parameters of the water quality was provided by applying Principal Component Analysis. According to results of the analysis, the best representative 9 parameters from the 19 water quality parameters were defined as parameters of BOD5, COD, TSS, ***, Zn, conductivity, NO2-N, Ni and NO3-N that taking part of the first two components.

The Predictive Role of New Markers in Early Diagnosis of Acute Kidney Injury in Patients with Acute Pancreatitis

Objective: Acute Kidney Injury (AKI) is a serious early complication that significantly increases the mortality rate in patients with acute pancreatitis (AP). AKI can improve with an early diagnosis and appropriate treatment protocols. We aimed to evaluate the role of IL-6, IL-18, NGAL, IL-1β, TNF-α, Cys-C and KIM-1 in serum and IL-18 in urine in the early detection of AKI in patients with AP. Materials and Methods: Fifty-six patients, who were diagnosed with AP and underwent standard treatment, between July 2011 and December 2013, were included in the study. Patients were sampled for markers at admission and discharge, or when the patients died. Patients were grouped by presence or absence of AKI. Results: The mean age of the cases was 56.41 ± 20.23 (17 - 91), of which 21 (37.5%) of the cases were male and 35 (62.5%) were female. AKI was detected in 13 (23.2%) patients. The Cys-C level was significantly higher in AKI patients (at hospitalization p Conclusion: In this study, Cys-C and KIM-1 proved to be statistically significant to predict AKI in patients with AP. However further studies are required to support these conclusions.
