Synthesis and Cytotoxic Activity of Novel Tetrahydrocurcumin Derivatives Bearing Pyrazole Moiety

Tetrahydrocurcumin(THC)is a major metabolite of curcumin and plays an important role in curcumin-induced biological *** is a promising preventive and chemotherapeutic agent for cancer.A series of new pyrazole derivatives of THC have been synthesized as potent anticancer *** condensation of THC with various substituted hydrazines leads to new pyrazole derivatives of THC(1-18).The prepared compounds have been evaluated via in vitro MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide)assay for their cell proliferation-inhibitory activity against human lung adenocarcinoma(A549),human cervical carcinoma(HeLa)and human breast carcinoma(MCF-7)*** derivatives show significantly higher anticancer activity against all three tested cancer cell lines than the parent compound *** compounds(7,8,12,13 and 15)display promising anticancer activity against MCF-7 cell line with IC50 values ranging from 5.8 to 9.3 *** most active compound(8)is substituted with 4-bromophenyl group at the pyrazole ring and inhibits the growth of all three tested cancer cell lines with an IC50 values of(8.0 iM,A549),(9.8 iM,HeLa)and(5.8 iM,MCF-7).The obtained compounds can be a good starting point for the development of new lead molecules in the fight against cancer.

Outbreaks of Monkeypox(mpox)in over 100 non-endemic countries in 2022 represented a serious global health *** a neglected disease,mpox has become a global public health issue.A42R profilinlike protein from mpox(PDB ID:4QWO)represents a potential new lead for drug development and may interact with various synthetic and natural *** this report,the interaction of A42R profilinlike protein with six phytochemicals found in the medicinal plant Ficus religiosa(abundant in India)was *** on the predicted and compared protein–ligand binding energies,biological properties,IC_(50) values and toxicity,two compounds,kaempferol(C-1)and piperine(C-4),were *** characteristics and quantitative structure–activity relationship(QSAR)of these two compounds were determined,and molecular dynamics(MD)simulations were *** silico examination of the kaempferol(C-1)and piperine(C-4)interactions with A42R profilin-like protein gave best-pose ligandbinding energies of–6.98 and–5.57 kcal/mol,*** predicted IC_(50) of C-1 was 7.63 lM and 82 lM for *** data indicated that kaempferol and piperine are non-mutagenic,and the QSAR data revealed that piperlongumine(5.92)and piperine(5.25)had higher log P values than the other compounds *** simulations of A42R profilin-like protein in complex with C-1 and C-4 were performed to examine the stability of the ligand–protein ***/C and C-4 showed the highest affinity and activities,they may be suitable lead candidates for developing mpox therapeutic *** study should facilitate discovering and synthesizing innovative therapeutics to address other infectious diseases.

Chemometric Evaluation of the Heavy Metals in Urban Soil of Fallujah City, Iraq

To evaluate the concentrations of heavy metals and identification of their sources, 20 composite soil samples were collected randomly in Fallujah city, Iraq. To assess the pollution level, we used the soil metal index (SMI) and enrichment factor. We investigated the sources of heavy metal using the chemometric techniques such as correlation analysis, principal component analysis (PCA) and cluster analysis (CA). The concentrations of Pb and Co have a normal distribution in soil of the study area. Zn and Cd have an approximately normal distribution, and Ni, Cu, and Cr have a non-normal distribution. Our results revealed that the urban soils in Fallujah city were unpolluted by heavy metals and showed a significant to extremely high enrichments of heavy metals controlled by anthropogenic activities. The moderate to strong positive correlations among Cd, Co, Cu, Cr, Ni, Pb, and Zn suggest that these metals have common sources. The chemometric techniques identify that the source of Cu, Co and Pb is controlled by lithogenic origin and that the source of Cr, Ni and Cd is controlled by mixed sources (lithogenic and anthropogenic).

Detection of Clinically Significant Warm Reactive Anti-A1 in a Post Red Blood Cells Transfusion Patient with Acute Lymphoblastic Leukemia

The purpose of this article is to report that some anti-A1 that react at 37°C are IgG antibodies and are clinically significant, as they can cause the destruction of a proportion of A1 cells in vivo following the transfusion of red blood cells. Therefore, when a serum of an individual contains anti-A1, further testing of the plasma against group A1, A2, and O by referencing red blood cells and individual cells against anti-A1 lectin (Dolichos biflorus lectin) must be performed. Confirming the specificity of anti-A1 by referencing red blood cells is also important in selecting the appropriate blood for transfusion. ABO antibodies are naturally occurring and activate the complement cascade, making them more likely to cause severe transfusion reactions compared to antibodies to other RBC antigens.
