Herein, we exploited the biorea.tivity of novel polymer–lipid ma.ga.es. dioxide na.opa.ticles.(PLMDs. to remodel the tumor immune microenvironment (TIME) by increa.ing the loca. oxygen levels.a.d extra.ellula. pH a.d enha.cing ra.ia.ion-induced immunogenic cell dea.h. This.s.udy demons.ra.ed tha. PLMD trea.ment s.ns.tized hypoxic huma. a.d murine CRPC cells.to ra.ia.ion, s.gnifica.tly increa.ing ra.ia.ion-induced DNa.double-s.ra.d brea.s.a.d ultima.ely cell dea.h, which enha.ced the s.cretion of da.a.e-a.s.cia.ed molecula. pa.terns. a.tributa.le to the induction of a.topha.y a.d endopla.mic reticulum s.res.. Reoxygena.ion via.PLMDs.a.s. pola.ized hypoxic murine Ra.264.7 ma.ropha.es.towa.d the M1 phenotype, enha.cing tumor necros.s.fa.tor a.pha.relea.e, a.d thus.reducing the via.ility of murine CRPC TRa.P-C2 cells. In a.s.ngeneic TRa.P-C2 tumor model, intra.enous.injection of PLMDs.s.ppres.ed, while ra.ia.ion a.one enha.ced recruitment of regula.ory T cells.a.d myeloid-derived s.ppres.or cells. Pretrea.ment with PLMDs.followed by ra.ia.ion down-regula.ed progra.med dea.h-liga.d 1 a.d promoted the infiltra.ion of a.titumor CD8+ T cells.a.d M1 ma.ropha.es.to tumor s.tes. Ta.en together, TIME modula.ion by PLMDs.plus.ra.ia.ion profoundly dela.ed tumor growth a.d prolonged media. s.rviva. compa.ed with ra.ia.ion a.one. Thes. res.lts.s.gges. tha. PLMDs.plus.ra.ia.ion is.a.promis.ng trea.ment moda.ity for improving thera.eutic effica.y in ra.iores.s.a.t a.d immunos.ppres.ive s.lid tumors.
