NeCAB family of neuronal calcium-binding proteins in health and disease

The N-terminal eF-hand calcium-binding proteins 1–3(NeCAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one eF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NeCAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NeCAB2 interacts with predominantly striatal G-protein-coupled receptors, while NeCAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NeCAB proteins exhibit distinct subcellular localizations: NeCAB1 is found in the nucleus and cytosol, NeCAB2 resides in endosomes and the plasma membrane, and NeCAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NeCAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NeCAB proteins by controlling the availability of protein–protein interaction domains situated between the eF hands and the antibiotic biosynthesis monooxygenase domain. Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other *** to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma *** cell culture-based research(2d cell culture)is still playing a pivotal role,while several shortcomings have been recently under *** vivo,mouse models are usually adopted for pre-clinical analyses with expectations to overcome the issues of 2d cell ***,they do not fully recapitulate human dedifferentiated liposarcoma(ddLPS)***,three-dimensional(3d)culture systems have been the recent research focus in the cell biology field with the expectation to overcome at the same time the disadvantages of 2d cell culture and in vivo animal models and fill in the gap between *** the liposarcoma rarity,we believe that 3d cell culture techniques,including 3d cell cultures/co-cultures,and Patient-derived tumor Organoids(PdOs),represent a promising approach to facilitate liposarcoma investigation and elucidate its molecular mechanisms and effective therapy *** this review,we first provide a general overview of 3d cell cultures compared to 2d cell *** then focus on one of the recent 3d cell culture applications,Patient-derived Organoids(PdOs),summarizing and discussing several PdO ***,we discuss the current and future applications of PdOs to sarcoma,particularly in the field of liposarcoma.

Bound states in the continuum （BICs） are perfectly localized resonances despite embedding in the continuum ***,an isolated BIC is very sensitive to the structure ***,we report merging acoustic BICs in a single open resonator,robust against the structure *** find that both symmetry-protected BIC and Friedrich-Wintgen BIC are sustained in a single coupled waveguide-resonator *** varying the height and length of the resonator,these two BICs move toward each other and merge into a single one at a critical *** to an individual BIC,the merged BIC is robust against fabrication error because its Q-factor is proportional to ΔL-4,whereΔL embodies the structure *** essence of this extraordinary phenomenon is perfectly explained by the two-and three-level approximations of the effective nonHermitian ***,we present direct experimental demonstrations of the moving and merging of BICs in a coupled 3d waveguide-resonator,which are evidenced by the vanishing of the linewidth of Fano resonance in the transmission *** results may find exciting applications in designing high-quality acoustic sources,sensors and filters.

The feasibility of using N,N,N＇,N＇-tetra（2-ethylhexyl）diglycolamide （TeHdGA） in 75 vol.% n-dodecane-25 vol.% n-octanol as agents for the extraction and separation ofNd（III）, Sm（III）, dy（llI）, Fe（III）, Ni（II）, and Cs（I） from concentrated chlo- ride solution was investigated. different extraction behaviors were obtained towards rare earth elements （Ree） studied and Fe（IIl）, Ni（II） and Cs（I）. efficient separation of Nd（III）, Sm（III） and dy（III） from Fe（III）, Ni（II）, and Cs（I） was achieved by TeHdGA, depending on the HCI, HNO3 or H2SO4 concentration. A systematic investigation was carried out on the detailed extraction prop- erties of Nd（III）, Sm（III）, and dy（III） with TeHdGA from chloride media. The IR spectra of the extracted species were investi- gated.

In this paper, the problem of self-adjointness of the product of two differential operators is considered. A number of results concerning self-adjointness of the product L2L1 of two second-order self-adjoint differential operators are obtained by using the general construction theory of self-adjoint extensions of ordinary differential operators.
