Isorhamnetin downregulates MMP2 and MMP9 to Inhibit development of Rheumatoid Arthritis through SRC/ERK/CREB Pathway

Objective To investigate the effect of isorhamnetin on the pathology of rheumatoid arthritis(RA).Methods Tumor necrosis factor(TNF)-α-induced fibroblast-like synoviocytes(FLS)was exposed to additional isorhamnetin(10,20 and 40µmol/L).Overexpression vectors for matrix metalloproteinase-2(MMP2)or MMP9 or SRC were transfected to explore their roles in isorhamnetin-mediated RA-FLS function.RA-FLS viability,migration,and invasion were evaluated.Moreover,a collagen-induced arthritis(CIA)rat model was established.Rats were randomly divided to sham,CIA,low-,medium-,and high-dosage groups using a random number table(n=5 in each group)and administed with normal saline or additional isorhamnetin[2,10,and 20 mg/(kg·day)]for 4 weeks,respectively.Arthritis index was calculated and synovial tissue inflammation was determined in CIA rats.The levels of MMP2,MMP9,TNF-α,interleukin-6(IL-6),and IL-1β,as well as the phosphorylation levels of SRC,extracellular regulated kinase(ERK),and cyclic adenosine monophosphate response element-binding(CREB),were detected in RA-FLS and synovial tissue.Molecular docking was also used to analyze the binding of isorhamnetin to SRC.Results In in vitro studies,isorhamnetin inhibited RA-FLS viability,migration and invasion(Pamnetin downregulated the levels of MMP2,MMP9,TNF-α,IL-6,and IL-1βin RA-FLS(Pd isorhamnetin-inhibited RA-FLS migration and invasion,as well as the levels of TNF-α,IL-6,and IL-1β(Pamnetin bound to SRC and reduced the phosphorylation of SRC,ERK,and CREB(Pd the inhibitory effect of isorhamnetin on RA-FLS viability,migration and invasion,as well as the negative regulation of MMP2 and MMP9(Pdies,isorhamnetin decreased arthritis index scores(Pd alleviated synovial inflammation.Isorhamnetin reduced the levels of MMP2,MMP9,TNF-α,IL-6,and IL-1β,as well as the phosphorylation of SRC,ERK,and CREB in synovial tissue(P<0.05).Notably,the inhibit

Synthesis of granulated Li_(1.33)Mn_(1.67)O_(4) via two antisolvent methods for lithium adsorption from gas-produced water

Gas-produced water is an accompanying wastewater in the natural gas extraction process,and it is a potential liquid lithium resource that contains a considerable amount of lithium.This study investigated the feasibility of using manganese-based ion sieves to adsorb and extract lithium from gas-produced water.And we focused on the applicability of two different granulation methods,extrusion and droplet,in gas-produced water systems.Two types of H_(1.33)Mn_(1.67)O_(4) particles were prepared by the extrusion method(EHMO)and the droplet method(dHMO).The porosity of dHMO was much higher than that of EHMO,and the adsorption performance of dHMO increased with the decrease of binder concentration.dHMO prepared with a binder concentration of 0.14 g·ml^(-1)exhibited the best adsorption performance in gas-produced water,and the Li^(+)adsorption capacity could reach 25.14 mg·g^(-1).In gas-produced water,the adsorption equilibrium of dHMO only took 9 h,and the adsorption process conformed to the Langmuir model and pseudo-second-order kinetic model.The pore diffusion model(PdM)could well describe its adsorption process.Besides,dHMO showed a great selectivity to Li^(+),and the selectivity order of dHMO in gas-produced water was Li^(+)>Ba^(2+)>>Mg^(2+),Ca^(2+),Sr^(2+)>>Na^(+)>>K^(+).After 20 cycles,the Li^(+)adsorption capacity was still higher than 17.30 mg·g^(-1),and the rate of manganese dissolution was less than 1%.

Optimizing piezoelectric performance of complex perovskite through increasing diversity of B-site cations

Piezoelectric materials with unique properties are hugely appealing for high sensitivity sensors and ultrasonic transducers.Here,Pb(Ni_(1/3)Nb_(2/3))O_(3)-Pb(Yb_(1/2)Nb_(1/2))O_(3)-PbTiO_(3)piezoelectric system was designed and investigated in detail.The optimized piezoelectric response(d_(33)=826 pC/N,d_(33)∗=1017 pm/V)together with good temperature stability and negligible performance degradation up to 10^(6) cycles was achieved in 0.58PNN-0.10PYN-0.32PT composition.Rayleigh analysis revealed the amplification of intrinsic and extrinsic contributions in MPB region due to more spontaneous polarization orientations and an increase in chemical disorder of system after the introduction of PYN.The origin of high piezoelectric response was investigated through the combination of scanning electron microscopy and piezoresponse force microscopy.The diversity of B-site cations was shown to enhance local structural heterogeneity,weaken long-range ferroelectric ordered domains,and lower free energy barrier via the formation of nanodomain configuration that promoted polarization rotation,thereby improving piezoelectric properties of the material.Therefore,it is suggested that increasing the diversity of B-site cations is an effective strategy for achieving outstanding piezoelectric performance.

One-pot and One-step Cu(0)-mediated Reversible-deactivation Radical Polymerization of N-Isopropylacrylamide(NIPam) in Water

Copper(0)-mediated reversible-deactivation radical polymerization(Cu(0)-mediated RdRP) of the water-soluble monomer Nisopropylacrylamide(NIPam) has been challenging with the problems of high dispersity, poor control over the molecular weights(MWs) or complex or multi reaction steps, etc. In this work, we report the well-controlled polymerization of NIPam in water via a facile one-pot and one-step Cu(0)-mediated RdRP. The results of this approach show that the key for kicking off the Cu(0)-mediated NIPam RdRPs is to ensure sufficient Cu~I at the very beginning, and the key to achieve a well-controlled chain growth is to provide adequate deactivation strength during the polymerization process. For NIPam, which has a high propagation rate constant, the deactivation control can be effectively enhanced by extra adding deactivator(i.e., Cu~II) to the system. Moreover, a low reaction temperature(4 ℃) is necessary in the controlled synthesis of higher MW poly(Nisopropylacrylamide)(PNIPam) to avoid the compromise in control caused by the phase transition from its lower critical solution temperature(LCST). Through this new kinetically controlled strategy, PNIPams with well-defined structure, narrow molecular weight distributions(MWds) and varied MWs were successfully achieved.

Modulated noble metal/2d MOF heterostructures for improved hydrogen storage of MgH2

The conjugation of external species with twodimensional（2d） materials has broad application prospects.In this study,we have explored the potential of noble metal/2d MOF heterostructures in hydrogen storage.Specifically,the MgH2-Ni-MOF@Pd system has shown remarkable hydrogen desorption/sorption performances,starting to liberate hydrogen at 181 0C,which is 230 0C lower than that of pristine MgH2.Under the catalytic effect of Ni-MOF@Pd,the dehydrogenation apparent activation energy of MgH2 is noticeably decreased from（133.5 ± 17.5） to（34.58 ± 1.87） kJ·mol-1,and the hydrogenation apparent activation energy is reduced from（70.41 ± 7.43） to（25.78 ± 4.64） kJ·mol-1,which is lowered by 63.4%.The fully-dehydrogenated MgH2-NiMOF@Pd composite rapidly uptakes hydrogen,with 2.62wt% at 100℃ and 6.06 wt% at 150℃ within 300 s,respectively.The mechanism analysis of MgH2 catalyzed by Ni-MOF@Pd has revealed that the transformation of Mg2Ni and Mg2NiH4could act as a "hydrogen pump",providing numerous channels for fast diffusion and transport of hydrogen atoms.Moreover,in the dehydrogenation process,the element Pd reacts with MgH2to form the MgPd alloy phase,which makes MgH2take precedence to decompose through the Mg-Pd alloy rather than self-decomposition,further reducing thermal stability and improving de/hydrogenation kinetics.The synergistic effect of Mg-Pd,Mg2Ni,and the special ultra-thin 2d sheet structure of the additive is the main reason for the good hydrogen storage property of MgH2-Ni-MOF@Pd.Our findings provide inspiration for designing efficient multifunctional additives with unique morphologies to optimize the hydrogen desorption/sorption behaviors of hydrogen storage materials.

功能磁共振评估宫内发育迟缓仔鼠肾脏微观结构及灌注改变

目的探讨体素内不相干运动(intravoxel incoherent motion,IVIM)、纵向弛豫时间定量成像(T1 mapping)、横向弛豫时间定量成像(T2 mapping)等功能磁共振成像技术评估宫内发育迟缓(intrauterine growth restriction,IUGR)仔鼠的肾脏微观结构及灌注改变的价值。方法通过孕期低蛋白饮食法建立IUGR仔鼠模型。低蛋白饮食孕鼠所产仔鼠随机分为IUGR8周组、IUGR12周组,正常饮食孕鼠所产仔鼠随机分为正常8周组、正常12周组,各组n=8。比较各组仔鼠肾脏皮质、髓质的表观弥散系数(apparent diffusion coefficient,AdC)、真实扩散系数(d_(t))、伪扩散系数(d*)、灌注分数(f)、T1值、T2值,以及血肌酐、血尿素氮水平。结果IUGR 12周组肾脏髓质dt值高于IUGR 8周组,IUGR 12周组肾脏髓质d*值低于正常12周组与IUGR 8周组(P0.05)。各组血肌酐及血尿素氮含量比较差异无统计学意义(P>0.05)。IUGR 8周组仔鼠肾脏可见肾小球增生肥大,未见明显纤维化改变;IUGR 12周组仔鼠肾脏可见肾小球萎缩,囊腔狭窄,间质出现炎性细胞浸润及纤维化。结论IVIM磁共振成像可用于评估及动态观察IUGR仔鼠肾脏微观结构及灌注损伤,T1 mapping磁共振成像可用于评估IUGR仔鼠肾脏损伤,T1 mapping联合T2 mapping磁共振成像可进一步分辨IUGR仔鼠的肾脏纤维化。

This prospective multicenter phase II study evaluated the clinical efficacy of neoadjuvant nivolumab-exclusive(N)and nivolumab–chemotherapy(N/C)combinations based on Pd-L1 expression.Eligible patients exhibited resectable clinical stage IIA–IIIB(AJCC 8th edition)NSCLC without EGFR/ALK alterations.Patients received either mono-nivolumab(N)or nivolumab+nabpaclitaxel+carboplatin(N/C)for three cycles based on Pd-L1 expression.The primary endpoint was the major pathological response(MPR).Key secondary endpoints included the pathologic complete response(pCR),objective response rate(ORR),and event-free survival(EFS).Baseline Pd-L1 expression and perioperative circulating tumor dNA(ctdNA)status were correlated with pCR and EFS.Fifty-two patients were enrolled,with 46 undergoing surgeries.The MPR was 50.0%(26/52),with 25.0%(13/52)achieving pCR,and 16.7%and 66.7%for patients with Pd-L1≥50%in N and N/C groups,respectively.Thirteen(25.0%)patients experienced grade 3 or higher immune-related adverse events during neoadjuvant treatment.Patients with post-neoadjuvant ctdNA negativity was more likely to have pCR(39.1%)compared with those remained positive(6.7%,odds ratio=6.14,95%CI 0.84-Inf,p=0.077).With a median follow-up of 25.1 months,the 18-month EFS rate was 64.8%(95%CI 51.9–81.0%).For patients with ctdNA–vs.ctdNA+,the 18m-EFS rate was 93.8%vs 47.3%(HR,0.15;95%CI 0.04,0.94;p=0.005).Immunochemotherapy may serve as an optimal neoadjuvant treatment even for patients with Pd-L1 expression≥50%.ctdNA negativity following neoadjuvant treatment and surgery could help identify superior pathological and survival benefits,which requires further confirmation in a prospective clinical trial(NCT04015778).

Lagoons are ecosystems for biodiversity and the livelihoods of coastal communities. The main objective of the study was to analyze the variability of arsenic concentrations in gastropods and bivalves in the Aby and Tendo lagoons, taking into account spatial, seasonal and hydrological variations. The study was carried out in four stages spread over two successive hydroclimatic cycles, including two seasons during the rainy season and two more during the dry season. The samples were taken in two areas of the Aby and Tendo lagoons. Arsenic levels were measured by ICP-MS. The results showed that mean arsenic concentrations in the muscles of organisms in Aby Lagoon ranged from 0.01 to 1.26 μg As/g, with a mean and median of 0.17 and 0.06 μg As/g, respectively. Fish had the highest levels of arsenic, followed by crustaceans, while molluscs and plants had lower and comparable concentrations of As. Arsenic concentrations in tilapia and jawbones varied significantly between sites and seasons, with higher concentrations at Tendo and during the rainy season. Arsenic concentrations in gastropods and bivalves were significantly higher than those of other species, with averages of 0.74 and 1.03 mg As/kg, respectively.

接种西安市某污水处理厂缺氧池生物膜构建升流式厌氧固定床生物膜(UAFB)反应器从而富集培养厌氧氨氧化(Anammox)生物膜,通过测定生物膜Anammox活性和酶活性并进行高通量测序分析,探讨城市污水处理厂现有填料转型培养Anammox生物膜的可行性及生物膜内菌群演替规律.结果表明,经29d培养后的UAFB-Anammox反应器TN去除率高达76.22%,快速启动成功;负荷提升阶段,表面氮负荷(SNLR)由0.23g/(m^(2)·d)提升至2.59g/(m^(2)·d),最大Anammox活性维持2.15g/(m^(2)·d),TN去除率高达83.68%.Illumina MiSeq测序结果发现,富集后的生物膜中优势菌属为Ca.Brocadia,相对丰度为33.38%,富集效果明显.同时缺氧生物膜上反硝化菌denitratisoma以Anammox的产物NO_(3)^(-)-N为基质,逐渐与AnAOB形成共生存关系.这说明利用城市污水处理厂现有缺氧生物膜可以快速培养Anammox生物膜,对Anammox工艺在城市污水处理厂升级改造中的应用提供一定参考.

Physico-Chemical and Nutritional Properties of a Food Broth Based on Nere (Parkia biglobosa)

Purpose: diet and eating habits are major risk factors for the health and the development of disease, such as, for example, metabolic disorder leading to cardiovascular pathology and cancer, decreased immunity exposing to infections. This study of the physico-chemical and nutritional properties of a soumara-based food broth was carried out with the aim of promoting the consumption of organic broth made from nere seeds (soumara). That is to alleviate certain metabolic diseases, which is a matter of food safety, and also to limit the risk for the health about the consumption of some cooking stocks on the market. Methods: Several natural ingredients such as nere seeds (soumara), ginger, black pepper, parsley and garlic were used to create a nere-based stock. All these ingredients were freeze-dried and the powder obtained was used to make the broth, regarding their physical and chemical properties. Results: The broth had a good protein content of 17.41 ± 0.367 g/100g, a lipid content of 16.80 ± 0.08 g/100g and a fiber content of 8.66 ± 0.04 g/100g. In terms of nutritional values, the broth showed good levels of calcium 184.21 ± 0.09 mg/100g, potassium 50.04 ± 1.45 mg/100g and iron and zinc. In terms of antioxidant activity, the broth also showed good antioxidant activity. Conclusion: Regarding the properties of our food broth, whose composition is based on natural ingredients, could be recommended for consumption and, its properties, could play an important role in preventing and combating certain metabolic diseases.
