Colorectal cancer remains a leading malignancy in humans. The importance of epigenetic modification in the development of this disease is now being recognized. The reversible and dynamic nature of epigenetic modifications provides a promising strategy in colorectal cancer chemoprevention and treatment. Luteolin(LUT), a flavone dietary phytochemical, can modulate various signaling pathways involved in carcinogenesis. many studies have demonstrated that LUT inhibits colorectal carcinogenesis by activating the Nrf2/ARE pathway. However, the potential epigenetic mechanism underlying Nrf2/ARE pathway activation remains unclear. In this study, we aimed to explore the anticancer potential of LUT in colorectal adenocarcinoma HCT116 cells and the epigenetic regulation of the Nrf2/ARE pathway. Specifically, our data showed that LUT suppressed cell proliferation and cellular transformation of HCT116 cells in a dose-dependent manner. Additionally, qPCR and western blotting were applied to determine the mRNA and protein expression of Nrf2 and its downstream genes after LUT treatment. Bisulfite genomic sequencing revealed that methylation of the Nrf2 promoter region was decreased by LUT, corresponding with the increased m RNA expression of Nrf2. Decreased protein levels and enzyme activities of epigenetic modifying enzymes, such as DNmTs and HDACs, were also observed in LUT-treated HCT116 cells. In summary, our findings suggest that LUT may exert its anti-tumor activity in part via epigenetic modifications of the Nrf2 gene with subsequent induction of its downstream antioxidative stress pathway.

A practical and effective approach to increase the energy storage capacity of lithium ion batteries(LIBs)is to boost their areal *** thick electrodes is one of the most crucial ways to achieve high areal capacity but limited by sluggish ion/electron transport,poor mechanical stability,and high-cost manufacturing *** we address these constraints by engineering a unique hierarchical-networked 10 mm thick all-carbon electrode,providing a scalable strategy to produce high areal capacity LIB *** hierarchical-networked structure utilizes micrometer-sized carbon fibers(mCFs)as building blocks,nano-sized carbon nanotubes(CNTs)as good continuous network with excellent electrical conductivity,and pyrolytic carbon as the binder and active material with excellent storage *** combination of the above features endows our HNT-mCF/CNT/PC electrode with excellent performance including high reversible capacity of 15.44 mah cm^(-2) at 2.0 mA cm^(-2) and exhibits excellent rate capability of 2.50 mah cm^(-2) under 10.0 mA cm^(-2) current *** Li-ion storage mechanism in HNT-mCF/CNT/PC involves dual-storage mechanism including intercalation and surface adsorption(pseudocapacitance)confirmed by the cyclic voltammetry and symmetric cell *** work provides insights into the construction of high mechanical stability thick electrode for the next generation high areal capacity LIBs and beyond.

multiple sclerosis(mS)is an inflammatory neurodegenerative disease of the central nervous system(CNS).Pathological characteristics of the disease include activation of CNS-intrinsic immune cells,such as microglia and astrocytes,and loss of neuronal connections,myelin and blood-brain barrier(BBB)integrity as well as peripheral immune cell infiltration into the *** has long been considered a predominantly immunological disease,which has led to the development of essentially only immune-directed *** this traditional“outside-in”mS hypothesis,a dysregulation of the peripheral immune system causes immune cell infiltration into the CNS,leading to autoreactivity against myelin sheath components and secondary BBB ***,recent findings indicate that overactivation of microglia and astrocytes represents an important first step in mS pathology,as appears to be the case for other neurodegenerative diseases such as Alzheimer’s disease(AD)and Parkinson’s disease(PD).

AIm:To evaluate the clinical efficacy and safety of epirubicin,cisplatin,and 5-FU combination chemotherapy for the sorafenib-refractory metastatic hepatocellular carcinoma(HCC).mETHODS:From April 2009 to June 2012,31 patients who were diagnosed with metastatic and progressive HCC after sorafenib treatment were retrospectively *** were treated with the combination of epirubicin(50 mg/m2Ⅳ;day 1),cisplatin(60 mg/m2Ⅳ;day 1),and 5-FU(1000 mg/m2Ⅳ;day 1-3)[Epirubicin,cisplatin,5-FU combination(ECF)],repeated every 4 ***:The overall response rate was 12.9%.Patients who responded to ECF chemotherapy showed a longer overall survival(OS)and time to progression(TTP)relative to those in the non-responder group(OS:20.4 mo vs 4.9 mo,Pmo vs 2.2 mo,Pmary liver mass also exhibited a longer OS and TTP relative to those with progressive disease(OS:13.4 mo vs 5.3 mo,P=0.003;TTP:9.4 mo vs 2.3 mo,P=0.003).The most common hematologic toxicity was thrombocytopenia(87.2%),and the incidence of grade 3-4 neutropenia was 53.9%.Age older than 60,a stable primary mass,and a good response to chemotherapy were prognostic factors for OS and ***:This combination cytotoxic chemotherapy can serve as another treatment option after sorafenib failure for the subset of patients with advanced metastatic HCC.

To assess the effect of strain and strain rate on texture evolution of an extruded mg–6Al–3Zn alloy,compression tests were carried *** were prepared in the extrusion direction(ed)and normal direction(ND).The compression tests were performed at 250℃ and with different strain rates of 0.01 sec−1 and 1 sec−1 and different *** observation and texture investigation show that at early stages of deformation,extension twins lead to the development of strong basal texture intensity along rolling direction(RD)in ed samples and contraction twins result in texture evolution along transverse direction(TD)in ND ***,microstructural investigation at high strains reveals that dynamic recrystallization occurs in both samples and consequently the basal texture intensity has been decreased.

mechanical, electrical, and thermal properties of the directionally solidified Bi–Zn–Al ternary eutectic alloy

A Bi-2.0Zn-0.2A1 （wt%） ternary eutectic alloy was prepared using a vacuum melting furnace and a casting furnace. The samples were directionally solidified upwards at a constant growth rate （V= 18.4 μm/s） under different temperature gradients （G = 1.15-3.44 K/mm） and at a constant temperature gradient （G = 2.66 K/mm） under different growth rates （V= 8.3-500 μm/s） in a Bridgman-type directional so- lidification furnace. The dependence ofmicrostructure parameter （2） on the solidification parameters （G and V） and that of the microhardness （Hv） on the microstructure and solidification parameters were investigated. The resistivity （ρ） measurements of the studied alloy were per- formed using the standard four-point-probe method, and the temperature coefficient of resistivity （α） was calculated from the ρ-Tcurve. The enthalpy （ah） and the specific heat （Cp） values were determined by differential scanning calorimetry analysis. In addition, the thermal conductivities of samples, obtained using the Wiedemann-Franz and Smith-Palmer equations, were compared with the experimental results. The results revealed that, the thermal conductivity values obtained using the Wiedemarm-Franz and Smith-Palmer equations for the Bi-2.0Zn-0.2Al （wt%） alloy are in the range of 5.2-6.5 W/Km and 15.2-16.4 W/Km, respectively.

We compared the efficacy and safety between once-daily dosing and on-demand use of udenafil for type 2 diabetic patients with erectile dysfunction （ed）. A multi-center, randomized, open-label, parallel-group, 12-week study was conducted. 161 patients who improved with on-demand 200 mg of udenafil according to Sexual Encounter Profile （SEP） diary Question 2 and 3 （Q2 and Q3） were randomized into 200 mg on-demand （n = 80） or 50 mg once-daily （n = 81） dosing groups for 8 weeks. The dosing period was followed by a 4-week treatment-free period. The primary efficacy endpoint was the change of the International Index of Erectile Function （IIEF） erectile function domain （EFD） score. The secondary efficacy endpoints included changes to the SEP diary Q2, Q3, IIEF Q3, Q4, other domains of IIEF, Global Assessment Question, and shift to the normal rate （EFD 〉 26）. Vascular endothelial markers were also assessed. The IIEF-EFD score of both groups improved after 8 weeks of treatment （P 〈 0.0001）. There was no statistically significant difference between two groups. Improvement was not maintained after the treatment-free follow-up period. Similar results were observed in the secondary efficacy endpoints. There was also no significant difference in vascular endothelial markers. Daily udenafil was well-tolerated, and there was no significant difference in the adverse drug reactions and adverse events between the two groups. Flushing and headache were the most frequent adverse events. Both regimens improved ed in diabetic patients and were well-tolerated. Further studies are needed to assess the effect of daily udenafil treatment in diabetic patients.
