CURlY lEAF ( ClF) encodes the methyltransferase subunit of the Polycomb Repressor Complex 2 (PRC2), which regulates the expression of target genes through H3K27 trimethylation. We isolated a new ClF mutant allele ( clf-78) using a genetic screen designed to identify microRNA (miRNA) deficient mutants. ClF mutant plants showed impaired miRNA activity caused by increased ubiquitination and enhanced degradation of ARGONAUTE 1 (AGO1) in specific tissues. Such ClF-mediated AGO1 regulation was evident when plants were exposed to UV radiation, which caused increased susceptibility of clf mutants to some UV-induced responses. Furthermore, we showed that ClF directly regulates FBW2, which in turn triggers AGO1 degradation in the clf mutants. Interestingly, AGO1 bound to a target appeared particularly prone to degradation in the mutant plants, a process that was exacerbated when the complex bound a non-cleavable target. Thus, prolonged AGO1–target interaction seems to favor AGO1 degradation, suggesting that non-cleavable miRNA targets may overcome translation inhibition by modulating AGO1 stability in plants.

Tomato production is influenced by shoot branching,which is controlled by different *** we produced tomato plants overexpressing the cytokinin-deactivating gene CYTOKININ OXYDASE 2(CKX2).CKX2-overexpressing(CKX2-OE)plants showed an excessive growth of axillary shoots,the opposite phenotype expected for plants with reduced cytokinin content,as evidenced by lC-MS analysis and ARR5-GUS *** TCP transcription factor SlBRC1b was downregulated in the axillary buds of CKX2-OE and its excessive branching was dependent on a functional version of the GRAS-family gene lATERAl SUPPRESSOR(lS).Grafting experiments indicated that increased branching in CKX2-OE plants is unlikely to be mediated by root-derived *** CKX2-OE plants with transgenic antisense plants for the strigolactone biosynthesis gene CAROTENOID ClEAVAGE DIOXYGENASE(CCD7-AS)produced an additive phenotype,indicating independent effects of cytokinin and strigolactones on increased *** the other hand,CKX2-OE plants showed reduced polar auxin transport and their bud outgrowth was reduced when combined with auxin ***,CKX2-OE basal buds did not respond to auxin applied in the decapitated *** results suggest that tomato shoot branching depends on a fine-tuning of different hormonal balances and that perturbations in the auxin status could compensate for the reduced cytokinin levels in CKX2-OE plants.

The isohydricity (pH constancy) as the property referred to mixtures of acids or bases, is illustrated on a simple example of the solutions: Hl (C0, mol/l) and Hl (C mol/l) mixed according to titrimetric mode (pH titration). A new derivation of the Michalowski formula C0=C+C2·10PK1 expressing this property is presented, and its applicability for determination of pK1 = – logK1 value is indicated. The principle of the isohydric method of pK1 determination is also outlined.
