AIM： To investigate the effect of stable c-Fos over- expression on immortalized human hepatocyte （IHH） proliferation. METHODs： IHHs stably transfected with c-Fos （IHH- Fos） or an empty vector （IHH-C） were grown in me- dium supplemented with 1% serum or stimulated with 10% serum. Cell proliferation was assessed by cell counts, 3H-thymidine uptake and flow cytometry analyses. The levels of cell cycle regulatory proteins （Cyclin DI, E, A） cyclin dependent kinases （cdk） cdk2, cdk4, cdk6, and their inhibitors p15, p16, p21, p27, total and phosphorylated GsK-3β and epidermal growth factor receptor （EGF-R） were assayed by Western blotting. Analysis of O/c/in D1 mRNA levels was performed by reverse transcription-polymerase chain reaction and real-time polymerase chain reaction （PCR） analysis. stability of Cyclin DI was studied by cycloheximide blockade experiments. REsULTs： stable c-Fos overexpression increased cell proliferation under low serum conditions and resulted in a two-fold increase in [3H]-thymidine incorpora- tion following serum addition. Cell cycle analysis by flow cytometry showed that c-Fos accelerated the cell cycle kinetics. Following serum stimulation, Cyclin D1 was more abundantly expressed in c-Fos overexpress- ing cells. Cyclin D1 accumulation did not result from increased transcriptional activation, but from nuclear stabilization. Overexpression of c-Fos correlated with higher nuclear levels of inactive phosphorylated GsK- 3β, a kinase involved in Cyclin D1 degradation and higher levels of EGF-R mRNA, and EGF-R protein com- pared to IHH-C both in serum starved, and in serum stimulated cells. Abrogation of EGF-R signalling in IHH- Fos by treatment with AG1478, a specific EGF-R tyro- sine kinase inhibitor, prevented the phosphorylation of GsK-3β induced by serum stimulation and decreased Cyclin D1 stability in the nucleus. The ease of accessing a virtually unlimited pool of resources makes Infrastructure as a service (Iaas) clouds an ideal platform for running data-intensive workflow applications comprising hundreds of computational tasks. However, executing scientific workflows in Iaas cloud environments poses significant challenges due to conflicting objectives, such as minimizing execution time (makespan) and reducing resource utilization costs. This study responds to the increasing need for efficient and adaptable optimization solutions in dynamic and complex environments, which are critical for meeting the evolving demands of modern users and applications. This study presents an innovative multi-objective approach for scheduling scientific workflows in Iaas cloud environments. The proposed algorithm, MOs-MWMC, aims to minimize total execution time (makespan) and resource utilization costs by leveraging key features of virtual machine instances, such as a high number of cores and fast local ssD storage. By integrating realistic simulations based on the WRENCH framework, the method effectively dimensions the cloud infrastructure and optimizes resource usage. Experimental results highlight the superiority of MOs-MWMC compared to benchmark algorithms HEFT and Max-Min. The Pareto fronts obtained for the Cybershake, Epigenomics, and Montage workflows demonstrate closer proximity to the optimal front, confirming the algorithm’s ability to balance conflicting objectives. This study contributes to optimizing scientific workflows in complex environments by providing solutions tailored to specific user needs while minimizing costs and execution times.

When, how often and for how long organisms mate can have strong consequences for individual fitness and are crucial aspects of evolutionary ecology. such determinants are likely to be of even greater importance in monandrous species and species with short adult life stages. Previous work suggests that mobility, a key dispersal? related trait, may affect the dynamics of copulations, but few studies have investigated the impact of individual mobility on mating latency, copulation duration and oviposition latency simultaneously. In this paper, we monitored the copulation dynamics of 40 males and 40 females, as well as the oviposition dynamics of the females of the Large White butterfly Pieris brassicae, a facultative long-distance disperser butterfly. Individuals from a breeding were selected to create a uniform distribution of mobility and we recorded the timing, number and duration of all copulations in a semiexperimental system. We showed that mobility, measured as the time spent in flight under stressful conditions (a proxy of dispersal tendency), correlates with all aspects of copulation dynamics: mobile males and females mated earlier and for shorter periods than less mobile individuals. In turn, late mating females increased the time between copulation and oviposition. These results feed the previously described mobility syndrome of R brassicae, involving morphological and physiological characters, with life-history traits. We suggest that the reduction of mating latency and copulation duration has an adaptive value in dispersing individuals, as their life expectancy might be shorter than that of sedentary individuals.

Trans-acting factors controlling mRNA fate are critical for the post-transcriptional regulation of inflammation-related genes, as well as for oncogene and tumor suppressor expression in human cancers. Among them, a group of RNA-binding proteins called "Adenylate-Uridylate-rich elements binding proteins"(AUBPs)control mRNA stability or translation through their binding to AU-rich elements enriched in the 3'UTRs of inflammation-and cancer-associated mRNA transcripts. AUBPs play a central role in the recruitment of target mRNAs into small cytoplasmic foci called Processing-bodies and stress granules(also known as P-body/sG). Alterations in the expression and activities of AUBPs and Pbody/sG assembly have been observed to occur with colorectal cancer(CRC)progression, indicating the significant role AUBP-dependent post-transcriptional regulation plays in controlling gene expression during CRC ***, these alterations contribute to the pathological expression of many early-response genes involved in prostaglandin biosynthesis and inflammation,along with key oncogenic pathways. In this review, we summarize the current role of these proteins in CRC development. CRC remains a major cause of cancer mortality worldwide and, therefore, targeting these AUBPs to restore efficient post-transcriptional regulation of gene expression may represent an appealing therapeutic strategy.

为克服既有原子力显微镜(AFM)悬臂式探针的谐振频率难以超过3.5 MHz,Q值低、成像速度低及在液体中成像效果欠佳等缺点,设计并制作了一种基于微机电系统(MEMs)谐振器的I2形探针,并成功实现了成像实验。为更进一步提高I2形探针的力灵敏度,从结构设计和检测方法入手,对原有探针进行改进。设计了简单有效的电阻差分检测方法,成功去处了输入端与输出端的耦合现象;采用局部离子注入的方法,提高了压阻敏感的传输效率。实验结果显示,在输入信号相同的条件下,改进后探针的输出信号与原有探针相比,输出信号得到了明显提高。计算结果表明,采用新方法制作的探针可使力灵敏度提高10倍以上。

Cladosporium cladosporioides is an emerging pathogenic fungus that causes Cladosporium rot in postharvest table grapes(Vitis vinifera). However,studies investigating the infection process of C. cladosporioides are lacking. Therefore, this study aimed to elucidate the infection process by investigating the influence of microclimatic parameters(temperature, wetness and fungal age) in C. cladosporioides pathogenesis, activities of grape defense-related enzymes and grape quality during the infection. C. cladosporioides effectively infects grapes by developing distinct colonies on the artificial wounds and berry surfaces, completing its life cycle within 48 h. The C. cladosporioides disease incidence optimally occurred at 20 ℃ and 25 ℃. Wetness played an influential role in the infectivity of C. cladosporioides and 7-day-old C. cladosporioides resulted in the most serious disease incidence of table grapes. As a result of infection, the quality of grapes was affected, including berry weight, p H, titratable acidity(TA), total soluble solids(Tss), and ascorbic acid level. This infection also induced defense-related enzymes, including polyphenoloxidase(PPO), peroxidase(POD), phenylalanine ammonialyase(PAL), and catalase(CAT), at certain times. The findings of this study demonstrated that Cladosporium rot development depended on the microclimatic parameters of grapes, significantly affected the grape quality and induced grape's defense-related enzymes.

Association between Family Environment and sexual Behaviour of Adolescents in Burkina Faso

studies conducted in Africa have demonstrated the existence of a significant association between family environment and the sexual behaviour of adolescents. Generally, empirical studies of this topic are based on parental control, and family or conjugal instability and socialization approaches. The objective of this study is to assess the association between family environment and the sexual behaviour of adolescents aged 12 to 19 years. The 2004 National survey of Adolescents (NsA) in Burkina Faso provides data for studying operationalization. Analyses were bivariate and multivariate. The former were conducted using cross tabulations while the latter used three logistic regression models. Results indicate a significant but low association among family environment variables. Absence of both parents, or a number of individuals under 20 years old higher than the number of adults in a household is not systematically associated with risky sexual behaviour. Parental control was shown to be more discriminating of sexual behaviour of adolescents from Burkina Faso, compared with communication with family members regarding sexuality. Despite current ongoing economic crises and sociocultural mutations in Burkinabe societies, household members and family still play major roles in adolescents’ education. Results reinforce sexual and reproductive health programs where parents and household members are central to strategies.
