AIM： To construct formulae for predicting the likelihood of ribavirin-induced anemia in pegylated interferon plus ribavirin for chronic hepatitis C. METHODS： Five hundred and sixty-one Japanese patients with hepatitis C virus genotype lb who had received combination treatment were enrolled and as- signed randomly to the derivation and confirmatory groups. Single nucleotide polymorphisms at or nearby ITPA were genotyped by real-time detection poly- merase chain reaction. Factors influencing significant anemia （hemoglobin concentration 〈 10.0 g/dL at week 4 of treatment） and significant hemoglobin decline （declining concentrations 〉 3.0 g/dL at week 4） were analyzed using multiple regression analyses. Prediction formulae were constructed by significantly independent factors. RESULTS： Multivariate analysis for the derivation group identified four independent factors associated with significant hemoglobin decline： hemoglobin decline at week 2 [P = 3.29× 10^4, odds ratio （OR） = 7.54 （g/dL）], estimated glomerular filtration rate [P = 2.16× 10^4, OR = 0.962 （ml/min/1.73 m2）], rs1127354 （P = 5.75 × 10^4, OR = 10.94） and baseline hemoglobin [P = 7.86 × 10^4, OR = 1.50 （g/alL）]. Using the model constructed by these factors, positive and negative predictive values and predictive accuracy were 79.8%, 88.8% and 86.2%, respectively. For the confirmatory group, they were 83.3%, 91.0% and 88.3%. These factors were closely correlated with significant anemia. However, the model could not be constructed, because no patients with rs1127354 minor genotype CA/AA had significant anemia. CONCLUSION： Reliable formulae for predicting the likelihood of ribavirin-induced anemia were constructed. Such modeling may be useful in developing individual tailoring and optimization of ribavirin dosage.

Reaction crystallization of struvite in water solutions containing 0.20 mass % of phosphate(V) ions by magnesium and ammonium ions addition was investigated experimentally. Process was carried out in DTM type crystallizer with liquid jet pump device in 298 K assuming stoichiometric conditions. Struvite crystals of mean size Lm 5.2-23.0 μm were produced depending on pH (9-11) and mean residence time of suspension in a crystallizer τ (900-3600 s). Under these conditions linear growth rate of struvite crystals (SIG MSMPR kinetic model) decreased 2-time with the increase in pH and 3-time with the elongation of mean residence time of crystal suspension from 7.11×10-9 m/s (pH 9, τ900 s) to 1.65×10-9 m/s (pH 11, τ3600 s). Nucleation rate varied within the 7.9×108-1.8×1010 1/(sm3) limits. Struvite product of maximal linear size exceeded 100 μm with 10 vol. % of < 3 μm fraction corresponded to pH 9 and τ3600 s.

The denominations of origin being perceived by the consumers in terms of both intrinsic qualities and of extrinsic qualities, have contributed in no uncertain manner to the increase of the typical products in question, positively influenced also by the noted emergencies coming from verified alimentary alarms during the beginning of the XXI century. The objective of this paper, is to determine the habits of purchase of typical and traditional products of the people of Messina （Sicily, Italy）. To do this a ＂motivational＂ survey was carried out by traditional methodology of descriptive researches. The adopted methodology of research was quantitative and random. The random research strives to identify the factors that depend on the behaviour of purchase and the appraisal of the existing relationships of cause-effect in a determined population. The interviewed were given a questionnaire composed of 17 questions on their knowledge of the typical products characteristics, the frequency of consumption, the monthly amounts destined to the purchase, the motivations that push them to the purchase of typical products rather than pre-packed. Methodologically, our statistical analysis has been performed using the log-likelihood ratio test in order to investigate the existence of association between the joint distributions of qualitative measured variables.

Serum chromogranin-A in hepatocellular carcinoma: Diagnostic utility and limits

AIM: The utility of serum alpha-fetoprotein (α-FP) for the detection of hepatocellular carcinoma (HCC) is questionable. High serum levels of chromogranin-A (CgA) have recently been reported in HCC. Impaired hepatic, renal, and heart functions influence circulating CgA. The aim of this study was to assess sensitivity and specificity of serum CgA as a marker of HCC in patients with liver cirrhosis (LC). METHODS: Serum CgA levels were measured by RIA in 339 patients of which 54 HCC, 132 LC, 45 chronic hepatitis (CH), 27 chronic heart failure (CHF), 36 chronic renal failure (CRF), 45 chronic inflammatory bowel disease (IBD) as disease controls and in 75 healthy controls. Patients with liver disease or IBD and concomitant renal and/or heart failure were excluded. Pearson correlation, non-parametric combination test and confidence interval analysis were used for statistical analysis. RESULTS: Serum CgA above normal values (100 ng/mL) were found in 83% of HCC patients, in 48% of LC patients, in 20% of CH patients, in 33% of IBD patients, in 92% of CRF patients, in 100% of CHF patients, and in none of the healthy controls. The mean CgA values in HCC (769±1046), in LC (249±369), in CH (87±94), in CRF (1390±1401), in CHF (577±539), in IBD (146±287) were significantly higher than those in healthy controls (48±18). HCC patients had higher CgA values (P200 ng/mL in 21% of the HCC patients and in none of the LC patients. No significant correlation was found between a-FP and CgA in patients with HCC and in patients with cirrhosis. CONCLUSION: When HCC is suspected and a-FP

The meiotic cohesin complex of S. cerevisiae shares with the mitotic one the Irr1/Scc3, Smc1, and Smc3 subunits, while the meiosis-specific subunit Rec8 replaces mitotic subunit Scc1/Mcd1. We noticed earlier that the irr1-1 mutation (F658G) severely affected meiosis. The irr1 1/IRR1 cells were entering meiosis before having completed mitotic cell division. Using meiotic two-hybrid assay and co-immunoprecipitation we show that in cells arrested in pachytene due to a lack of a gene-regulatory factor Ndt80, the Irr1 protein interacts with Rec8p and the irr1-1 mutation abolishes this interaction. These findings indicate an important role of Irr1p in early stages of meiosis.

Understanding the molecular mechanism of the protein assembly still remains a challenge in the case of many biological systems. In this frame, the mechanism which drives RodA hydrophobins to self-assemble onto the surface of the conidia of the human fungal pathogen Aspergillus fumigatus into highly ordered nanorods known as rodlets, is still unresolved. Here, AFM investigations were combined with Monte Carlo simulations to elucidate how these small amphiphilic proteins self-assemble into tightly packed rodlets and how they are further organized in nanodomains. It becomes that the assembly of RodA hydrophobins into rodlets and their parallel alignment within nanodomains result from their anisotropic properties. Monte Carlo simulations allowed us to confirm that anisotropic interactions between macromolecules are sufficient to drive them to assembly into rodlets prior to nanodomains formation. Better knowledge of the mechanism of hydrophobins assembly into rodlets offers new prospects for the development of novel strategies leading to inhibition of rodlet formation, which should allow more rapid detection of the conidia by the immune system.

Precipitation and Crystallization of Struvite from Synthetic Wastewater under Stoichiometric Conditions

Phosphate (V) ions were continuously removed from synthetic wastewater containing inorganic impurities using magnesium and ammonium ions. The product was magnesium ammonium phosphate (V) hexahydrate, struvite, MgNH4PO4 × 6H2O. Research ran in stoichiometric conditions in DT MSMPR type crystallizer with internal circulation of suspension. Increase in process environment pH from 9 to 11 resulted in 3-time decrease of mean struvite crystals size (from 40.1 to12.6mm). Elongation of mean residence time of suspension in a crystallizer up to 3600 s resulted in improvement of the product quality. Mean size of struvite crystals enlarged up to50.2mm. Based on kinetic calculations results (SIG MSMPR model) it was concluded, that linear struvite crystal growth rate varied within 5.04 × 10–9 – 1.69 × 10–8 m/s range, whereas nucleation rate within 1.4 × 107 – 1.7 × 1010 1/(s m3) limits. In solid product, besides struvite, also all impurities present in wastewater were identified analytically as hydroxides, phosphates and other salts.

Ethylene plays a crucial role in various biological processes and therefore its biosynthesis is strictly regu- lated by multiple mechanisms. Posttranslational regulation, which is pivotal in controlling ethylene biosynthesis, impacts 1-aminocyclopropane 1-carboxylate synthase （ACS） protein stability via the complex interplay of specific factors. Here, we show that the Arabidopsis thaliana protein phosphatase type 2C, ABI1, a negative regulator of abscisic acid signaling, is involved in the regulation of ethylene biosynthesis under oxidative stress conditions. We found that ABI1 interacts with ACS6 and dephosphorylates its C-terminal fragment, a target of the stress-responsive mitogen-activated protein kinase, MPK6. In addition, ABI1 controls MPK6 activity directly and by this means also affects the ACS6 phosphorylation level. Consistently with this, ozone-induced ethylene production was significantly higher in an ABI1 knockout strain （abiltd） than in wild-type plants. Importantly, an increase in stress-induced ethylene production in the abiltd mutant was compen- sated by a higher ascorbate redox state and elevated antioxidant activities. Overall, the results of this study provide evi- dence that ABI1 restricts ethylene synthesis by affecting the activity of ACS6. The ABI1 contribution to stress phenotype underpins its role in the interplay between the abscisic acid （ABA） and ethylene signaling pathways.
