Treatment with a JAK1/2 inhibitor ameliorates murine autoimmune cholangitis induced by IFN overexpression

The interferon(IFN)signaling pathways are major immunological checkpoints with clinical significance in the pathogenesis of autoimmunity.We have generated a unique murine model named ARE-Del,with chronic overexpression of IFNγ,by altering IFNγmetabolism.Importantly,these mice develop an immunologic and clinical profile similar to patients with primary biliary cholangitis,including high titers of autoantibodies and portal inflammation.We hypothesized that the downregulation of IFN signaling pathways with a JAK1/2 inhibitor would inhibit the development and progression of cholangitis.To study this hypothesis,ARE-Del^(+/−)mice were treated with the JAK1/2 inhibitor ruxolitinib and serially studied.JAK inhibition resulted in a significant reduction in portal inflammation and bile duct damage,associated with a significant reduction in splenic and hepatic CD4^(+)T cells and CD8^(+)T cells.Functionally,ruxolitinib inhibited the secretion of the proinflammatory cytokines IFNγand TNF from splenic CD4^(+)T cells.Additionally,ruxolitinib treatment also decreased the frequencies of germinal center B(GC B)cells and T follicular helper(Tfh)cells and led to lower serological AMA levels.Of note,liver and peritoneal macrophages were sharply decreased and polarized from M1 to M2 with a higher level of IRF4 expression after ruxolitinib treatment.Mechanistically,ruxolitinib inhibited the secretion of IL-6,TNF and MCP1 and the expression of STAT1 but promoted the expression of STAT6 in macrophages in vitro,indicating that M1 macrophage polarization to M2 occurred through activation of the STAT6-IRF4 pathway.Our data highlight the significance,both immunologically and clinically,of the JAK/STAT signaling pathway in autoimmune cholangitis.

AMachine Learning Approach to User Profiling for Data Annotation of Online Behavior

The user’s intent to seek online information has been an active area of research in user profiling.User profiling considers user characteristics,behaviors,activities,and preferences to sketch user intentions,interests,and motivations.Determining user characteristics can help capture implicit and explicit preferences and intentions for effective user-centric and customized content presentation.The user’s complete online experience in seeking information is a blend of activities such as searching,verifying,and sharing it on social platforms.However,a combination of multiple behaviors in profiling users has yet to be considered.This research takes a novel approach and explores user intent types based on multidimensional online behavior in information acquisition.This research explores information search,verification,and dissemination behavior and identifies diverse types of users based on their online engagement using machine learning.The research proposes a generic user profile template that explains the user characteristics based on the internet experience and uses it as ground truth for data annotation.User feedback is based on online behavior and practices collected by using a survey method.The participants include both males and females from different occupation sectors and different ages.The data collected is subject to feature engineering,and the significant features are presented to unsupervised machine learning methods to identify user intent classes or profiles and their characteristics.Different techniques are evaluated,and the K-Mean clustering method successfully generates five user groups observing different user characteristics with an average silhouette of 0.36 and a distortion score of 1136.Feature average is computed to identify user intent type characteristics.The user intent classes are then further generalized to create a user intent template with an Inter-Rater Reliability of 75%.This research successfully extracts different user types based on their preferences in online content,platforms,crit

A Review on the Recent Trends of Image Steganography for VANET Applications

Image steganography is a technique of concealing confidential information within an image without dramatically changing its outside look.Whereas vehicular ad hoc networks(VANETs),which enable vehicles to communicate with one another and with roadside infrastructure to enhance safety and traffic flow provide a range of value-added services,as they are an essential component of modern smart transportation systems.VANETs steganography has been suggested by many authors for secure,reliable message transfer between terminal/hope to terminal/hope and also to secure it from attack for privacy protection.This paper aims to determine whether using steganography is possible to improve data security and secrecy in VANET applications and to analyze effective steganography techniques for incorporating data into images while minimizing visual quality loss.According to simulations in literature and real-world studies,Image steganography proved to be an effectivemethod for secure communication on VANETs,even in difficult network conditions.In this research,we also explore a variety of steganography approaches for vehicular ad-hoc network transportation systems like vector embedding,statistics,spatial domain(SD),transform domain(TD),distortion,masking,and filtering.This study possibly shall help researchers to improve vehicle networks’ability to communicate securely and lay the door for innovative steganography methods.

Gravity-driven powder flow and the influence of external vibration on flow characteristics

The controlled and homogeneous flow of dry granular powders through hoppers is essential for applications,namely,packaging of food grains,fertilizers and additive manufacturing processes such as directed energy deposition for better product quality.One of the major issues encountered in the granular flows through hoppers is flow stagnation due to the well-known arching phenomenon.Vibration-assisted granular flow through hoppers is one of the mechanisms used for better mass flow control.In this work,the influence of external mechanical vibration on the powder flow is investigated experimentally and using discrete element simulations.First,the mass flow rate through the hopper increases with an increase in vibration amplitude and then decreases,signifying the existence of an optimal amplitude of vibration.The DEM simulations explained the underlying mechanisms for the existence of an optimal amplitude of vibration corresponding to the maximum mass flow rate.A range of vibration amplitudes from 0 mm to 3.5 mm is used to study the flow behaviour;the maximum flow of around 33 g/s to 35 g/s is observed for 0.75 mm to 1.25 mm vibration amplitude for the hopper-particle combination studied in this work.The work also reports the influence of vibration frequency,hopper,and particle dimension on the flow characteristics.The research facilitates the effective use of mechanical vibration to enhance powder flow that can further be extended to non-spherical and multi-material particles.

Real-Time Prediction of Urban Traffic Problems Based on Artificial Intelligence-Enhanced Mobile Ad Hoc Networks(MANETS)

Traffic in today’s cities is a serious problem that increases travel times,negatively affects the environment,and drains financial resources.This study presents an Artificial Intelligence(AI)augmentedMobile Ad Hoc Networks(MANETs)based real-time prediction paradigm for urban traffic challenges.MANETs are wireless networks that are based on mobile devices and may self-organize.The distributed nature of MANETs and the power of AI approaches are leveraged in this framework to provide reliable and timely traffic congestion forecasts.This study suggests a unique Chaotic Spatial Fuzzy Polynomial Neural Network(CSFPNN)technique to assess real-time data acquired from various sources within theMANETs.The framework uses the proposed approach to learn from the data and create predictionmodels to detect possible traffic problems and their severity in real time.Real-time traffic prediction allows for proactive actions like resource allocation,dynamic route advice,and traffic signal optimization to reduce congestion.The framework supports effective decision-making,decreases travel time,lowers fuel use,and enhances overall urban mobility by giving timely information to pedestrians,drivers,and urban planners.Extensive simulations and real-world datasets are used to test the proposed framework’s prediction accuracy,responsiveness,and scalability.Experimental results show that the suggested framework successfully anticipates urban traffic issues in real-time,enables proactive traffic management,and aids in creating smarter,more sustainable cities.

Safety of tranexamic acid in surgically treated isolated spine trauma

BACKGROUND Tranexamic acid(TXA),a synthetic antifibrinolytic drug,effectively reduces blood loss by inhibiting plasmin-induced fibrin breakdown.This is the first study in the United Kingdom to investigate the effectiveness of TXA in the surgical management of isolated spine trauma.AIM To assess the safety of TXA in isolated spine trauma.The primary and secondary outcomes are to assess the rate of thromboembolic events and to evaluate blood loss and the incidence of blood transfusion,respectively.METHODS This prospective observational study included patients aged≥17 years with isolated spine trauma requiring surgical intervention over a 6-month period at two major trauma centers in the United Kingdom.RESULTS We identified 67 patients:26(39%)and 41(61%)received and did not receive TXA,respectively.Both groups were matched in terms of age,gender,American Society of Anesthesiologists grade,and mechanism of injury.A higher proportion of patients who received TXA had a subaxial cervical spine injury classification or thoracolumbar injury classification score>4(74%vs 56%).All patients in the TXA group underwent an open approach with a mean of 5 spinal levels involved and an average operative time of 203 min,compared with 24 patients(58%)in the non TXA group who underwent an open approach with an average of 3 spinal levels involved and a mean operative time of 159 min.Among patients who received TXA,blood loss was<150 and 150–300 mL in 8(31%)and 15(58%)patients,respectively.There were no cases of thromboembolic events in any patient who received TXA.CONCLUSION Our study demonstrated that TXA is safe for isolated spine trauma.It is challenging to determine whether TXA effectively reduces blood loss because most surgeons prefer TXA for open or multilevel cases.Further,larger studies are necessary to explore the rate,dosage,and mode of administration of TXA.

Dental-derived stem cells(DSCs)are attractive cell sources due to their easy access,superior growth capacity and low immunogenicity.They can respond to multiple extracellular matrix signals,which provide biophysical and biochemical cues to regulate the fate of residing cells.However,the direct transplantation of DSCs suffers from poor proliferation and differentiation toward functional cells and low survival rates due to local inflammation.Recently,elegant advances in the design of novel biomaterials have been made to give promise to the use of biomimetic biomaterials to regulate various cell behaviors,including proliferation,differentiation and migration.Biomaterials could be tailored with multiple functionalities,e.g.,stimuli-responsiveness.There is an emerging need to summarize recent advances in engineered biomaterials-mediated delivery and therapy of DSCs and their potential applications.Herein,we outlined the design of biomaterials for supporting DSCs and the host response to the transplantation.

Incense smoke(IS)inhalation exposure system:Physicochemical characterization,IS particle deposition and clearance in human airway using MPPD model

Incense smoke(IS)is source of indoor air pollution and key risk for diverse human diseases.Less in-formation is available regarding controlled IS rodent inhalation exposure system and IS particulate matter(PM)deposition in human airways.Study aimed to demonstrate stable ISPM physicochemical parameters of 10 incense products inside the customized whole body inhalation exposure chamber(without animal)connected to smoke generation unit via aerosol mixing device.IS analyzed for size segregated PM emission,ISPM in vitro aerodynamics(MMAD and GSD determination),fine and ultrafine particle's SEM,SEM-EDX and PAH analysis.Using real life exposure scenario by utilizing MMAD,GSD and PM concentration after Tier 1 exposure assessment as key input parameters,ISPM dosimetry in infant(3 months)and adult(21 years male and female)human airways was calculated using multiple-path particle dosimetry(MPPD 3.04)modeling.Mass median aerodynamic diameter(MMAD)and geo-metric standard deviation(GSD)ranged between 0.55 and 2.10μm and 1.22 to 1.77(polydisperse)respectively.PM1.0 and PM0.1 showed multiple morphology and presence of heavy and trace elements.PAH like acenaphthylene,anthracene,fluorene,naphthalene and phenanthrene were detected(0.84-143.17μg/g).MPPD results showed higher ISPM deposition in pulmonary region and lowest in trachea bronchial region.ISPM deposition in tissue was higher in lower,peripheral lung as compared to upper and central lung tissue.Whole body inhalation exposure system showed stable IS atmosphere(physi-cochemical parameters)indicating the device suitability in future inhalation studies.MPPD ISPM deposition fraction and clearance data showed deep lung penetrating and retention behavior with higher risk in infant followed by female and then male.These modeled particle deposition and clearance data may be useful in risk assessment analysis of IS.

Post processing of additive manufactured Mg alloys:Current status,challenges,and opportunities

Magnesium(Mg)and its alloys are emerging as a structural material for the aerospace,automobile,and electronics industries,driven by the imperative of weight reduction.They are also drawing notable attention in the medical industries owing to their biodegradability and a lower elastic modulus comparable to bone.The ability to manufacture near-net shape products featuring intricate geometries has sparked huge interest in additive manufacturing(AM)of Mg alloys,reflecting a transformation in the manufacturing sectors.However,AM of Mg alloys presents more formidable challenges due to inherent properties,particularly susceptibility to oxidation,gas trapping,high thermal expansion coefficient,and low solidification temperature.This leads to defects such as porosity,lack of fusion,cracking,delamination,residual stresses,and inhomogeneity,ultimately influencing the mechanical,corrosion,and surface properties of AM Mg alloys.To address these issues,post-processing of AM Mg alloys are often needed to make them suitable for application.The present article reviews all post-processing techniques adapted for AM Mg alloys to date,including heat treatment,hot isostatic pressing,friction stir processing,and surface peening.The utilization of these methods within the hybrid AM process,employing interlayer post-processing,is also discussed.Optimal post-processing conditions are reported,and their influence on the microstructure,mechanical,and corrosion properties are detailed.Additionally,future prospects and research directions are proposed.

A Hybrid and Lightweight Device-to-Server Authentication Technique for the Internet of Things

The Internet of Things(IoT)is a smart networking infrastructure of physical devices,i.e.,things,that are embedded with sensors,actuators,software,and other technologies,to connect and share data with the respective server module.Although IoTs are cornerstones in different application domains,the device’s authenticity,i.e.,of server(s)and ordinary devices,is the most crucial issue and must be resolved on a priority basis.Therefore,various field-proven methodologies were presented to streamline the verification process of the communicating devices;however,location-aware authentication has not been reported as per our knowledge,which is a crucial metric,especially in scenarios where devices are mobile.This paper presents a lightweight and location-aware device-to-server authentication technique where the device’s membership with the nearest server is subjected to its location information along with other measures.Initially,Media Access Control(MAC)address and Advance Encryption Scheme(AES)along with a secret shared key,i.e.,λ_(i) of 128 bits,have been utilized by Trusted Authority(TA)to generate MaskIDs,which are used instead of the original ID,for every device,i.e.,server and member,and are shared in the offline phase.Secondly,TA shares a list of authentic devices,i.e.,server S_(j) and members C_(i),with every device in the IoT for the onward verification process,which is required to be executed before the initialization of the actual communication process.Additionally,every device should be located such that it lies within the coverage area of a server,and this location information is used in the authentication process.A thorough analytical analysis was carried out to check the susceptibility of the proposed and existing authentication approaches against well-known intruder attacks,i.e.,man-in-the-middle,masquerading,device,and server impersonations,etc.,especially in the IoT domain.Moreover,proposed authentication and existing state-of-the-art approaches have been simulated in the real environment of IoT to verif
