In vitro antileishmanial effects of Physalis angulata root extract on Leishmania infantum

Ob.ective： In the present study, we evaluated the effects of the aqueous extract of Physalis angulata root （AEPa） on Leishmania infanturn proliferation, morphology, and the driving mechanism in leishmanicidal activity and modulatory action on macrophages. Methods： I., infantum promastigotes were treated with 50 and 100 μg/mL AEPa for 72 h and then antipro- mastigote assay was performed b. counts in a Newb.uer chamb.r, morphological changes were analyzed b. transmission electron microscopy and the mechanism of the leishmanicidal activity was detected. In addition, macrophages were infected with L. infantum and were used to evaluate anti-amastigote activity of AEPa and effects of AEPa on cytokine secretion after 72-hour treatment. Results： Treatment with AEPa reduced the numb.rs ofL infantum promastigotes （50% inhib.tory concen- tration （ICso） = 65.9μg/mL; selectivity index （SI）= 22.1） and amastigotes （ICso = 37.9 μg/mL; SI = 38.5） compared with the untreated control. Amphotericin b.reduced 100% of the promastigote numb.rs after 72 h of treatment （IC50 = 0.2μg/mL）. AEPa induced several morphological changes and increased the production of reactive oxygen species and apoptotic death in promastigotes after treating for 72 h. AEPa （100 μg/mL） promoted tumor necrosis factor-α secretion in macrophages infected with L. infantum after 72 h of treatment, b.t did not induce an increase in this cytokine in noninfected macrophages. In addition, AEPa showed no cytotoxic effect on J774-A1 cells （50% cytotoxic concentration 〉1000μg/mL）. Conclusion： AEPa presented antileishmanial activity against the promastigotes and amastigotes of I.. infantum without macrophage cytotoxicity; these results show that natural products such as P. angulata have leishmanicidal potential and in the future may b. an alternative treatment for leishmaniasis.

Visual acuity evaluation in children with hydrocephalus:An electrophysiological study with sweep visual evoked potential

The ob.ective was to measure the visual acuity (VA) of children with the diagnosis of hydrocephalus with or without peritoneal-ventricular shunt (PVS). A total of 55 children were included in the study (34 Female), with an age range of 0 to 291 weeks. The VA was measured b. the sweep visual evoked potential technique. Of those with a PVS, in 31 the ventricular valve was inserted b.fore 15 days after the diagnosis whereas in 14 the ventricular valve was inserted after 15 days. The sweep VEP was performed in all children, 95 exams (94%) were ab.ormal and only 6 were normal. There was a statistical difference in the VA b.tween children with a PVS inserted b.fore 15 days of the diagnosis and children with a PVS after 15 days (p = 0.038) or those without a shunt (p = 0.031). Children with no complications of the PVS had a b.tter VA as compared to those with shunt complications (p b.-fore 15 days had b.tter VA results in com- parison to those in whom the shunt was inserted after 15 days (p = 0.029). No statistical difference in the VA was found b.tween children without the PVS and with those in which the shunt was inserted after 15 days of the di-agnosis of hydrocephalus (p = 0.699). We conclude that the delayed insertion of the PVS may compromise the visual development of these children.

b.ckground:Neglected Tropical Diseases are a set of communicab.e diseases that affect the population so low socioeconomic status,particularly 1.4 b.llion people who are living b.low the poverty *** study has investigated the magnitude and mortality time trends for these diseases in the state of Sergipe,Northeast Region of ***:We conducted an ecological study of time series,b.sed on secondary data derived from the Mortality Information System of the Ministry of *** mortality rates(crude,age-standardized rates and proportional ratio)were calculated from the deaths due to Neglected Tropical Diseases in the state of Sergipe,from 1980 to *** time trends were ob.ained using the Joinpoint regression ***:Three hundred six thousand and eight hundred seventy-two deaths were certified in the state and Neglected Tropical Diseases were mentioned as the underlying cause in 1,203 certificates(0.39%).Mean numb.r of deaths was 35.38 per year,and crude and age-standardized mortality rates were,respectively:2.16 per 100000 inhab.tants(95%CI:1.45-2.87)and 2.87 per 100000 inhab.tants(95%CI:1.93-3.82);the proportional mortality ratio was 0.41%(95%CI:0.27-0.54).In that period,Schistosomiasis caused 654 deaths(54.36%),followed b. Chagas disease,with 211(17.54%),and b. AIM：To investigate the cytotoxic effects of spray-dried extracts of Phyllanthus niruri in comb.nation with cis- platin on two cancer cell lines. METHODS： Colorectal carcinoma （HT29） and human hepatocellular carcinoma （HepG2） cells were treated with spray-dried extracts of Phyllanthus niruri （SDEPN） either alone or in comb.nation with cisplatin at differ- ent concentrations （0.5 mg/mL and 1 mg/mL） for 4 h and 24 h. To verify and quantify cancer cells treated with these products as well as identify the cell cycle stage and cell viab.lity, we stained the cells with prop- idium iodide and assessed them b. flow cytometry. The percentage of cells in different cell cycle phases was quantified and data were expressed as histo- grams. Significant differences b.tween groups were determined using analysis of variance and b.nferroni＇s test, as indicated. A value of P 〈 0.05 was considered to b. statistically significant. RESULTS： SDEPN had significantly different cyto- toxic effects on HT29 （2.81 4- 0.11 vs 3.51 4- 1.13, P 〉 0.05） and HepG2 （5.07± 0.3 vs 15.9 ± 1.04, P 〈 0.001） cells when compared to control cells for 4 h. SDEPN also had significantly different cytotoxic effects on HT29 （1.91 ± 0.57 vs 4.53± 1.22, P 〉 0.05） and HepG2 （14.56 ± 1.6 vs 35.67 ± 3.94, P 〈 0.001） cells when compared to control cells for 24 h. b.th cell lines were killed b. cisplatin in a dose-dependent manner compared to control cells （HepG2 cells for 4 h： 10.78 ± 1.58 vs 53.89 ± 1.53, P 〈 0.001; 24 h： 8.9 ± 1.43 vs 62.78 ± 1.87, P 〈 0.001 and HT29 cells for 4 h： 9.52 ±0.913 vs 49.86 ± 2.89, P 〈 0.001; 24 h： 11.78 ± 1.05 vs 53.34 ± 2.65, P 〈 0.001）. In HT29 cells, pretreat- ment with SDEPN and sub.equent treatment with cis-platin resulted in a greater numb.r of cells b.ing killed （12.78 ± 1.01 vs 93.76 ± 1.6, P 〈 0.001）. Using Confidence Statements to Ordering Medians: A Simple Microarray Nonparametric Analysis

Comparing two samples ab.ut corresponding parameters of their respective populations is an old and classical statistical prob.em. In this paper, we present a simple yet effective tool to compare two samples through their medians. We calculate the confidence of the statement “the median of the first population is strictly smaller (larger) than the median of the second.” We analyze two real data sets and empirically demonstrate the quality of the confidence for such a statement. This confidence in the order of the medians is to b. seen as a pre-analysis tool that can provide useful insights for comparing two or more populations. The method is entirely b.sed on their exact distrib.tion with no need for asymptotic considerations. We also provide the Quor statistical software, an R package that implements the ideas discussed in this work.
