Human exposure to consumer chemicals has become a worldwide *** this work,a comprehensive sampling strategy is presented,to our knowledge being the first to study all relevant exposure pathways in a single cohort using multiple methods for assessment of exposure from each exposure *** selected groups of chemicals to be studied are consumer chemicals whose production and use are currently in a state of transition and are;per-and polyfluorinated alkyl substances(PFASs),traditional and“emerging”brominated flame retardants(BFRs and EBFRs),organophosphate esters(OPEs)and phthalate esters(PEs).Information about human exposure to these contaminants is needed due to existing data gaps on human exposure intakes from multiple exposure pathways and relationships between internal and external *** environment,food and biological samples were collected from 61 participants and their households in the Oslo area(Norway)on two consecutive days,during winter ***,dust,hand wipes,and duplicate diet(food and drink)samples were collected as indicators of external exposure,and blood,urine,blood spots,hair,nails and saliva as indicators of internal exposure.A food diary,food frequency questionnaire(FFQ)and indoor environment questionnaire were also *** 2000 samples were collected in total and participant views on their experiences of this campaign were collected via *** 91%of our participants were positive about future participation in a similar project,some tasks were viewed as *** the food diary and collection of duplicate food/drink portions were the tasksmost frequent reported as“hard”/“very hard”.Nevertheless,a strong positive correlation between the reported total mass of food/drinks in the food record and the total weight of the food/drinks in the collection bottles was observed,being an indication of accurate performance of the participants despite the challenges of the sampling campaign.

Background: Cardiac toxicity is currently defined as a symptomatic decrease in Left Ventricular Ejection Fraction (LVEF) of more than 5% or an asymptomatic decrease of at least 10% to a value of under 50% in repeated evaluations on conventional transthoracic echocardiogram (TTE), as well as a Global Longitudinal Strain (GLS) value Aims: To highlight using GLS rather than modified Simpson 2d-LVEF for the evaluation of long-term cardiotoxicity. Case Presentation: The case concerns a 73-year-old female patient with a history of breast cancer chemotherapy and anthracyclines-based therapy who presented symptoms of late cardiac toxicity related to the chemotherapeutic treatment. In the following years, the patient remained asymptomatic with a 2d-LVEF of 48%, dilation of the left atrium was found, and the reservoir phase strain was severely decreased. Conclusion: The preferred method for evaluating cardiovascular complications associated with chemotherapy is the TTE, which is performed prior to the start of treatment, during therapy, and in the follow-up. Myocardial deformation as a predictor of cardiotoxicity allows the identification of subclinical heart failure.

The SARS-CoV-2 spike protein contains a functionally important fatty acid(FA)binding site,which is also found in some other coronaviruses,***-CoV and *** occupancy of the FA site by linoleic acid(LA)reduces infectivity by‘locking’the spike in a less infectious ***,we use dynamical-nonequilibrium molecular dynamics(d-NEMd)simulations to compare the allosteric responses of spike variants to LA removal.d-NEMd simulations show that the FA site is coupled to other functional regions of the protein,*** receptor-binding motif(RBM),N-terminal domain(NTd),furin cleavage site,and regions surrounding the fusion peptide.d-NEMd simulations also identify the allosteric networks connecting the FA site to these functional *** comparison between the wild-type spike and four variants(Alpha,delta,delta plus,and Omicron BA.1)shows that the variants differ significantly in their responses to LA *** allosteric connections to the FA site on Alpha are generally similar to those on the wild-type protein,with the exception of the RBM and the S71–R78 region,which show a weaker link to the FA *** contrast,Omicron is the most different variant,exhibiting significant differences in the RBM,NTd,V622–L629,and furin cleavage *** differences in the allosteric modulation may be of functional relevance,potentially affecting transmissibility and *** comparison of the effects of LA on SARS-CoV-2 variants,including emerging variants,is warranted.

AIM: To investigate the role of sorafenib(SFN) in autophagy of hepatocellular carcinoma(HCC). We evaluated how SFN affects autophagy signaling pathway in human HCC cell lines. METHOdS: Two different human HCC cell lines, Hep3 B and Huh7, were subjected to different concentrations of SFN. Cell viability and onset of apoptosis were determined with colorimetric assay and immunoblotting analysis, respectively. The changes in autophagy-related proteins, including LC3, ULK1, AMPK, and LKB, were determined with immunoblotting analysis in the presence or absence of SFN. To assess autophagic dynamics, autophagic flux was measured with chloroquine, a lysosomal inhibitor. The autophagic responsiveness between different HCC cell lines was compared under the autophagy enhancing ***: Hep3 B cells were significantly more resistant to SFN than Huh7 cells. Immunoblotting analysis revealed a marked increase in SFN-mediated autophagy flux in Huh7 cells, which was, however, absent in Hep3 B cells. While both starvation and rapamycin enhanced autophagy in Huh7 cells, only rapamycin increased autophagy in Hep3 B cells. Immunoblotting analysis of autophagy initiation proteins showed that SFN substantially increased phosphorylation of AMPK and consequently autophagy in Huh7, but not in Hep3 B ***: The autophagic responsiveness to SFN is distinct between Hep3 B and Huh7 cells. Resistance of Hep3 B cells to SFN may be associated with altered autophagy signaling pathways.
