c.lc.um signaling is essential for lymphoc.te ac.ivation, with genetic.disruptions of store-operated c.lc.um (c.^(2+)) entry resulting in severe immunodefic.enc.. The inositol 1,4,5-trisphosphate rec.ptor (IP_(3)R), a homo- or heterotetramer of the IP_(3)R1-3 isoforms, amplifies lymphoc.te signaling by releasing c.^(2+) from endoplasmic.retic.lum stores following antigen stimulation. Although knoc.out of all IP_(3)R isoforms in mic. c.uses immunodefic.enc., the seeming redundanc. of the isoforms is thought to explain the absenc. of variants in human immunodefic.enc.. In this study, we identified c.mpound heterozygous variants of ITPR3 (a gene enc.ding IP_(3)R subtype 3) in two unrelated c.uc.sian patients presenting with immunodefic.enc.. To determine whether ITPR3 variants ac. in a nonredundant manner and disrupt human immune responses, we c.arac.erized the c.^(2+) signaling c.pac.ty, the lymphoc.te response, and the c.inic.l phenotype of these patients. We observed disrupted c.^(2+) signaling in patient-derived fibroblasts and immune c.lls, with abnormal proliferation and ac.ivation responses following T-c.ll rec.ptor stimulation. Rec.nstitution of IP_(3)R3 in IP_(3)R knoc.out c.ll lines led to the identific.tion of variants as func.ional hypomorphs that showed reduc.d ability to disc.iminate between homeostatic.and induc.d states, validating a genotype–phenotype link. These results demonstrate a func.ional link between defec.ive endoplasmic.retic.lum c.^(2+) c.annels and immunodefic.enc. and identify IP_(3)Rs as diagnostic.targets for patients with spec.fic.inborn errors of immunity. These results also extend the known c.use of c.^(2+)-assoc.ated immunodefic.enc. from store-operated entry to impaired c.^(2+) mobilization from the endoplasmic.retic.lum, revealing a broad sensitivity of lymphoc.tes to genetic.defec.s in c.^(2+) signaling.

Polyborate distribution in aqueous B（OH）3 solutions at 298.15 K was c.lc.lated using measured pH and literature equilibrium c.nstants. A systematic.study on the struc.ure, stability and Raman spec.ra of the H3BO3 hydrated c.usters, [H3BO3（H2O）n, n=1-14], was probed by DFT in both gaseous and aqueous phase at B3LYP/aug-c.-p VDZ level. All of these stable c.nfigurations were desc.ibed and the most stable hydrated c.usters were c.osen. The mic.o-hydration proc.ss of H3BO3 was desc.ibed in detail: when n≤6, only the equatorial hydration exists in small hydration c.uster;when n≥7, the axial hydration happens;when n≥12, a fully hydration struc.ure forms with 1 axial hydration water and 9 equatorial hydration water. Transition state（TS） of H3BO3 dissoc.ation in aqueous solution has also been loc.ted using transit-guided quasi-newton method, indic.ting the axial hydration is the "powder hose" for the H3BO3 dissoc.ation. The predic.ed equilibrium intra-B-O distanc. varies from 1.38 to 1.43  at the present level of theory. The equatorial hydration distanc. ranges from 0.32 to 0.37 nm and the axial hydration distanc. from 0.16 to 0.18 nm with the saturated hydration number of 10. The c.MD simulation shows that only the first hydration sphere c.n be found, and the hydration distanc. is 3.65 with the hydration number about 11.8. The results reveal a good agreement with the mic.o-hydration from ab initio and the time-spac. averaged struc.ure from X-Ray sc.ttering.

(c.nvergent and disc.iminant) validity with infertility patients,who were administered the Infertility Self-Effic.c. (ISE) sc.le with other measures; and 4 expert ac.eptability,as determined by reproduc.ive health professionals. Setting:Rec.uitment at a fertility c.nter,a national infertility organization,and medic.l trials web pages. Patient(s):A total of 213 partic.pants (159 women; 54 men) with a diagnosis of infertility in the past 2 years were rec.uited for the various study phases. Intervention( s):None. Main Outc.me Measure(s):Partic.pants’infertility self-effic.c. was assessed with an online assessment battery using the ISE sc.le; c.mparison mental health measures assessed c.rrent levels of fertility problem distress,perc.ived stress,and c.ping style.
