Advancing male age differentially alters levels and localization patterns of PLCzeta in sperm and testes from different mouse strains

Sperm-specific phospholipase C zeta(PLCζ)initiates intracellular calcium(Ca2+)transients which drive a series of concurrent events collectively termed oocyte *** investigations have linked abrogation and absence/reduction of PLCζwith forms of male infertility in humans where oocyte activation ***,very few studies have examined potential relationships between PLCζand advancing male age,both of which are increasingly considered to be major effectors of male *** efforts in humans may be hindered by inherent PLCζvariability within the human population,alongside a lack of sufficient controllable ***,utilizing immunoblotting,immunofluorescence,and quantitative reverse transcription PCR(qRT-PCR)we examined for the first time PLCζprotein levels and localization patterns in sperm,and PLCζmRNA levels within testes,from mice at 8 weeks,12 weeks,24 weeks,and 36 weeks of age,from two separate strains of mice,C57BL/6(B6;inbred)and CD1(outbred).Collectively,advancing male age generally diminished levels and variability of PLCζprotein and mRNA in sperm and testes,respectively,when both strains were ***,advancing male age altered the predominant pattern of PLCζlocalization in mouse sperm,with younger mice exhibiting predominantly post-acrosomal,and older mice exhibiting both post-acrosomal and acrosomal populations of PLCζ.However,the specific pattern of such decline in levels of protein and mRNA was ***,our results demonstrate a negative relationship between advancing male age and PLCζlevels and localization patterns,indicating that aging male mice from different strains may serve as useful models to investigate PLCζin cases of male infertility and subfertility in humans.

Argon laser photocoagulation versus intrastromal voriconazole injection in treatment of mycotic keratitis

AIm： To compare argon laser photocoagulation and intrastromal injection of voriconazole as adjunctive treatment modalities in cases of resistant mycotic corneal ulcers. mETHODS： Two groups each of them included 20 cases of resistant mycotic corneal ulcers. Both groups treated with local and systemic specific antimicrobial drugs guided with culture and sensitivity results. In one group argon laser photocoagulation was used as an adjunctive therapy to the specific antifungal drugs and in the other group, intrastromal injection of voriconazole was done besides the specific antifungal drugs. The 40 cases included in the study were proven according to culture and sensitivity to be 28 cases with pure fungal results and 12 cases with mixed （fungal and bacterial）. In argon laser group, argon laser irradiation of the corneal ulcer was performed using argon laser 532 nm wavelength （Carl Zeiss LSL 532s AG; meditec, Inc.） after fluorescein staining. In the other group, voriconazole solution （500 pg/mL） was prepared and injected in the corneal stroma. all cases were followed up for 3mo after healing was achieved. RESULTS： Complete healing of the epithelial defect and resolution of stromal infiltration with no adverse effects were achieved in argon laser group in duration ranged from 2-4wk in 90% of cases. In voriconazole group 4 cases needed amniotic membrane graft due to thinning and 16 cases healed in duration ranged from 2- 6wk （80% of cases）. CONCLUSION： Argon superior to intrastromal treatment of resistant fungal laser photocoagulation is voriconazole injection in corneal ulcers.

The coronavirus disease 2019(COVID-19)represents a global health and economic *** injuries have been approved to be associated with severe acute respiratory syndrome coronavirus(SARS-CoV-2)*** viral tropism pattern of SARS-CoV-2 can induce hepatic injuries either by itself or by worsening the conditions of patients with hepatic ***,other factors have been reported to play a crucial role in the pathological forms of hepatic injuries induced by SARS-CoV-2,including cytokine storm,hypoxia,endothelial cells,and even some treatments for *** the other hand,several groups of people could be at risk of hepatic COVID-19 complications,such as pregnant women and *** present review outlines and discusses the interplay between SARS-CoV-2 infection and hepatic injury,hepatic illness comorbidity,and risk ***,it is focused on the vaccination process and the role of developed vac-cines in preventing hepatic injuries due to SARS-CoV-2 infection.

Influence of climatic conditions,topography and soil attributes on the spatial distribution of site productivity index of the species rich forests of Jalisco,mexico

This paper presents an approach based on field data to model the spatial distribution of the site productivity index （SPI） of the diverse forest types in Jalisco, mexico and the response in SPI to site and cli-matic conditions. A linear regression model was constructed to test the hypothesis that site and climate variables can be used to predict the SPI of the major forest types in Jalisco. SPI varied significantly with topog-raphy （elevation, aspect and slope）, soil attributes （pH, sand and silt）, climate （temperature and precipitation zones） and forest type. The most important variable in the model was forest type, which accounted for 35% of the variability in SPI. Temperature and precipitation accounted for 8 to 9% of the variability in SPI while the soil attributes accounted for less than 4% of the variability observed in SPI. No significant differences were detected between the observed and predicted SPI for the individual forest types. The linear regression model was used to develop maps of the spatial variability in predicted SPI for the individual forest types in the state. The spatial site productivity models developed in this study provides a basis for understanding the complex relationship that exists between forest productivity and site and climatic conditions in the state. Findings of this study will assist resource managers in making cost-effective decisions about the management of individual forest types in the state of Jalisco, mexico.

Synthesis and Antimicrobial Activity of New 2,5-Disubstituted 1,3,4-Oxadiazoles and 1,2,4-Triazoles and Their Sugar Derivatives

A series of new 2,5-disubstituted-l,3,4-oxadiazole and 1,2,4-triazole derivatives were synthesized by hetero- cyclization of acid hydrazide I and thiosemicarbazide derivative 2. Furthermore, the acyclic C-nucleoside analogs were prepared by cyclization of their corresponding sugar hydrazones by reaction with acetic anhydride. The antimicrobial activity of the prepared compounds was evaluated and some of the synthesized compounds revealed good activities against fungi.

Tumor necrosis factor-alpha -308G/A polymorphism and risk of hepatocellular carcinoma in hepatitis C virus-infected patients

Tumor necrosis factor-alpha (TNF-α) is an important cytokine in generating an immune response against infection with hepatitis C virus (HCV). The functions of TNF-α may be altered by single-nucleotide polymorphisms (SNPs) in its gene structure. We hypothesized that SNPs in TNF-α may be important in determining the outcome of an HCV infection. To test this hypothesis, we investigated the role of the polymorphism -308G/A, which is located in the promoter region of the TNF-α gene, in the progression of HCV infection in Egyptian patients using a quantitative real-time polymerase chain reaction (qRT-PCR). The distribution of this polymorphism and its impact on the serum level of TNF-α was compared between 90 HCV-infected patients [45 with HCV-induced cirrhosis and 45 with HCV-related hepatocellular carcinoma (HCC)] and 45 healthy Egyptian volunteers without any history of liver disease. Our results showed that at the TNF-α -308 position, the G/G allele was most common (78.5% ) in the study population, with the G/A and A/A alleles occurring less frequently (13.3% and 8.1% , respectively). Frequencies of G/G, G/A, and A/A genotypes were 87%, 7%, and 6% in patients with liver cirrhosis and were 94% , 4% , and 2% in patients with HCC, respectively. Serum levels of TNF-α were significantly higher in HCV-infected patients than in healthy controls, indicating that the TNF-α -308 polymorphism does not influence the production of TNF-α. The serum level of TNF-α was positively correlated with HCV infection. Taken together, these findings suggest that the TNF-α -308 polymorphism may not be a host genetic factor associated with the severity of HCV infection, but may be an independent risk factor for HCC.

A mODEL OF A NEAR-ZERO ENERGY HOmE(NZEH)USING PASSIVE DESIGN STRATEGIES AND PV TECHNOLOGY IN HOT CLImATES

INTRODUCTION Recent development has seen a drastic increase in energy use trends in Saudi Arabian buildings leading to a demand for an effective course of action for energy conservation and production.A case study-based research initiative explor-ing near-zero energy potential in Saudi Arabia was undertaken.A 4-bedroom detached single-family faculty residence at King Fahd University of Petroleum and minerals(KFUPm)representing common regional housing design trends was utilized.A base case simulation model of the house was developed and val-idated using short-term and real-time energy consumption *** sets of strategies:passive design strategies,representative codes and standards,and renewable technology were employed in the new design of the *** strategies com-prised a green roof,a ventilated wall system,a sloped roof,and insulation for thermal *** alternatives helped reduce the annual energy consumption of the house by 17.2%.The most recent version of the International Energy Conserva-tion Code(IECC 2012)was also incorporated along with ASHRAE Standard 62.2 for *** code and standard together reduced the annual energy consumption by 31.1%.Solar PV was then utilized to reduce grid utilization for the remainder of the house energy *** strategy provided 24.7%of the total energy consumed annually.A combination of strategies showed a 70.7%energy consumption reduction,thereby decreasing the energy index of the house from 162.9 to 47.7 kWh/m^(2)/*** Zero Energy Building(ZEB)concepts and strategies utilized in this study demonstrate a socially responsible approach to achieving near-zero energy performance for an existing house.

Wireless Sensor Network(WSN)forms an essential part of *** is embedded in the target environment to observe the physical parameters based on the type of *** nodes inWSN are constrained by different features such as memory,bandwidth,energy,and its processing *** WSN,data transmission process consumes the maximum amount of energy than sensing and processing of the ***,diverse clustering and data aggregation techniques are designed to achieve excellent energy efficiency in *** this view,the current research article presents a novel Type II Fuzzy Logic-based Cluster Head selection with Low Complexity Data Aggregation(T2FLCH-LCDA)technique for *** presented model involves a two-stage process such as clustering and data ***,three input parameters such as residual energy,distance to Base Station(BS),and node centrality are used in T2FLCH technique for CH selection and cluster ***,the LCDA technique which follows Dictionary Based Encoding(DBE)process is used to perform the data aggregation at ***,the aggregated data is transmitted to the BS where it achieves energy *** experimental validation of the T2FLCH-LCDAtechnique was executed under three different scenarios based on the position of *** experimental results revealed that the T2FLCH-LCDA technique achieved maximum energy efficiency,lifetime,Compression Ratio(CR),and power saving than the compared methods.

Estimating oak forest parameters in the western mountains of Iran using satellite-based vegetation indices

Remote sensing is an important tool for studying and modeling forest *** indices obtained from Landsat-8 remotely sensed data were used to estimate the forest parameters in the western mountains of *** sample points were selected on the map of Bayangan County,Kermanshah province,*** each point,a cluster of five rectangular plots of 8100 m2 and 200 m apart was *** clusters were primarily sampled to determine the variance of the forest *** coefficient of variation was used as a criterion for sample *** density,canopy cover and basal area per hectare were calculated for each *** indices were extracted fromthe Landsat-8 images for each *** linear and nonlinear regressions were conducted to develop the *** models were validated using an independent data *** of model parameters was statistically *** results showed that Normalized difference vegetation index(NDVI)and Transformed normalized difference vegetation index(TNDVI)followed by Simple ratio vegetation index(SRVI)were the best predictors,explaining up to 91%of the variations with high *** NDVI,Soil adjusted vegetation index 2(SAVI2)and SRVI,the cubic model was more appropriate than the linear model for predicting the forest *** this model,the R-square value increased while NRmSE decreased *** Infrared percentage vegetation index(IPVI),the quadratic model was better,but,for other vegetation indices,nonlinear models were not superior to linear ***,it can be concluded that Landsat-8 imagery is suitable for predicting forest parameters in the oak forests of western *** course,large plots must be selected,and pre-classification is necessary to gain accurate and precise estimations.

This paper proposes three fractional discrete chaotic systems based on the Rulkov,Chang,and Zeraoulia–Sprott rational *** dynamics of the proposed maps are investigated by means of phase plots and bifurcations *** stabilization schemes are proposed for each of the three maps and the convergence of the states is established by using the Lyapunov ***,a combination synchronization scheme is proposed whereby a combination of the fractional Rulkov and Chang maps is synchronized to the fractional Zeraoulia-Sprott *** results are used to confirm the findings of the paper.
