Leukocyte-technetium-99m uptake in Crohn's disease:Does it show subclinical disease?

AIM:To evaluate inflammatory activity in patients with Crohn's disease (CD) using technetium-99m-hexamethylpropyleneamine oxime (99mTc-hMpAO) granulocyte ***: Twenty patients (7 male and 13 female) with CD and five healthy volunteers were selected for 99mTc-hMpAO granulocyte scintigraphy. The Crohn's Disease Activity Index (CDAI), blood tests and C-reactive protein (CRp) of each patient were performed 7 d before the scintigraphic images. The leukocytes were labeled according to the International Society of Radiolabeled Blood Elements (ISORBE) consensus protocol and the scintigraphic images, including single photon emission computed tomography, were obtained 30 min and 2 h after injection of the radiolabeled ***:The labeling yield of the leukocytes with the lipophilic complex 99mTc-hMpAO was 55.0%±10%. Six of the 20 patients (30%) presented congruent results for the three parameters investigated (CDAI, Scintigraphic Index and CRp). On the other hand, 14 patients (70%) did not show congruent results. There was no significant correlation between the indices analyzed according to the Spearman test (p>0.05,n=20).CONCLUSION: The results suggest that 99mTc-hMpAO-labeled leukocyte scintigraphy could be important for determining inflammatory activity in CD even in the absence of clinical symptoms.

Aeromonas hydrophila septicemia was a serious infectious disease occurred in *** majority harmed fish was crucian carp（Carassius auratus gibelio）.It was reported that immune prevention was the effect measure to prevent this disease using *** *** it is necessary to develop monoclonal antibody（McAb） against the IgM from Crucian *** this study,the purification of serum immunoglobulin from Crucian carp was carried out by using Goat IgG *** concentration of purified IgM was 2.5 mg·*** using hybridoma-monoclonal antibody technology,hybridoma cell line secreted McAb against the IgM from crucian carp had been established and the characteristics of mab8 had been *** isotyping analysis,the results showed that it belong to IgG1,it was able to recognize the heavy chain of the IgM on western blot,and did not has any cross reaction with the Ig from 19 different fish species including common carp（Cyprinus carpio）,bighead carp（Aristichthys nobilis）,grass carp（Ctenopharhyngodon idella） and bream（Abramis brama） *** results demonstrated that McAb was highly specific and sensitive to the Ig of Crucian carp,and had the great potential to be used for analyzing the structure of crucian carp IgM,monitoring the immunity level of cultured Crucian carp and pathogen diagnosis.

Bioceramic material of the quaternary system;SiO2- CaO-Na2O-p2O5 that has composition similar to Bio- glass? 45S5 was prepared by the sol-gel method from locally obtained bentonite clay (BTC). The monolith obtained was sintered at 1000?C for 2 h to facilitate densification and phase transformation. X-ray diffraction (XRD) analysis revealed the presence of sodium calcium silicate, Na2Ca2Si3O9 as major crystal phase, and another secondary orthorhombic phase, NaCapO4. Fourier transform infrared (FTIR) spectroscopic investigation confirmed the presence of Si-O-Si bonds and a crystalline phosphate in the glass network. Scanning electron microscopy (SEM) revealed a network of micropores and interconnected macropores. Overall, the material displays features amenable for possible utilization in tissue engineering scaffolds.

为了研究污泥龄对活性污泥系统处理微量磺胺类药物(5μg/L)的影响,共运行了4个试验室规模(3L)的序批式活性污泥反应器(SBR),其污泥龄分别为2、8、14、20d。批次摇瓶试验通过设置3个工况(正常运行,加入生物抑制剂,无微生物)来讨论在1个运行周期(8h)中对浓度为5μg/L磺胺甲恶唑的吸附作用、生物降解作用和挥发损失。试验结果显示对磺胺甲恶唑的总去除量为2.14±0.60μg/g SS,吸附作用占总去除量的63%;磺胺嘧啶为1.14±0.63μg/g SS,83%;磺胺间二甲氧为2.33±0.67μg/g SS,35%;磺胺甲基嘧啶为2.45±0.85μg/g SS,55%;磺胺类药物的去除效果与污泥的污泥龄有着非常显著的关系(p<0.02)。通过运行加入磺胺甲恶唑(进水5μg/L)的反应器60d,4个反应器对磺胺甲恶唑的平均去除率分别为10%、41%、51%、58%,处理效果随着污泥龄的增加而变好,同时单位污泥去除率随着污泥龄的增加而降低,SRT=2d的反应器由于存在大量的丝状菌,使得单位污泥对磺胺甲恶唑去除率大大高于其他3个反应器。通过分子生物学分析,发现微生物群落结构的变化不大,从而说明了影响磺胺类药物处理效果的主要因素在于更强的吸附能力,更高的污泥浓度。

Effect of herpesvirus infection on pancreatic duct cell secretion

AIM： To examine the effect of acute infection caused by herpesvirus （pseudorabies virus, pRV） on pancreatic ductal secretion. METhODS： The virulent Ba-DupGreen （BDG） and non- virulent Ka-RREp01acgfp （KEG） genetically modified strains of pRV were used in this study and both of them contain the gene for green fluorescent protein （GFp）. Small intra/ interlobular ducts were infected with BDG virus （10^7 pFU/mL for 6 h） or with KEG virus （10^10 pFU/mL for 6 h）, while non-infected ducts were incubated only with the culture media. The ducts were then cultured for a further 18 h. The rate of hCO3 secretion [base efflux -λ（B）] was determined from the buffering capacity of the cells and the initial rate of intracellular acidification （1） after sudden blockage of basolateral base loaders with dihydro-4,4,- diisothiocyanatostilbene-2,2,-disulfonic acid （500 μmol/L） and amiloride （200 μmol/L）, and （2） after alkali loading the ducts by exposure to Nh4Cl. All the experiments were performed in hCO3-buffered Ringer solution at 37 ℃ （n = 5 ducts for each experimental condition）. Viral structural proteins were visualized by immunohistochemistry. Virallyencoded GFp and immunofluorescence signals were recorded by a confocal laser scanning microscope.RESULTS： The BDG virus infected the majority of accessible cells of the duct as judged by the appearance of GFp and viral anUgens in the ductal cells. KEG virus caused a similarly high efficiency of infection. After blockage of basolateral base loaders, BDG infection significantly elevated -λB-） 24 h after the infection, compared to the non-infected group. however, KEG infection did not modify -λB-）. After alkali loading the ducts, -λB-） was significantly elevated in the BDG group compared to the control group 24 h after the infection. As we found with the inhibitor stop method, no change was observed in the group KEG compared to the non-infected group. Renal Function after Major Uro-Oncologic Surgery and Dexmedetomidine Infusion

Objective: Acute kidney injury in major surgery is associated with increased postoperative mortality. This study aimed to evaluate renal function after major urologic surgery and intraoperative dexmedetomidine infusion. Methods: Thirty oncologic patients with normal renal function scheduled for prostatectomy or nephrectomy, anesthetized with combined epidural and general anesthesia, were randomized to receive either intraoperative blind infusion of dexmedetomidine (Dexmedetomidine Group, n = 15, 0.5 μg/kg load dose plus 0.7 μg/kg/h) or 0.9% saline (Control Group, n = 15) until the end of surgery. Intraoperative and cumulative 24-hour diuresis, serum creatinine (SCr), calculated creatinine clearance (ClCr) and serum cystatin C (SCys) at postoperative days 1, 2 and 3 and 2 weeks after surgery were evaluated. Results: Mean ± standard deviation values for intraoperative diuresis in Dexmedetomidine and Control Groups were 566 ± 396 mL and 298 ± 153 mL, respectively (p = 0.014). Cumulative 24-hour diuresis in Dexmedetomidine and Control Groups was 1947 ± 266 mL and 1748 ± 237 mL, respectively (p = 0.91). Mean values of SCr, ClCr and SCys were not significantly different from their baseline values in both groups and no significant differences were seen between groups at any moment for two weeks (p > 0.05). Conclusion: According to the doses used in this study, despite an intraoperative increase in diuresis, intraoperative infusion of dexmedetomidine did not influence renal performance up to two weeks after major uro-oncologic surgery, as evaluated by SCr, ClCr and SCys.

Babesiosis is a tick-transmitted disease that causes severe economic losses to the cattle industry in Brazil. Water buffaloes (Bubalus bubalis) are often carriers of Babesia spp., but there are no studies that provide an accurate estimation of this infection in animals raised in regions of endemic stability. This study was conducted to investigate Babesia bovis and B. bigemina infections in 108 water buffaloes (50 calves and 58 adult females) located in areas of São paulo state, where the animals were continuously exposed to Rhipicephalus microplus ticks. B. bovis and B. bigemina infections were screened by microscopic examination of blood smears, nested pCR (npCR) and quantitative real-time pCR (qpCR), which were also used to estimate the number of copies (NC) of the cytochrome b (mt-cytB) gene in the blood samples. B. bigemina was found in blood smears of three calves from Alambari herd (all with less than 0.1% parasitemia). Molecular techniques were more sensitive than blood smears to diagnose piroplasms in water buffaloes: 20.37% and 100.00% for B. bovis-infected animals and 59.26% and 100.00% for B. bigemina-infected animals, respectively for npCR and qpCR. The NC of mt-cytB gene of B. bovis and B. bigemina in blood samples revealed significant effects (p p ≤ 0.05) for B. bigemina (2.80 ± 0.06) than for B. bovis (2.61 ± 0.05). Within each herd-age, differences between the species’ NC values were found only in Alambari calves, which showed significantly higher (p ≤ 0.05) NC of B. bigemina (3.48 ± 0.13). The calves and cows from Ibaté showed the lowest NC of B. bigemina (2.29 ± 0.13 and 2.63 ± 0.14) and B. bovis (2.54 ± 0.11 and 2.37 ± 0.12), respectively. These data suggest a high prevalence of B. bovis and B. bigemina infection in the buffalo population in endemic areas of São paulo state.

AIM： To investigate the relationship between mucin 6 （MUC6） VNTR length and hpylori infection. METhODS： Blood samples were collected from patients visiting the Can Tho General hospital for upper gastrointestinal endoscopy. DNA was isolated from whole blood, the repeated section was cut out using a restriction enzyme （pvu 11） and the length of the allele fragments was determined by Southern blotting. hpylori infection was diagnosed by ^14C urea breath test. For analysis, MUC6 allele fragment length was dichotomized as being either long （〉 13.5 kbp） or short （≤ 13.5 kbp） and patients were classified according to genotype [long- long （LL）, long-short （LS）, short-short （SS）]. RESULTS： 160 patients were studied （mean age 43 years, 36% were males, 58% h pylori positive）. MUC6 pvuⅡ-restricted allele fragment lengths ranged from 7 to 19 kbp. Of the patients with the LL, LS, SS MUC6 genotype, 43% （24/56）, 57% （25/58） and 76% （11/46） were infected with hpylori, respectively （p = 0.003）. CONCLUSION： Short MUC6 alleles are associated with h pylori infection.
