Epidemiology and outcomes of acute liver failure in Australia

BACKGROUND Acute liver failure (ALF) is a life-threatening syndrome with varying aetiologies requiring complex care and multidisciplinary management. Its changing incidence, aetiology and outcomes over the last 16 years in the Australian context remain uncertain. AIM To describe the changing incidence, aetiology and outcomes of ALF in South Eastern Australia. METHODS The database of the Victorian Liver Transplant Unit was interrogated to identify all cases of ALF in adults (> 16 years) in adults hospitalised between January 2002 and December 2017. Overall, 169 patients meeting criteria for ALF were identified. Demographics, aetiology of ALF, rates of transplantation and outcomes were collected for all patients. Transplant free survival and overall survival (OS) were assessed based on survival to discharge from hospital. Results were compared to data from a historical cohort from the same unit from 1988- 2001. RESULTS Paracetamol was the most common aetiology of acute liver failure, accounting for 50% of cases, with an increased incidence compared with the historical cohort (P = 0.046). Viral hepatitis and non-paracetamol drug or toxin induced liver injury accounted for 15% and 10% of cases respectively. Transplant free survival (TFS) improved significantly compared to the historical cohort (52% vs 38%, P = 0.032). TFS was highest in paracetamol toxicity with spontaneous recovery in 72% of cases compared to 31% of non-paracetamol ALF (P < 0.001). Fifty-nine patients were waitlisted for emergency liver transplantation. Nine of these died while waiting for an organ to become available. Forty-two patients (25%) underwent emergency liver transplantation with a 1, 3 and 5 year survival of 81%, 78% and 72% respectively. CONCLUSION Paracetamol toxicity is the most common aetiology of ALF in South-Eastern Australia with a rising incidence over 30 years. TFS has improved, however it remains low in non-paracetamol ALF.

Computer symbol calculation of invariants of three-manifolds obtained from trefoil knots

FOR Witten invariants, there have been many studies (see refs. [1—9]). Numerical computer calculation of some Witten invariants τ_r (M) of several three-manifolds was done by Freed andGrompf and Neil. In this note, we calculate _p(M, A) by computer for all manifolds obtained by integral surgeries on a left trefoil knot, up to p≤13. The notations here are standard (see references [3, 6, 9]).

Regional-scale relationship among biological soil crusts,invasive annual grasses,and disturbance

Introduction:Sagebrush ecosystems in western North America are being replaced by the invasion of annual grasses,particularly Bromus *** experimental situations and in localized landscapes,prior studies have documented that biological soil crusts(biocrusts)can reduce annual grass presence and that biocrusts are highly vulnerable to physical *** conservation would benefit from verification of these patterms at scales that matter to local *** study tests if these patterns appear at a regional ***:A previously collected data set of vegetation provided sampling of biocrust cover across the Great Basin within the state of Nevada,USA Data were analyzed with non-par ametric methods including odds ratios and generalized aditive models(GAM).Results:From a data set of 608 vegetation plots within the Great Basin ecoregion,proportion of plots with high annual grass cover differed between sites with high versus low biocrust cover(p=0.0015).A negative relationship between annual grass cover and biocrust cover was confirmed with GAM(p=0.009).For a model of biocrust cover,cattle disturbance was found to be an explanatory variable(p<0.00001).Conclusions:The patterns do appear at the regional scale,with high levels of cattle activity corresponding to low cover of biocrusts,and low cover of biocrusts corresponding to high cover of annual grasses.

Evaluating Subdivisions for Identifying Extraneous Flow in Separate Sanitary Sewer Systems

Separate sanitary sewer systems are designed to convey sewage waste from municipal areas to a central treatment facility;they are not designed to handle water associated with precipitation events. However, intercept of groundwater (infiltration) and of flows through manholes or unauthorized connections (inflows) introduces rainwater into the sanitary sewer system. Infiltration/Inflow (I/I) increases the costs associated with treatment and can create additional environmental problems. Identifying and quantifying the volume I/I can be complicated and costly. A simple quantitative method was developed to quantify the extent of I/I occurring in sewer sheds. The method uses measured sewer flows, water usage, precipitation values, and land cover data to calculate the volume of extraneous flows. To assess its utility, the method was used to compare two urban sewer sheds, Holiday Knolls and Eagle View. Both sewer sheds showed evidence of I/I in excess of 200 gallons per day per inch-mile of sewer pipe (gpd/in-mile). Holiday Knolls, the older subdivision had an average I/I of 1912 gpd/in-mile, while Eagle View had an average of 1143 gpd/in-mile. The devel- oped method provided simple means to calculate I/I and to identify sewer sheds in need of repair.

An integrated analysis of air pollution from US coal-fired power plants

TIR-Learner, a New Ensemble Method for TIR Transposable Element Annotation, Provides Evidenee for Abundant New Transposable Elements in the Maize Genome

Transposable elements (TEs) make up a large and rapidly evolving proportion of plant genomes. Among Class II DNA TEs, TIR elements are flanked by characteristic terminal inverted repeat sequences (TIRs). TIR TEs may play important roles in genome evolution, including generating allelic diversity, inducing structural variation, and regulating gene expression. However, TIR TE identification and annotation has been hampered by the lack of effective tools, resulting in erroneous TE annotations and a significant underestimation of the proportion of TIR elements in the maize genome. This problem has largely limited our understanding of the impact of TIR elements on plant genome structure and evolution. In this paper, we propose a new method of TIR element detection and annotation. This new pipeline combines the advantages of current homology-based annotation methods with powerful de novo machine-learning approaches, resulting in greatly increased efficiency and accuracy of TIR element annotation. The results show that the copy number and genome proportion of TIR elements in maize is much larger than that of current annotations. In addition, the distribution of some TIR superfamily elements is reduced in centromeric and pericentromeric positions, while others do not show a similar bias. Finally, the incorporation of machine-learning techniques has enabled the identification of large numbers of new DTA (hAT) family elements, which have all the hallmarks of bona fide TEs yet which lack high homology with currently known DTA elements. Together, these results provide new tools forTE research and new insight into the impact ofTIR elements on maize genome diversity .

Mining diversity of the natural biorefinery housed within Tipula abdominalis larvae for use in an industrial biorefinery for production of lignocellulosic ethanol

Although they are the largest taxonomic group of animals, relatively few insects have been examined for symbiotic relationships with micro-organisms. However, this is rapidly changing because of the potential for examination of the natural insectmicrobe-lignocellulose interactions to provide insights for biofuel technology. Micro-organisms associated with lignocellulose-consuming insects often facilitate the digestion of the recalcitrant plant diet; therefore these microbial communities may be mined for novel lignocellulose-degrading microbes, or for robust and inexpensive biocatalysts necessary for economically feasible biofuel production from lignocellulose. These insect-microbe interactions are influenced by the ecosystem and specific lignocellulose diet, and appre- ciating the whole ecosystem-insect-microbiota-lignocellulose as a natural biorefinery provides a rich and diverse framework from which to design novel industrial processes. One such natural biorefinery, the Tipula abdominalis larvae in riparian ecosystems, is reviewed herein with applications for biochemical processes and overcoming challenges involved in conversion of lignocellulosic biomass to fuel ethanol. From the dense and diverse T. abdominalis larval hindgut microbial community, a cellulolytic bacterial isolate, 27C64, demonstrated enzymatic activity toward many model plant polymers and also produced a bacterial antibiotic. 27C64 was co-cultured with yeast in fermentation of pine to ethanol, which allowed for a 20% reduction of commercial enzyme. In this study, a micro-organism from a lignocellulose-consuming insect was successfully applied for improvement of biomass-to-biofuel technology.

Analysis of cellulolytic and hemicellulolytic enzyme activity within the Tipula abdominalis （Diptera： Tipulidae） larval gut and characterization of Crocebacterium ilecola gen. nov., sp. nov., isolated from the Tipula abdominalis larval hindgut

In forested stream ecosystems of the north and eastern United States, larvae of the aquatic crane fly Tipula abdominalis are important shredders of leaf litter detritus. T. abdominalis larvae harbor a dense and diverse microbial community in their hindgut that may aide in the degradation oflignocellulose. In this study, the activities ofcellulolytic and hemicellulolytic enzymes were demonstrated from hindgut extracts and from bacterial isolates using model sugar substrates. One of the bacterial isolates was further characterized as a member of the family Microbacteriaceae. Taxonomic position of the isolate within this family was determined by a polyphasic approach, as is commonly employed for the separation of genera within the family Microbacteriaceae. The bacterial isolate is Gramtype positive, motile, non-sporulating, and rod-shaped. The G ＋ C content of the DNA is 64.9 mol%. The cell wall contains B2y type peptidoglycan, D- and L-diaminobutyric acid as the diamino acid, and rhamnose as the predominant sugar. The predominant fatty acids are 12-methyltetradecanoic acid （ai-C15：0） and 14-methylhexadecanoic acid （ai-C17：0）. The isolate forms a distinct lineage within the family Microbacteriaceae, as determined by 16S rRNA sequence analysis. We propose the name Crocebacterium ilecola gen. nov., sp. nov., to accommodate this bacterial isolate. The type species is T202T （ATCC BAA-1359; GenBank Accession DQ826511）.
