Antiplasmodial mechanism of Lawsonia inermis: An in silico based investigation

Background:Lawsonia inermis has been widely reported to be used as an herbal treatment for Malaria.However,despite several experimental studies about its antimalarial activities,the approach through which the herbal plant suppresses plasmodium infection is yet to be found.Consequently,this study uses computational approaches to understand the biological targets and pathways involved in the antiplasmodial activities of Lawsonia inermis compounds.Methods:The Gas Chromatography-Mass Spectrometry technique identified the phytocompounds present in the herbal plant.GeneCards,OMIM,and NCBI databases were explored to collate target proteins for further network pharmacology analysis.The phytocompounds were subjected to Absorption,Distribution,Metabolism,Excretion and Toxicity(ADMET)and druglikeness analysis.The STRING algorithm and Cytoscape were employed to develop and analyze the relationships among target proteins and compounds/targets/pathways network of the putative targets of the phytocompounds.Further computational analysis was carried out to identify potential drug leads.Results:Based on the Network Pharmacology studies,phytocompounds in Lawsonia inermis exhibit antiplasmodial activity by interacting with therapeutic genes that play essential roles in metabolism and signaling pathways.Notable among the genes are MMP9,MAPK1,HMOX1 and IDO1.Meanwhile,the most influenced pathways include the metabolic pathway,PI3K-Akt signaling pathway,and HIF-1 signaling pathway.ADMET analysis,molecular docking analysis,and molecular dynamics simulation revealed that 3-phenyl-2-Isoxazoline and 2-Dimethylamino-3’-methoxyacetophenone are recommendable drug leads for Malaria treatment as they form stable and favorable complexes with Matrix metalloproteinase-9(MMP9)target.Conclusion:The 3-phenyl-2-Isoxazoline and 2-Dimethylamino-3’-methoxyacetophenone phytocompounds from Lawsonia inermis herbal plant are predicted as antimalarial drug candidates and recommended for further wet-lab studies.

Oral and peri-oral signs and symptoms of herbal dentifrices in patients in two oral medicine clinics in Lagos—A preliminary study

Background: Herbal dentifrices appear to have become an attractive alternative for some consumers and its use has gained appreciable acceptance in Nigeria. This could be partly due to the perception that herbal toothpastes like other herbal products are “natural”, devoid of chemicals and therefore superior to regular toothpastes. Aggressive advertisement of the products may also contribute to this new trend. Mucosal symptoms such as glossitis and intolerance to spicy foods seen in patients attending the oral diagnosis clinic are usually associated with anaemia and nutritional deficiency states. The disturbing trend of an increasing number of patients presenting with such oral mucosal symptoms associated with the use of herbal dentifrices prompted this study. This trend was observed in two isolated oral medicine clinics. Objective: To sensitize oral health, allied professionals and consumers on the possible adverse effects of regular use of some herbal dentifrices sold in Nigeria. Materials and Methods: Routine patients of two oral medicine clinics in Lagos, namely the Randle General Hospital and the Lagos University Teaching Hospital who complained of some adverse mucosal signs and symptoms following the regular use of two identified herbal toothpastes made up the study population. The study was conducted between April 2010-April 2011. A detailed history and examination was carried out on these patients and clinical photographs of oral signs observed was taken in consenting patients. Data analysis was done using the Epi-info 6 software. P values ≤ 0.05 was considered statistically significant and data was presented in table format. Results: A total of 45 patients, 9 (20%) males and 36 (80%) fe-males were seen. The age range of patients was 14 - 78 years;mean age 45.5 ± 14.9 years. The duration of use of herbal dentifrices ranged from 2 weeks - 84 months (mean 24 ± 11.5 months). The COVID-19 Pandemic: Present or Gone, Where Do Adolescents Stand?

Introduction: The initial global response to the SARS-CoV-2 pandemic focused on adults as adolescents mainly had subclinical infections. Adolescents, however, could transmit the virus and hence be a risk to the elderly, young children and immunocompromised. Adolescents are also at risk of long-term complications. This study sought to determine the prevalence of antibodies to SARS-CoV-2 among adolescents in Southern Nigeria, and to determine the association between SARS-CoV-2 seropositivity and age, gender, socioeconomic class, school type and nutritional status. Methods: This descriptive cross-sectional study was carried out in Edo state of southern Nigeria. We recruited 270 in-school adolescents aged 10 - 19 years. A WHO-adapted, pre-tested questionnaire was used to obtain data on participant history and risk factors. The questionnaire was interviewer-administered. Adolescents whose parents gave consent were recruited through a multi-stage randomized sampling method. Capillary blood for the assay was collected and analysed using a lateral-flow immunoassay device. The primary outcome was serostatus of the study. The positive predictors of serostatus were finally determined using a multiple logistics regression model. Statistical significance was set at p Results: The seroprevalence of SARS-CoV-2 was 45.9%. IgM antibodies were found in 5.6% of participants. Older adolescents had higher seropositivity rates than younger adolescents (2 = 8.101, p = 0.017). There was no association between gender and seropositivity. Adolescents in public schools were more likely to be seropositive than those in private schools (2 = 25.242, p = 0.001). A higher seroprevalence was noticed among study participants in the lower socioeconomic class (x2 = 6.928, p = 0.031), and those in overcrowded classes. (2 = 79.303, p = 0.001). An Assessment of Saltwater Intrusion in Coastal Regions of Lagos, Nigeria

This paper explains various factors that contribute to saltwater intrusion, including overexploitation of freshwater resources and climate change as well as the different techniques essential for effective saltwater intrusion management. The impact of saltwater intrusion along coastal regions and its impact on the environment, hydrogeology and groundwater contamination. It suggests potential solutions to mitigate the impact of saltwater intrusion, including effective water management and techniques for managing SWI. The application of A.I (assessment index) serves as a guideline to correctly identify wells with SWI ranging from no intrusion, slight intrusion and strong intrusion. The challenges of saltwater intrusion in Lagos and the salinization of wells were investigated using the hydro-chemical parameters. The study identifies four wells (“AA”, “CMS”, “OBA” and “VIL”) as having high electric conductivities, indicating saline water intrusion, while other wells (“EBM”, “IKJ, and “IKO”) with lower electric conductivities, indicate little or no salt-water intrusion, and “AJ” well shows slight intrusion. The elevation of the wells also played a vital role in the SWI across coastal regions of Lagos. The study recommends continuous monitoring of coastal wells to help sustain and reduce saline intrusion. The findings of the study are important for policymakers, researchers, and practitioners who are interested in addressing the challenges of saltwater intrusion along coastal regions. We assessed the SWI across the eight (8) wells using the Assessment Index to identify wells with SWI. Wells in “CMS” and “VIL” has strong intrusions. A proposed classification system based on specific ion ratios categorizes water quality from good (+) to highly (-) contaminated (refer to Table 4). These findings underscore the need for attention and effective management strategies to address groundwater unsuitability for various purposes.

Ultimate bearing capacity(UBC)is a key subject in geotechnical/foundation engineering as it determines the limit of loads imposed on the foundation.The most reliable means of determining UBC is through experiment,but it is costly and time-consuming which has led to the development of various models based on the simplified assumptions.The outcomes of the models are usually validated with the experimental results,but a large gap usually exists between them.Therefore,a model that can give a close prediction of the experimental results is imperative.This study proposes a grasshopper optimization algorithm(GOA)and salp swarm algorithm(SSA)to optimize artificial neural networks(ANNs)using the existing UBC experimental database.The performances of the proposed models are evaluated using various statistical indices.The obtained results are compared with the existing models.The proposed models outperformed the existing models.The proposed hybrid GOA-ANN and SSA-ANN models are then transformed into mathematical forms that can be incorporated into geotechnical/foundation engineering design codes for accurate UBC measurements.

Globally,type 2 diabetes mellitus(T2DM)is considered as one of the most common diseases,which is associated with irreversible risk factors such as age,gene,race,ethnicity,and reversible factors such as diet,physical activity,and smoking.Noncompliance to dietary regimen is the major factors for rapidly rising incidence of T2DM among developing countries.Recently,elevated HbA1c level has also been considered as one of the leading risk factors for developing microvascular and macrovascular complications.Improvement in the elevated HbA1c level can be achieved through diet management;thus,this study found that improvement in patients’dietary knowledge,attitude toward,and compliance to diet regimen will promote a better control of the disease.In conclusion,the stakeholders(health-care providers,health facilities,agencies involved in T2DM care)should educate the patients on the importance of dietary modification in the disease management.Similarly,there is a need for nurses and other health care professionals to advocate and promote a better holistic management that will encourage patients participation and family involvement in the care.

Effect of Intake Conditions and Nozzle Geometry on Spray Characteristics of Group-Hole Nozzle

The group-hole nozzle concept is proposed to meet the requirement of nozzle hole minimization and reduce the negative impact of poor spatial spray distributions.However,there are limited researches on the effects of intake conditions and nozzle geometry on spray characteristics of the group-hole nozzle.Therefore,in this study,an accurate spray model coupled with the internal cavitating flow was established and computational fluid dynamics(CFD)simulations were done to study the effects of intake conditions and nozzle geometry on spray characteristics of the group-hole nozzle.Experimental data obtained using high-speed digital camera on the high-pressure common rail injection system was used to validate the numerical model.Effects of intake conditions(injection pressure and temperature)and nozzle geometry(orifice entrance curvature radius and nozzle length)on the flow and spray characteristics of the group-hole nozzle were studied numerically.The differences in Sauter mean diameter(SMD),penetration length and fuel evaporation mass between single-hole nozzle and group-hole nozzle under different nozzle geometry were also discussed.It was found that the atomization performance of the group-hole nozzle was better than that of the single-hole nozzle under same intake conditions,and the atomization effect of the short nozzle was better than that of the long nozzle.With increase in the orifice entrance curvature radius,the average velocity and turbulent kinetic energy of the fuel increased,which was conducive to improving the injection rate and flow coefficient of the nozzle.Meanwhile,the penetration length and SMD value rose,while evaporation mass dropped.When the ratio of the orifice entrance curvature radius(R)to the diameter of injection hole(D)was 0.12,the spray characteristics reached a constant state due to elimination of cavitation.Conclusions were made based on these.This study is expected to be a guide for the design of the group-hole nozzle.

Treatment satisfaction and medication adherence among hypertensive patients in a teaching hospital in Ekiti State, Nigeria

Objective:This study investigates treatment satisfaction and medication adherence among hypertensive patients attending the medical outpatient clinic of a teaching hospital in Ekiti State,Nigeria.Materials and Methods:The study design was descriptive and cross-sectional using quantitative research strategy.A systematic sampling technique was used in selecting 270 participants;a structured questionnaire was used for data collection.Medication adherence was assessed using the 8-item Morisky Medication Adherence Scale,and treatment satisfaction was assessed using the 14-item Treatment Satisfaction Questionnaire for Medication.Results:The study findings revealed that majority(73.7%)of the study participants had good knowledge of hypertension,while 44.4%had a good level of treatment satisfaction.Test of association of participants’sociodemographic characteristics revealed age(χ^(2)=23.773,P=0.003)and occupation(χ^(2)=21.251,P=0.019)to be significantly associated with treatment satisfaction.The level of medication adherence by the participants was observed to be relatively poor(50.4%).The educational status(χ^(2)=23.914,P=0.001)and monthly income(χ^(2)=10.406,P=0.034)of the participants showed significant association with medication adherence.Conclusion:The study recommends that improving patients’understanding of their disease and treatment with subsidizing hypertensive medications by the government will enhance medication adherence.

Influence of mulch and poultry manure application on soil temperature,evapotranspiration and water use efficiency of dry season cultivated okra

Sustainable vegetable production especially during the dry season requires adequate conservation of soil water.This study was conducted to evaluate the sole and interactive effects of mulching(M)and poultry manure(PM)application on soil temperature(ST),crop evapotranspiration(ETc)and water use efficiency(WUE)of okra.The experiment was a Randomized Complete Block Design(RCBD)with three replicates.The treatments were M at 0 and 6t ha^(-1)and PM at 0,10 and 20t ha^(-1).Soil temperature was measured using digital thermometer while ETc was determined by water depletion method using a Time Domain Reflectometer.Irrigation at field capacity was applied manually at 2-day intervals.Independent appli-cation of mulch significantly lowered ST while joint application of 20t ha^(-1)PM(PM20)and M signifi-cantly(p≤0.05)reduced ST at 5 cm and 10 cm soil depth compared with the unmulched plots in both seasons.Application of 10t ha^(-1)PM(PM10)without M recorded the highest ETc(43.7 mm),while joint application of PM20 and M reduced ETc by about 93%compared with PM10 only.Okra used water most efficiently when PM20 was applied under mulched plot.There was 62.2%increase in WUE under mulched plots compared with the control while the residual effect of PM10 and M significantly increased WUE by 65.5%.It was evident that M alongside application of PM is a good strategy for regulating ST,moderating ETc and increasing okra WUE,especially during dry season farming.

Postoperative pain assessment and management among nurses in selected hospitals in Benin City,Edo State,Nigeria

Objective:This study was designed to determine the nurse assessment of postoperative pain and its management in selected hospitals,Benin City,Edo State,Nigeria.Materials and Methods:A descriptive cross-sectional survey was adopted.The target population consist of 222 purposely nurses who are in the cadre of nursing officer II to chief nursing officer who works in the various surgical wards/units of the selected health facilities.The data were collected from the participants using the pretested structured questionnaire developed by the researcher.Results:Results showed that 66.2%of nurses had a poor level of knowledge on postoperative pain assessment.The McGill Pain Questionnaire was the most used pain assessment tool with a mean score of 2.84 whereas the Dallas Pain Questionnaire was the least used with a mean score of 1.90.“Providing clean,calm,and well-ventilated ward environment”(3.69±0.61)was the most used nonpharmacological method for postoperative pain management,followed by“distraction,relaxation,and guided imagery”(3.52±0.50),“dressing,bandage,splint,and reinforce wound sites postoperatively”(3.39±0.54),and“early ambulation/exercise”(3.20±0.62).The most used pharmacological interventions were“acetaminophen”(3.63±0.55),“topical anesthetic”(2.92±0.62),“nonselective nonsteroidal anti-inflammatory drugs”(2.87±0.43),and“mixed opioid agonist-antagonist”(2.56±0.56).Conclusion:There is a poor level of knowledge on postoperative pain assessment among nurses in this study setting.It is,therefore,pertinent for hospitals to organize continuous in-service training for postoperative pain assessment and management,especially on nonpharmacological approaches among nurses.
