Hepatoprotective and in vivo antioxidant activities of the hydroethanolic leaf extract of Mucuna pruriens (Fabaceae) in antitubercular drugs and alcohol models

AIM: Hepatotoxicity is a significantly increasing health problem worldwide, and the extent of the problem has stimulated interest in the search for hepatotherapeutic agents from plants. This study investigated the hepatoprotective and in vivo antioxidant activities of the hydroethanolic extract of Mucuna pruriens leaves in antitubercular and alcohol-induced hepatotoxicity assays in rats. METHOD: In each of the models used, seven groups were allotted. The different groups received normal saline(10 mL·kg-1, p.o.); hepatotoxicant(isoniazid-rifampicin, INH-RIF, 100 mg·kg-1, i.p. or 20% ethanol 5 g·kg-1, p.o.) and normal saline(10 mL·kg-1, p.o.); hepatotoxicant and extract at doses of 100, 200, and 400 mg·kg-1 p.o.; hepatotoxicant and silymarin 50 mg·kg-1 p.o.; and extract at 400 mg·kg-1 p.o.. On the 21st day of treatment, blood was collected for assessment of serum biochemical parameters and harvested liver samples were assessed for antioxidants. RESULTS: The hepatotoxicants significantly(P < 0.05-0.001) increased the levels of alanine transaminase(ALT), aspartate transaminase(AST), alkaline phosphatase(ALP), bilirubin, and malondialdehyde(MDA); and reduced the levels of catalase(CAT), superoxide dismutase(SOD), glutathione peroxidase(GPx), and reduced glutathione GSH compared to control. M. pruriens significantly reversed(P < 0.05-0.001) the elevation in the level of ALT, AST, ALP, and bilirubin caused by the hepatotoxicants. The extract(200 and 400 mg·kg-1) significantly reversed(P < 0.05) the diminution in the level of in vivo antioxidants and increased the level of MDA produced by INH-RIF. M. pruriens(100-400 mg·kg-1) elicited significant reduction(P < 0.001) in the level of MDA compared to the alcohol group. Silymarin also reversed the deleterious effects of the hepatotoxicants. CONCLUSION: The hydroethanolic extract of Mucuna pruriens leaves possesses hepatoprotective activity with enhancement of in vivo antioxidants as a possible mechanism of action.

AIM: Diabetes Mellitus is associated with significant morbidity and mortality worldwide. The need for enhanced efficacy and safety, and cheaper and more readily available new drugs has increased the search for new antidiabetic drugs from plants. This study was conducted to investigate the antidiabetic activity of the hydroethanolic leaf extract of B. coccineus in rats. METHODS: The effect of B. coccineus extract(100–800 mg·kg–1, p.o.) on blood glucose levels in normal and glucose loaded rats, and alloxan-induced diabetic rats was determined. After 10 days of treatment, blood samples were collected from rats for lipid and insulin profiling. Animals were thereafter sacrificed and the kidneys, heart, and liver were harvested for antioxidant indices assay. RESULTS: In normal rats, B. coccineus did not cause significant reduction in blood glucose. At the dose of 800 mg·kg–1, significant increase in blood glucose level was not observed 30 min. after glucose load. B. coccineus administered acutely did not generally produce significant reduction in blood glucose level in diabetic rats. Administered subacutely, the extract significantly reduced blood glucose level in diabetic rats from the 3rdday with peak effect observed at the dose of 800 mg·kg–1on the 10thday. The extract generally preserved in vivo antioxidant levels in the kidneys, heart, and liver, increased the level of high density lipoprotein and insulin, and reduced the level of triglycerides and low density lipoprotein compared to diabetic control. CONCLUSION: The findings in this study suggest that the hydroethanolic leaf extract Byrsocarpus coccineus possesses antidiabetic activity possibly mediated through inhibition of intestinal glucose absorption, in vivo antioxidant activity, and enhancement of regeneration of beta cells of the pancreas and insulin secretion.

Dynamic and ballistic impact behavior of biocomposite armors made of HDPE reinforced with chonta palm wood(Bactris gasipaes) microparticles

The mechanical behavior of chonta palm wood(Bactris gasipaes) microparticles reinforced high density polyethylene(HDPE) under high strain-rate compressive and ballistic impact loading were *** palm wood microparticles were introduced into the HDPE via an extrusion process using parallel twin screw extruder to produce biocomposite containing 10, 20, 25, and 30 wt % chonta wood microparticles. In addition to mechanical tests, fractographic analysis was done to understand the failure mechanism in the biocomposites under dynamic and ballistic impact loads. The results indicate that both quasi-static and dynamic mechanical properties of HDPE are enhanced by reinforcement with chonta palm wood particles. The biocomposites containing 25 wt % wood microparticles exhibited the highest strength, stiffness, ballistic impact resistance and impact energy absorption capability. Introduction of microparticles of chonta palm wood as reinforcement into a polymeric matrix such as HDPE is therefore a promising method to develop biocomposites with enhanced capacity to withstand dynamic impact loading and absorb impact energy.

本研究采用7对引物对新疆26个天山云杉群体中的945个样本进行了EST-SSR标记分析,揭示其遗传多样性。研究发现,新源和阜康等17个地区群体的基因多样性指数介于0.374~0.520之间;在新源和阜康的种群中发现的位点等位基因数表明海拔和等位基因多样性存在一定正相关关系。根据Hardy-weinberg平衡定律,基于EST-SSR标记的天山云杉天然群体的稀有等位基因频率没有出现偏离(Fis=-0.018),其中92.84%的遗传变异出现于群体内部。通过与其他云杉物种对比分析,表明天山云杉具有中度的遗传多样性水平;在新源、阜康地区发现了7个独有的等位基因。通过天山云杉群体的遗传多样性分析,为天山云杉种质资源的保护和遗传改良提供了实践参考。

Cardioprotective and renoprotective effects of Cocos nucifera water in offspring of high fat diet fed Wistar rat dams

Nigeria is an Afrotropical region with considerable ecological heterogeneity and levels of biotic *** its vertebrate fauna,reptiles have broad distributions,thus,they constitute a compelling system for assessing the impact of ecological variation and geographic isolation on species *** red-headed rock agama,Agama agama,lives in a wide range of habitats and,thus,it may show genetic structuring and ***,we tested the hypothesis that ecology affects its genetic structure and population *** inference phylogenetic analysis of a mitochondrial DNA(mtDNA)gene recovered four well-supported matrilines with strong evidence of genetic structuring consistent with eco-geographic *** differences among populations based on the mtDNA also correlated with geographic distance. The ecologicalniche model for the matrilines had a good fit and robust performance. Population divergence alongthe environmental axes was associated with climatic conditions, and temperature ranked highestamong all environmental variables for forest specialists, while precipitation ranked highest for theforest/derived savanna, and savanna specialists. Our results cannot reject the hypothesis that nicheconservatism promotes geographic isolation of the western populations of Nigerian A. ***, ecological gradients and geographic isolation impact the genetic structure and population divergenceof the lizards. This species might be facing threats due to recent habitat fragmentation,especially in western Nigeria. Conservation actions appear necessary.
