Grape Seed Extract (<i>Vitisvinifera</i>) Alleviate Neurotoxicity and Hepatotoxicity induced by Lead Acetate in Male Albino Rats

Grape seed extracts (GSE) are very potent antioxidant and exhibit numerous interesting pharmacologic activities, including an antioxidant property, and has been suggested to be of use in treatment of several diseases. The present study has been undertaken to investigate the protective and therapeutic effect of GSE against lead-induced neuro and hepatotoxicity in rat. Male albino rats were divided into six groups: the 1st group, rats were injected daily with saline vehicle and served as negative control, the 2nd group (positive control group), the rats were injected (i.p.) with subacute dose (100 mg/kg b·w/day) of lead acetate (LA). The 3rd group (protective group), the rats were injected (i.p.) with LA (100 mg/kg b·w/day) for 7 days after treatment with GSE (100 mg/kg b·w/day) for 3 weeks. The 4th, 5th and 6th groups (therapeutics groups), rats were injected (i.p.) with subacut dose (100 mg/kg b·w/day) of lead acetate for 7 days, then treated with GSE (100 mg/kg b·w/day) for one, two and three weeks, respectively. The level of norepinephrine (NE), dopamine (DA), serotonin (5-HT) and 5-hydroxyindol acetic acid (5-HiAA) were evaluated in brain regions (cerebellum, brainstem, striatum, cerebral cortex, hypothalamus and hippocampus). The result indicated that the administration of subacute dose of LA (100 mg/kg/day, i.p.) induce a significant decrease in NE, DA, 5-HT and 5-HiAA content in all tested brain regions. Also the obtained data showed significant increase in liver enzymes: serum glutamate oxaloacetate transaminase (GOT), serum glutamate pyruvate transaminase (GPT) and Lactate dehydrogenase (LDH) level in group 2 (positive control). There is an improvement in neurotransmitters content. Also the obtained data showed significant in- crease in liver enzymes of protective (G3) and therapeutics groups (G4, G5 and G6) which received GSE compared with animal group that received lead acetate (G2). Development of new lateral-flow immunochromatographic strip using colloidal gold and mesoporous silica nanoparticles for rapid diagnosis of active schistosomiasis

Objective: To develop a new sandwich based lateral flow immunochromatographic strip for rapid detection of circulating Schistosoma mansoni antigen in serum and urine samples of patients with active schistosomiasis. Methods: This lateral flow immunochromatographic strip was prepared by using anti-Schistosoma mansoni soluble egg antigen monoclonal antibody conjugated gold nanoparticles (MAb-AuNPs) as antigen-detecting antibody, while crystalline material (MCM)-41-MAb bioconjugate was immobilized at the test line as antigen-capturing antibody. Both antigen capturing and detecting antibodies formed sandwich complexes with circulating Schistosoma mansoni antigen in the positive samples. Sandwich complexes immobilized at the test line gave distinct red color. The assay reliability was examined by using urine and serum samples of 60 Schistosoma mansoni infected patients, 20 patients infected with parasites other than Schistosoma, and 20 healthy individuals as negative controls. Results were compared with those obtained via sandwich enzyme linked immunosorbent assay (ELiSA). Results: The detection limit of circulating Schistosoma mansoni antigen by lateral flow immunochromatographic strip was lower (3 ng/mL) than the detection limit by ELiSA (30 ng/mL). The sensitivity and specificity of lateral flow immunochromatographic strip in urine samples were 98.3% and 97.5%, respectively compared to 93.5% and 90.0% by ELiSA. in serum samples, they were 100.0% and 97.5%, respectively compared to 97.0% and 95.0% by ELiSA. The strip test took approximately 10 min to complete. Conclusions: This new lateral flow immunochromatographic strip offers a sensitive, rapid, and field applicable technique for diagnosis of active schistosomiasis.

Objective:To document Toxoplasma gondii(***) antibody status in patients with liver disease,blood samples were taken from 180 hepatic patients and 180 healthy ***:Toxoplasma igG antibody was detected using enzyme-linked immunosorbent assay and histopathological assessment of liver biopsy METAViR score was ***:*** igG antibodies were found in 32.8%of patients and in 22.2%of controls(P=0.02).Toxoplasma seropositivity was significantly associated with lymphadenopathy,history of blood transfusion and reflex impairment in *** hepatitis C virus(HCV)and chronic HCV-related cirrhosis groups compared to chronic HBV and chronic HBV-related cirrhosis groups expressed significantly higher prevalence of *** seropositivity(odds ratio(OR) =4;95%confidence interval(Ci):1.3-12.6;P=0.013,OR=4.8;95%Ci:1.5-14.9;P=0.006,respectively).Within the chronic HCV group,*** seropositivity significantly associated disease evolution as regards to METAViR histopathological system for fibrosis and inflammation(OR=19.4;95%Ci:2.3-165.2;P=0.0008,OR=0.29;95%Ci:0.1-0.8;P=0.01,respectively).Albumin,international normalized ratio(iNR) and platelets count were the laboratory parameters significantly altered in Toxoplasma-positrvc chronic HCV patients(P=0.00 l,0.03,0.04,respectively).Child-Pugh scoring for cirrhosis in chronic HCV group placed the majority of seropositive patient in class C with significant statistical difference compared to Child A reference group(OR= 0.08;95%67:0.01-0.5;P=0.003).Conclusions:Toxoplasma seropositivity was high in patients with cirrhosis and associated higher grades of inflammation and necrosis signifying disease evolution,suggesting that cirrhotic patients may thus form a risk group for toxoplasmosis.

This paper is concerned with a vital topic in image processing:color image forgery detection. The development of computing capabilitieshas led to a breakthrough in hacking and forgery attacks on signal, image,and data communicated over networks. Hence, there is an urgent need fordeveloping efficient image forgery detection algorithms. Two main types offorgery are considered in this paper: splicing and copy-move. Splicing isperformed by inserting a part of an image into another image. On the otherhand, copy-move forgery is performed by copying a part of the image intoanother position in the same image. The proposed approach for splicingdetection is based on the assumption that illumination between the originaland tampered images is different. To detect the difference between the originaland tampered images, the homomorphic transform separates the illuminationcomponent from the reflectance component. The illumination histogramderivative is used for detecting the difference in illumination, and henceforgery detection is accomplished. Prior to performing the forgery detectionprocess, some pre-processing techniques, including histogram equalization,histogram matching, high-pass filtering, homomorphic enhancement, andsingle image super-resolution, are introduced to reinforce the details andchanges between the original and embedded sections. The proposed approachfor copy-move forgery detection is performed with the Speeded Up RobustFeatures (SURF) algorithm, which extracts feature points and feature vectors. Searching for the copied partition is accomplished through matchingwith Euclidian distance and hierarchical clustering. in addition, some preprocessing methods are used with the SURF algorithm, such as histogramequalization and single-mage super-resolution. Simulation results proved thefeasibility and the robustness of the pre-processing step in homomorphicdetection and SURF detection algorithms for splicing and copy-move forgerydetection, respectively.

Childhood Poisoning Cases Admitted to Zagazig University Hospitals during the Year 2018: A Retrospective Study

Background: Childhood poisoning is considered major socioeconomic and public health problem as there are thousands of children admitted to the emergency departments and millions of calls are made to poison control centers every year. Aim of the Work: Determine the prevalence and pattern of childhood poisoning cases admitted to Zagazig University hospitals. Subjects and Methods: This is a retrospective study on children Results: A total of 624 childhood poisoning cases, more males than females (55.3% versus 44.7% respectively), and more in age group of 3 - 6 years old (40.86%), more in rural than urban communities (65.06% against 34.94% respectively). Oral exposure was the most against other routes (84.94%). Most cases were unintentional (92.8%). The prevalence of childhood poisoning in descending order was;compound therapeutic medications (32.69%) followed by pesticides (26.92%) then corrosives (17.31%), while volatile hydrocarbons (benzene or kerosene) accounted for (15.38%) and carbon monoxide (3.85%) and others (3.85%). Overall, minor cases were the commonest (63%) while only 3.8% of cases were severe. About 88.94% of cases were discharged after completed management while death rate was 0.96%. Conclusion: Most childhood poisoning cases were males, accidentally, mainly by oral route and in rural areas, most commonly in age group of 3 - 6 years. Most cases were due to therapeutic medications then pesticide exposures. Most of cases were completely cured.

Numerical Treatments of Functional Fredholm integral Equation in 2D with Discontinuous Kernels

This work proposes a new definition of the functional Fredholm integral equation in 2D of the second kind with discontinuous kernels (FT-DFiE). Furthermore, the work is concerned to study this new equation numerically. The existence of a unique solution of the equation is proved. in addition, the approximate solutions are obtained by two powerful methods Toeplitz Matrix Method (TMM) and Product Nystr?m Methods (PNM). The given numerical examples showed the efficiency and accuracy of the introduced methods.

Synthesis, Characteristic and Antimicrobial Activity of Some New Spiro[indol-thiazolidon-2,4-diones] and Bis(5-fluorospiro[indoline-3,2’-thiazolidine]-2,4’-dione) Probes

in a search for new antimicrobial agents, some new spiro[indol-thiazolidon- 2,4-diones] (6a-c) were synthesized by condensation of 5-substituted isatins 1 with sulfanilamide in MeOH, followed by aroylation with p-nitrobenzoyl chloride in DMF to get compounds 4a-c. Cycloaddition of 4a-c with thioglycolic acid in a dry non-polar solvent (dioxane) gave the targets 6a-c. Also, bis(5-fluorospiro[indoline-3,2’-thiazolidine]-2,4’-dione) (9) was synthesized by condensation of 5-fluoroindoline-2,3-dione with benzene-1,4-diamine (2:1 by mol) in MeOH, which followed by cycloaddition with thioglycolic acid in dioxane gave compound 8. Acylation of the later with 2,2,2-trifluoroacetic anhydride in THF has yielded the target 9. Structures of the products have been deduced from their elemental analysis and spectral data. The in vitro antimicrobial activity of the new systems 6a-c, and 9 was tested.

Mismatch repair genes (hMLH1, hPMS1, hPMS2, GTBP/hMSH6,HMSH2) in the pathogenesis of hepatocellular carcinoma

AiM: DMA mismatch repair (MMR) is an important mechanism for maintaining fidelity of genomic DNA. Abnormalities in one or more MMR genes are implicated in the development of many cancers. We investigated the role of expression of MMR genes (hMLH1, hPMS1, hPMS2, GTBP/hMSH6, hMSH2) in hepatocellular carcinogenesis. METHODS: We evaluated the expression level of MMR genes in 33 hepatocellular carcinoma (HCC) cases using the multiplex reverse transcription (RT) PCR assays, as well as in 16 cases of normal adjacent hepatic tissues. β-actin gene was used as an internal control and calibrator for quantification of gene expression. RESULTS: Out of the 33 studied cases, 25 were HCV positive and 30 (90.9%) showed reduced expression in one or more of the studied MMR genes. Reduced expression was found in hMSH2(71.9%), hMLH1 (53.3%), GTBP(51.1%), hPMS2 (33.3%) and hPMS1 (6%). A significant correlation was found between reduced expression of hPMS2(P= 0.0069) and GTBP(P= 0.0034), hPMS2 and non-cirrhosis (P= 0.0197), hMLH1 and high grade. On the other hand, 57.1%, 50%, 20%, 18.8%, and 6% of the normal tissues distant to tumors showed reduced expression of hMSH2, hMLH1, GTBP, hPMS2, and hPMS1 respectively. Multivariate analysis revealed a significant correlation between the expression level of hMSH2(P= 0.008), hMLH1 (P= 0.001) and GTBP (P= 0.032) and HCC, between hPMS2, GTBP and HCV-associated HCC (PiON: Reduced expression of MMR genes seems to play an important role in HCV-associated HCC. hPMS2 is likely involved at an early stage of hepatocarcinogenesis since it was detected in normal adjacent tissues. Reduced expression of hPMS2 provides a growth advantage and stimulates proliferation which encourages malignant transformation in non-cirrhotic HCV-infected patients via acquisition of more genetic damages.

Applying the WHO instead of CDC growth charts may double obesity rates

Aims: This study compares the WHO (2007) and the National Health Examination Survey (NHANES iii) reference intervals and investigates the differences when applied on a Canadian cohort of older children and adolescents. Methods: We calculated height, weight and BMi z-scores of 4375 consecutive patients (1993 female, 45.6%) aged 5 - 20 years attending outpatient clinics at a single tertiary care centre using reference data of the latest NHANES (iii) survey and the WHO (2007) growth charts. To address age dependency, data was stratified into age groups. Results: Using the NHANES iii reference intervals, medians of weight (+0.46), height (+0.29) and BMi z-scores (+0.46) were significantly non-zero. The WHO (2007) growth charts yielded medians of +2.05, +0.32, +0.53 for weight, height and BMi z-scores respectively, all significantly non-zero. When comparing both growth charts, Canadian children had significantly different weight and BMi z-scores (p 95th percentile) doubled from 8.6% to 16.0%. A significant age dependency was observed with higher WHO (2007) weight z-scores (>7 years) and higher BMi z-scores (7 to 13 years) and no significant difference was observed for height z-scores across all age groups. Gender differences were observed for weight z-scores (>9 years) and BMi (males: 9 - 11 years, p = 0.0118;11 - 13 years, p = 0.0069) whereas no significant difference was found in height z-scores across all age groups. Conclusion: Our results reveal substantial differences between both reference populations and thus interpretation needs be done with caution, especially when labelling results as abnormal.
