The effects of selenium on the translation efficiency of selenoproteins Gpx1， Gpx4， and Txnrd1 in oxLDL-induced EA.hy926 cells

Selenium is a crucial trace element that contributes to physiological processes in the body as selenoproteins. Selenoproteins serve as an integral role in the body in controlling the redox state of cells and protecting against damage induced by oxidative stress. This study aimed to investigate the effects and possible mechanism of selenium on selenoproteins expression in EA.hy926 cells induced by oxLDL. The impact of selenium on the viability of EA.hy926 cells was detected by the MTT method， and intracellular ROS level and mitochondrial membrane potential were assessed by fluorescent probe DCFH-DA and JC-1， respectively. RNA-seq， quantitative real-time PCR (RT-qPCR)， and western blot were used to investigate the selenoprotein expression. Selenoprotein mRNA translation efficiency was analyzed by ribosome profiling (Ribo-Seq) coupled with transcriptomics. Our data showed that selenium supplementation (0.5 μmol/L) significantly decreased ROS production， reduced mitochondrial inner membrane potential and increased the proliferative activity of EA.hy926 cells induced by oxLDL. Moreover， The protective effects of selenium against oxLDL-induced EA.hy926 cell injury were associated with the upregulation of the expressions of selenoproteins GPX1， GPX4， and TXNRD1. Furthermore， the expressions of selenoproteins GPX1 and GPX4 were hierarchically controlled， but the expressions of selenoproteins TXNRD1 were mainly regulated by oxLDL. Finally， Ribo-Seq coupled with transcriptomics results demonstrated that the expressions of selenoproteins GPX1， GPX4， and TXNRD1 were regulated at the translation process level. These findings suggested that selenium could have preventive effects in oxLDL induced EA.hy926 cell injury by regulating the selenoprotein expression， and the selenoproteins expressions at the translation level in vascular endothelial cells need further study.

Drought is one of the major abiotic threat to rice production in the context of climate change.Super Basmati is an elite,fine grain basmati rice variety grown in Punjab,Pakistan.Due to drought sensitive in nature,its yield has been facing an alarming situation in production because of gradual decrease in irrigated water for a couple of years.Three reported novel QTLs for drought tolerance were selected for incorporation into Super Basmati by employing marker assisted selection strategy.IR55419-04 with novel QTLs was used as a donor parent.Foreground selection was performed by applying PCR based QTL linked SSR markers followed by recombinant selection by using 2-4 flanking markers.Background selection was exercised by using polymorphic SSR markers for maximum genome recovery of the Super Basmati.The individuals homozygous at the target QTLs and with maximum background of Super Basmati at the rest of the non-target genome was selected for evaluation of drought tolerance.Under drought stress conditions,the yields of all introgressed lines(ILs)were 44.2%-125.7%higher than recurrent parent.Six superior ILs that are drought tolerant and very similar to Super Basmati in terms of agronomic and grain quality traits are marked for release as drought-tolerant varieties in arid regions or for use in breeding programs of high grain quality and drought-tolerant parents.

Background and Aims:Liver transplantation(LT)using ABO-incompatible(ABOi)grafts can extend the donor pool to a certain extent and hence reduce the waiting time for transplantation.However,concerns of the impending prognosis associated with this option,especially for patients with liver failure and higher model for end-stage liver disease(MELD)scores,who tend to be more fragile during the waiting period before LT.Methods:Recipients undergoing LT for acute-onchronic liver failure or acute liver failure were retrospectively enrolled at four institutions.Overall survival was compared and a Cox regression analysis was performed.Propensity score matching was performed for further comparison.Patients were stratified by MELD score and cold ischemia time(CIT)to determine the subgroups with survival benefits.Results:Two hundred ten recipients who underwent ABOi LT and 1,829 who underwent ABO compatible(ABOc)LT were enrolled.The 5-year overall survival rate was significantly inferior in the ABOi group compared with the ABOc group after matching(50.6%vs.75.7%,p0.05).Comparison of the survival rates revealed no statistically significant difference for patients with MELD scores≥40(p>0.05).For patients with MELD scores of 31-39,the overall survival rate was significantly inferior in the ABOi group compared with the ABOc group(p<0.001);however,the rate was increased when the liver graft CIT was<8 h.Conclusions:For recipients with MELD scores≤30,ABOi LT had a prognosis comparable to that of ABOc LT and can be regarded as a feasible option.For recipients with MELD scores≥40,ABOi should be adopted with caution in emergency cases.For recipients with MELD scores of 31-39,the ABOi LT prognosis was worse.However,those patients benefited from receiving ABOi grafts with a CIT of<8 h.

Sprouts are ready-to-eat and are recognized worldwide as functional components of the human diet.Recent advances in innovative agricultural techniques could enable an increase in the production of healthy food.The use of light-emitting diode(LED)in indoor agricultural production could alter the biological feedback loop,increasing the functional beneﬁts of plant foods such as wheat and lentil sprouts and promoting the bioavailability of nutrients.The effects of white(W),red(R),and blue(B)light were investigated on the growth parameters and nutritional value of wheat and lentil sprouts.In the laboratory,seeds were sown under three different LED treat-ments:white,red,and blue light,while normal incandescent light served as a control.Percentage seed germina-tion improved by 18.34%and 12.67%for wheat and 18.34%and 12.67%for lentil sprouts under LED treatments R and B,respectively.An increase in total soluble protein and sugar by 33.4%and 9.23%in wheat and by 31.5%and 5.87%in lentils was observed under the R LED treatment.Vitamin C concentrations in wheat and lentils were signiﬁcantly increased by R LED compared to all other treatments.Other parameters,including potassium and sodium concentrations,were signiﬁcantly increased under red and blue light compared to the control;white light,on the other hand,signiﬁcantly decreased all these parameters.According to the experimental data,red and blue LED light could be beneﬁcial in the production of functional wheat and lentil sprouts with high nutrient concentrations.

A consensus meeting of national experts from all major national hepatobiliary centres in the country was held on May 26,2023,at the Pakistan Kidney and Liver Institute&Research Centre(PKLI&RC)after initial consultations with the experts.The Pakistan Society for the Study of Liver Diseases(PSSLD)and PKLI&RC jointly organised this meeting.This effort was based on a comprehensive literature review to establish national practice guidelines for hilar cholangiocarcinoma(hCCA).The consensus was that hCCA is a complex disease and requires a multidisciplinary team approach to best manage these patients.This coordinated effort can minimise delays and give patients a chance for curative treatment and effective palliation.The diagnostic and staging workup includes high-quality computed tomography,magnetic resonance imaging,and magnetic resonance cholangiopancreato-graphy.Brush cytology or biopsy utilizing endoscopic retrograde cholangiopancreatography is a mainstay for diagnosis.However,histopathologic confirmation is not always required before resection.Endoscopic ultrasound with fine needle aspiration of regional lymph nodes and positron emission tomography scan are valuable adjuncts for staging.The only curative treatment is the surgical resection of the biliary tree based on the Bismuth-Corlette classification.Selected patients with unresectable hCCA can be considered for liver transplantation.Adjuvant chemotherapy should be offered to patients with a high risk of recurrence.The use of preoperative biliary drainage and the need for portal vein embolisation should be based on local multidisciplinary discussions.Patients with acute cholangitis can be drained with endoscopic or percutaneous biliary drainage.Palliative chemotherapy with cisplatin and gemcitabine has shown improved survival in patients with irresectable and recurrent hCCA.

花后弱光对软质小麦籽粒产量与淀粉品质的影响

为探明花后弱光胁迫对软质小麦籽粒产量、淀粉粒度分布及黏度参数的影响,以软质小麦品种荃麦725和扬麦15为材料,在小麦开花后设置3个弱光处理即S1 (遮光10%)、S2(遮光20%)、S3(遮光30%)处理,以自然光照条件为对照,分析花后不同程度弱光条件下软质小麦籽粒产量、淀粉粒度分布与黏度参数的差异。结果表明,与对照相比,花后弱光降低了小麦粒重、穗粒数和产量,提高了籽粒蛋白质含量,但使籽粒淀粉含量、籽粒蛋白质和淀粉积累量显著减少。花后弱光降低了软质小麦B型淀粉粒(粒径≤10μm)体积、表面积百分比,增加了A型淀粉粒(粒径>10μm)体积、表面积百分比,对A、B型淀粉粒数目百分比无显著影响。在B型淀粉粒中,弱光对粒径0.1～2.8μm的淀粉粒体积百分比的影响较粒径2.8～10μm的淀粉粒大。在A型淀粉粒中,弱光对粒径>22μm的淀粉粒体积百分比的影响大于粒径10～22μm的淀粉粒。弱光导致小麦籽粒淀粉峰值黏度、低谷黏度、最终黏度、稀懈值、回升值等参数显著下降。花后弱光虽然可增加软质小麦籽粒蛋白质含量,但显著降低籽粒淀粉含量以及蛋白质和淀粉积累量,不利于粒重与产量形成;弱光对B型淀粉粒的影响大于A型淀粉粒,会降低B型淀粉粒体积和表面积占比,但可增加A型淀粉粒体积和表面积占比,降低淀粉峰值黏度等参数。

Dramatic changes in climatic conditions that supplement the biotic and abiotic stresses pose severe threat to the sustainable rice production and have made it a difficult task for rice molecular breeders to enhance production and productivity under these stress factors. The main focus of rice molecular breeders is to understand the fundamentals of molecular pathways involved in complex agronomic traits to increase the yield. The availability of complete rice genome sequence and recent improvements in rice genomics research has made it possible to detect and map accurately a large number of genes by using linkage to DNA markers. Linkage mapping is an effective approach to identify the genetic markers which are co-segregating with target traits within the family. The ideas of genetic diversity, quantitative trait locus（QTL） mapping, and marker-assisted selection（MAS） are evolving into more efficient concepts of linkage disequilibrium（LD） also called association mapping and genomic selection（GS）, respectively. The use of cost-effective DNA markers derived from the fine mapped position of the genes for important agronomic traits will provide opportunities for breeders to develop high-yielding, stress-resistant, and better quality rice cultivars. Here we focus on the progress of molecular marker technologies, their application in genetic mapping and evolution of association mapping techniques in rice.

COVID-19 Outbreak Prediction by Using Machine Learning Algorithms

COVID-19 is a contagious disease and its several variants put under stress in all walks of life and economy as well.Early diagnosis of the virus is a crucial task to prevent the spread of the virus as it is a threat to life in the whole world.However,with the advancement of technology,the Internet of Things(IoT)and social IoT(SIoT),the versatile data produced by smart devices helped a lot in overcoming this lethal disease.Data mining is a technique that could be used for extracting useful information from massive data.In this study,we used five supervised ML strategies for creating a model to analyze and forecast the existence of COVID-19 using the Kaggle dataset“COVID-19 Symptoms and Presence.”RapidMiner Studio ML software was used to apply the Decision Tree(DT),Random Forest(RF),K-Nearest Neighbors(K-NNs)and Naive Bayes(NB),Integrated Decision Tree(ID3)algorithms.To develop the model,the performance of each model was tested using 10-fold cross-validation and compared to major accuracy measures,Cohan’s kappa statistics,properly or mistakenly categorized cases and root means square error.The results demonstrate that DT outperforms other methods,with an accuracy of 98.42%and a root mean square error of 0.11.In the future,a devisedmodel will be highly recommendable and supportive for early prediction/diagnosis of disease by providing different data sets.

Convolutional Neural Network Model for Fire Detection in Real-Time Environment

Disasters such as conflagration,toxic smoke,harmful gas or chemical leakage,and many other catastrophes in the industrial environment caused by hazardous distance from the peril are frequent.The calamities are causing massive fiscal and human life casualties.However,Wireless Sensors Network-based adroit monitoring and early warning of these dangerous incidents will hamper fiscal and social fiasco.The authors have proposed an early fire detection system uses machine and/or deep learning algorithms.The article presents an Intelligent Industrial Monitoring System(IIMS)and introduces an Industrial Smart Social Agent(ISSA)in the Industrial SIoT(ISIoT)paradigm.The proffered ISSA empowers smart surveillance objects to communicate autonomously with other devices.Every Industrial IoT(IIoT)entity gets authorization from the ISSA to interact and work together to improve surveillance in any industrial context.The ISSA uses machine and deep learning algorithms for fire-related incident detection in the industrial environment.The authors have modeled a Convolutional Neural Network(CNN)and compared it with the four existing models named,FireNet,Deep FireNet,Deep FireNet V2,and Efficient Net for identifying the fire.To train our model,we used fire images and smoke sensor datasets.The image dataset contains fire,smoke,and no fire images.For evaluation,the proposed and existing models have been tested on the same.According to the comparative analysis,our CNN model outperforms other state-of-the-art models significantly.

The global population is increasing rapidly as compared to food production;approximately three times more food would be required in 2050.Climate change affects crop production by causing sudden changes in weather conditions,including rain,storms,heat waves,doughiness,and water shortages.Farming with smart technology provides a productive solution.Smart farming is a productive solution that provides a great resource of income and improves the countries’economy by exporting consumable goods and preventing food security problems.Smart agriculture provides a combination of flexibility,remote access,and automation through the use of intelligent control technologies.Many countries are working towards smart and intelligent agriculture farming that analyzes crop,soil fertility,pests and weeds,and other problems caused by mismanagement and incompetence.However,smart agricultural farming is less widely adopted in agriculture as a result of high costs and little understanding of technology.In this study,An artificial climate control chamber(ACCC)was designed for cultivating plants by controlling the optimal parameters,especially the light spectrum.In ACCC,influential plant factors such as light,moisture,humidity,and fertilizer concentration have been controlled intelligently.Light spectrum was controlled by time periods in the previous system,while in the system proposed in this study,the light was controlled by image processing.In an artificial control chamber,the plant growth stages have been determined through image processing techniques.Datasets of image images have been used to organize specific intensities of the light spectrum.This intelligent system provides aid in the speed breeding procedure through variant spectrums of light and fertilizers combinations.In the research study,the yield and quality of intelligent farming are enhanced.
