Cultural characteristics of chromium resistant unicellular cyanobacteria isolated from local environment in Pakistan

Many unicellular cyanobacteria were isolated from different places： fields, ponds, polluted water, and soils from Muredkey and Kasur tannery areas, near Lahore, Pakistan. Different media like BG 11 medium, Bold Basal medium, Chu＇s # 10 medium and Gotham＇s medium, in standard forms and with slight variations of ingredients, and different pH, temperature and light regimes were checked for the optimum growth of the isolates. The isolation pro- cedure was repeated with different concentrations of chromium to select the resistant strains. These selected strains grew on chromium of the range 100-200 μg/ml in BG 11 medium. Cyanobacteria were maintained in solid and liquid media with/without shaking. Cyanobacterial strains were collected from natural habitats that were accompanied by a diversified group of organisms including bacteria, protozoan, and rotifers etc. In order to eliminate these agents termed as contaminants, we used several methods including phenol treatment, use of antibiotic and careful manual picking of unicellular cyanobacteria. Resistance of these strains against different heavy metals （ZnSO4, MnSO4, NiSO4, COCl2, Pb（NO3）3, CuSO4, HgCl2, AgNO3 and CdCl2） and antibiotics （erythromycin, streptomycin, kanamycin, chloramphenicol, neomycin） was evaluated. Optimum temperature was 30℃ with variable pH for the reduction of Cr^6＋ in to Cr^3＋ in majority of strains.

AIM： To map Usher phenotype in a consanguineous Pakistani family and identify disease-associated mutation in a causative gene to establish phenotype-genotype correlation.· METHODS： A consanguineous Pakistani family in which Usher phenotype was segregating as an autosomal recessive trait was ascertained. On the basis of results of clinical investigations of affected members of this family disease was diagnosed as Usher syndrome（USH）. To identify the locus responsible for the Usher phenotype in this family, genomic DNA from blood sample of each individual was genotyped using microsatellite Short Tandem Repeat（STR） markers for the known Usher syndrome loci. Then direct sequencing was performed to find out disease associated mutations in the candidate gene.· RESULTS： By genetic linkage analysis, the USH phenotype of this family was mapped to PCDH15 locus on chromosome 10q21.1. Three different point mutations in exon 11 of PCDH15 were identified and one of them,c.1304AC was found to be segregating with the disease phenotype in Pakistani family with Usher ***, c.1304 A C transversion mutation predicts an amino-acid substitution of aspartic acid with an alanine at residue number 435（p.D435A） of its protein ***, in silico analysis revealed conservation of aspartic acid at position 435 and predicated this change as pathogenic.·CONCLUSION：Theidentificationofc.1304ACpathogenic mutation in PCDH15 gene and its association with Usher syndrome in a consanguineous Pakistani family is thefirst example of a missense mutation of PCDH15 causing USH1 phenotype. In previous reports, it was hypothesized that severe mutations such as truncated protein of PCDH15 led to the Usher I phenotype and that missense variants are mainly responsible for non-syndromic hearing impairment.

Shrubby seablite or lani(Suaeda fruticosa Forssk)is a perennial euhalophyte with succulent leaves,which could be planted on arid-saline lands for restoration and cultivated as a non-conventional edible or cash *** about the impacts of maternal saline environment on seed attributes of this important euhalophyte is *** study investigated the effects of maternal salinity on yield,size and stress tolerance of *** *** of *** were grown in a green net house under increasing maternal salinity levels(0,300,600 and 900 mM NaCl)until seed *** yield,size,stress tolerance and germination of the descended seeds under different maternal saline conditions were *** grown under saline conditions(300,600 and 900 mM NaCl)produce a substantially higher quantity of seeds than plants grown under non-saline condition(0 mM NaCl).Low maternal salinity(300 mM NaCl)improves seed *** produced under all maternal salinity levels display a higher tolerance to low temperature(night/day thermoperiod of 10℃/20℃),whereas seeds produced under 300 mM NaCl maternal saline condition show a better tolerance to high temperature(night/day thermoperiod of 25℃/35℃)during *** from all maternal saline conditions germinate better in the 12 h photoperiod(12 h light/12 h dark)than in the dark(24 h dark);however,seeds produced from low and moderate maternal saline conditions(300 and 600 mM NaCl)show a higher germination in the dark than those from control and high maternal saline conditions(0 and 900 mM NaCl).In general,maternal salinity is found to improve yield,size and stress tolerance of *** seeds.

Clustering algorithms optimization can minimize topology maintenance overhead in large scale vehicular Ad hoc networks(VANETs)for smart transportation that results from dynamic topology,limited resources and noncentralized *** performance of a clustering algorithm varies with the underlying mobility model to address the topology maintenance overhead issue in VANETs for smart *** design a robust clustering algorithm,careful attention must be paid to components like mobility models and performance objectives.A clustering algorithm may not perform well with every mobility ***,we propose a supervisory protocol(SP)that observes the mobility pattern of vehicles and identies the realistic Mobility model through microscopic *** analytical model can be used to determine an efcient clustering algorithm for a specic mobility model(MM).SP selects the best clustering scheme according to the mobility model and guarantees a consistent performance throughout VANET *** simulation has performed in three parts that is the central part simulation for setting up the clustering environment,In the second part the clustering algorithms are tested for efciency in a constrained atmosphere for some time and the third part represents the proposed *** simulation results show that the proposed scheme outperforms clustering algorithms such as honey bee algorithm-based clustering and memetic clustering in terms of cluster count,re-afliation rate,control overhead and cluster lifetime.

The lactose-fermenter Enterobacteriaceae are the most frequent cause of clinical infection in our country. The objective of this study was to isolate and identify the most common lactose-fermenter Enterobacteriaceae from clinical samples, including urine, blood, wounds, and sputum, obtained from the local hospital and from environmental samples from a chicken farm, agriculture soil, and water from the Tigris River in Baghdad City. The study also aimed at establishing the antibiotic resistance patterns of the isolated bacteria. A total of 155 bacterial isolates were identified from 10 genera according to the Vitek 2 system. The most common bacterial isolates from the clinical and environmental samples were Escherichia coli and Klebsiella pneumoniae, respectively. The antibiotic resistance patterns showed that all clinical and environmental isolates were multidrug resistant to β-lactam (except carbapenems) drug and aminoglycosides and more sensitive to carbapenems.

Distance relays are normally used to protect transmission lines. They respond to the impedance Between the relay location and the fault location as the impedance per mile of a transmission Line is fairly constant, these relays respond to the distance to a fault on the transmission line - and Hence their name as will be seen shortly, under certain conditions it may be desirable to make Distance relays respond to some parameter other than the impedance, such as the admittance or the Reactance up to the fault location. Electrical power is most important in everyday life it consists of generators, transformers, and transmission and distribution lines etc. system of protection offering considerable economic and technical advantages. Unlike phase and neutral overcurrent protection, the key advantage of distance protection is that its fault coverage of the protected circuit is virtually independent of source impedance variations during the transmissions faults can occur which cause damage to the power system process. The system also loses its stability and comfortable zones when fault occurs. When a fault occur in an element of a power system then automatic protective device is needed to isolate the faults element as soon as possible to keep the healthy section of the system in normal condition. A protective system includes circuit breakers and protective relays to isolate the faults section of the system from the healthy sections. These protective elements should be able to analyze the type of fault that occurred during abnormal conditions. In this, we have analyzed different kinds of faults over a transmission line placed between two buses. The simulation studies is carried out using PSCAD/EMTDC software, and the results obtained is verified theoretically. Distance relays are used to detect and protect from faults in transmission lines.
