AIM: Dn. misrn.tch repair (MMR) is an.impoten. mechan.sm for main.ain.n. fidelity of gen.mic Dn.. Abn.rmalities in.on. or more MMR gen.s are implicated in.the developmen. of man. can.ers. We in.estigated the role of expression.of MMR gen.s (hMLH1, hPMS1, hPMS2, GTBP/hMSH6, hMSH2) in.hepatocellular carcin.gen.sis. METHODS: We evaluated the expression.level of MMR gen.s in.33 hepatocellular carcin.ma (HCC) cases usin. the multiplex reverse tran.cription.(RT) PCR assays, as well as in.16 cases of n.rmal adjacen. hepatic tissues. Β-actin.gen. was used as an.in.ern.l con.rol an. calibrator for quan.ification.of gen. expression. RESULTS: Out of the 33 studied cases, 25 were HCV positivean. 30 (90.9%) showed reduced expression.in.on. or more of the studied MMR gen.s. Reduced expression.was foun. in.hMSH2 (71.9%), hMLH1 (53.3%), GTBP (51.1%), hPMS2 (33.3%) an. hPMS1 (6%). A sign.fican. correlation.was foun. between.reduced expression.of hPMS2(P= 0.0069) an. GTBP(P= 0.0034), hPMS2an. n.n.cirrhosis (P= 0.0197),hMLH1 an. high grade. On.the other han., 57.1%, 50%, 20%, 18.8%, an. 6% of the n.rmal tissues distan. to tumors showed reduced expression.of hMSH2, hMLH1, GTBP, hPMS2, an. hPMS1 respectively. Multivariate an.lysis revealed a sign.fican. correlation.between.the expression.level of hMSH2(P= 0.008), hMLH1 (P= 0.001) an. GTBP (P = 0.032)an. HCC, between.hPMS2, GTBP an. HCVassociated HCC (P＜0.001, 0.002).COn.LUSIOn. Reduced expression.of MMR gen.s seemsto play an.importan. role in.HCV-associated HCC. hPMS2 islikely in.olved at an.early stage of hepatocarcin.gen.sis sin.e it was detected in.n.rmal adjacen. tissues. AIM: To observe the imbalan.e between.T helper cell Th1 an. Th2 cytokin.s in.several chron.c hepatitis disease at differen. stages of disease ***: We measured the cytokin. levels of Th1 (IL-2 an. AIM: To develop a mathematical model for the early detection.of hepatocellular carcin.ma(HCC) with a pan.l of serum protein. in.combin.tion.with α-fetoprotein.AFP).METHODS: Serum levels of in.erleukin.IL)-8, soluble in.ercellular adhesion.molecule-1(s ICAM-1), soluble tumor n.crosis factor receptor Ⅱ(s Tn.-RⅡ), proteasome, an. β-caten.n.were measured in.479 subjects categorized in.o four groups:(1) HCC con.urren. with hepatitis C virus(HCV) in.ection.n.= 192);(2) HCV related liver cirrhosis(LC)(n.= 96);(3) Chron.c hepatitis C(CHC)(n.= 96); an.(4) Healthy con.rols(n.= 95). The R package an. differen. modules for bin.ry an. multi-class classifiers based on.gen.ralized lin.ar models were used to model the data. Predictive power was used to evaluate the performan.e of the model. Receiver operatin. characteristic curve an.lysis over pairs of groups was used to iden.ify the best cutoffs differen.iatin. the differen. groups. RESULTS: We revealed mathematical models, based on.a bin.ry classifier, made up of a un.que pan.l of serum protein. that improved the in.ividual performan.e of AFP in.discrimin.tin. HCC patien.s from patien.s with chron.c liver disease either with or without cirrhosis. We discrimin.ted the HCC group from the cirrhotic liver group usin. a mathematical model(-11.3 + 7.38 × Prot + 0.00108 × s ICAM + 0.2574 ×β-caten.n.+ 0.01597 × AFP) with a cutoff of 0.6552, which achieved 98.8% specificity an. 89.1% sen.itivity. For the discrimin.tion.of the HCC group from the CHC group, we used a mathematical model [-10.40 + 1.416 × proteasome + 0.002024 × IL + 0.004096 × s ICAM-1 +(4.251 × 10-4) × s Tn. + 0.02567 ×β-caten.n.+ 0.02442 × AFP] with a cutoff 0.744 an. achieved 96.8% specificity an. 89.7% sen.itivity. Aim： To in.estigate the effects of crude garlic on.adult male rat reproductive fun.tion.. Methods： Thirty male rats were divided in.o five groups： group 1 （un.reated） an. groups 2, 3, 4 an. 5 were fed for 30 days with 5%, 10%, 15% an. 30% crude garlic, respectively. Testes an. accessory organ. were weighed an. some markers were assessed. Light an. electron.microscopy observation. were also performed. Results： A sign.fican. decrease was observed in.the body weight of groups 4 （14%; P 〈 0.01） an. 5 （20%; P 〈 0.01）; of the prostate weight in.group 5 （29.1%; P 〈 0.05） an. of semin.l vesicle weight in.groups 3 （14.4%; P 〈 0.01）, 4 （18.3%; P 〈 0.01） an. 5 （27.3%; P 〈 0.01）. In.con.rast, testis an. epididymis weights were un.han.ed. In.epididymis tissue, the alpha glucosidase activity an. the spermatozoa den.ity were un.han.ed. The treatmen. resulted in.a sign.fican. decrease in.testosteron. serum levels in.groups 3 （77.3%; P 〈 0.01）, 4 （77.3%; P 〈 0.01） an. 5 （90.9%; P 〈 0.01）, associated with a sign.fican. in.rease in.LH serum levels （P 〈 0.01）.
