Novel CYP1B1 mutations and a possible prognostic use for surgical management of congenital glaucoma

AIM: To identify CYP1B1 gene mutations and evaluate their possible role as a prognostic factor for success rates in the surgical management of Egyptian congenital glaucoma patients.METHODS: Totally 42 eyes of 29 primary congenital glaucoma patients were operated on with combined trabeculotomy/trabeculectomy with mitomycin-C and followed up at 1 d, 1 wk, 1, 6 and 12 mo postoperatively. Genomic DNA was extracted from peripheral blood leukocytes. Coding regions of CYP1B1 gene were amplified using 13 pairs of primers, screened for mutations using single-strand conformation polymorphism followed by sequencing of both strands. Efficacy of the operation was graded as either a success [maintaining intraocular pressure(IOP) less than 21 mm Hg with or without antiglaucoma medication], or a failure(IOP more than 21 mm Hg with topical antiglaucoma medications). RESULTS: Seven novel mutations out of a total of 15 different mutations were found in the CYP1B1 genes of 14 patients(48.2%). The presence of CYP1B1 gene mutations did not correlate with the failure of the surgery(P=0.156, odds ratio=3.611, 95%CI, 0.56 to 22.89); while the positive consanguinity strongly correlated with failure of the initial procedure(P=0.016, odds ratio=11.25, 95%CI, 1.57 to 80.30). However, the Kaplan-Meier survival analysis revealed a significantly lower time of IOP control in the subgroup with mutations in CYP1B1 versus the congenital primary glaucoma group without mutations(log rank test, P=0.015).CONCLUSION: Seven new CYP1B1 mutations are identified in Egyptian patients. Patients harboring confirmed mutations suffered from early failure of the initial surgery. CYP1B1 mutations could be considered as a prognostic factor for surgery in primary congenital glaucoma.

The main cable bent around the saddle of the suspension bridge is subjected to the wind,the vehicle,the bridge’s own weight and the corrosive media.The coupling of the three loads and the environments causes the friction,the corrosion,and the fatigue(tribo-corrosion-fatigue)among the wires inside the main cable.In this paper,a wire bending tribo-corrosion-fatigue test rig was designed and developed.The effect of fatigue load on the bending friction behaviors between the cable wires in ultrapure water and 3.5%NaCl solution was explored.The tribological properties and electrochemical corrosion behaviors under different fatigue loading ranges were investigated.The tribo-corrosion-fatigue interaction between the cable wires was quantitatively characterized,and the mechanism of the interaction was analyzed.The results demonstrate that the increasing fatigue load exacerbates the coupling damage of the cable wires attributed to the enhanced interaction.The findings carry theoretical importance when assessing the main cable’s deterioration and the load-bearing safety of a suspension bridge.

Effect of wear debris on fretting fatigue crack initiation

Both wear and fatigue occur in fretting condition,and they interact with one another during the whole process.Fretting fatigue is commonly analysed without considering the effect of wear in partial slip regime,although wear affects the lifetime of crack initiation.This paper investigates,for the first time,the effect of wear debris on fretting fatigue crack initiation.To investigate the effect of debris,first fretting wear characteristics in partial slip regime are analysed for loading conditions.Then,the effect of wear on fretting fatigue crack initiation is investigated using Ruiz parameters and critical plane methods without considering the debris effect.Through the results,we can see that loading conditions affect the wear profiles in different ways.Moreover,wear has a significant effect on the fatigue in partial slip regime without considering debris especially on the crack initiation location.Finally,considering wear debris in the analysis,its effect on critical plane parameters is investigated.It is found that by considering the wear debris effect,the frtting fatigue crack initiation location is shifted towards the trailing edge.The predictions of both crack initiation location and lifetime show a good agreement with the experimental data.

In this paper,a methodology integrating crystal plasticity(CP),the eXtended finite element method(XFEM)and the cohesive zone model(CZM)is developed for an Al-Cu-Mg alloy to predict fatigue crack propagation(FCP)across grain boundary(GB)of Al-Cu-Mg alloy during stageІІ.One GB model is incor-porated into FCP constitutive law to describe grain interaction at GB.A bicrystal containing GB is built up to simulate FCP behavior through L participated GBs.Modelling features including GB characteristic,cumulative plastic strain(CPS)distribution and crystal slipping evidence can be identified.The numer-ical results are compared with published experimental data to check the accuracy of model.This work demonstrates that the combination of CP containing GB constitutive laws,XFEM and CZM is a promising methodology in predicting twist angle-controlled crack deflection through GBs.

Enhanced Heap-Based Optimizer Algorithm for Solving Team Formation Problem

Team Formation(TF)is considered one of the most significant problems in computer science and optimization.TF is defined as forming the best team of experts in a social network to complete a task with least cost.Many real-world problems,such as task assignment,vehicle routing,nurse scheduling,resource allocation,and airline crew scheduling,are based on the TF problem.TF has been shown to be a Nondeterministic Polynomial time(NP)problem,and high-dimensional problem with several local optima that can be solved using efficient approximation algorithms.This paper proposes two improved swarm-based algorithms for solving team formation problem.The first algorithm,entitled Hybrid Heap-Based Optimizer with Simulated Annealing Algorithm(HBOSA),uses a single crossover operator to improve the performance of a standard heap-based optimizer(HBO)algorithm.It also employs the simulated annealing(SA)approach to improve model convergence and avoid local minima trapping.The second algorithm is the Chaotic Heap-based Optimizer Algorithm(CHBO).CHBO aids in the discovery of new solutions in the search space by directing particles to different regions of the search space.During HBO’s optimization process,a logistic chaotic map is used.The performance of the two proposed algorithms(HBOSA)and(CHBO)is evaluated using thirteen benchmark functions and tested in solving the TF problem with varying number of experts and skills.Furthermore,the proposed algorithms were compared to well-known optimization algorithms such as the Heap-Based Optimizer(HBO),Developed Simulated Annealing(DSA),Particle SwarmOptimization(PSO),GreyWolfOptimization(GWO),and Genetic Algorithm(GA).Finally,the proposed algorithms were applied to a real-world benchmark dataset known as the Internet Movie Database(IMDB).The simulation results revealed that the proposed algorithms outperformed the compared algorithms in terms of efficiency and performance,with fast convergence to the global minimum.

This paper investigates a polygonal finite element(PFE)to solve a two-dimensional(2D)incompressible steady fluid problem in a cavity square.It is a well-known standard benchmark(i.e.,lid-driven cavity flow)-to evaluate the numerical methods in solving fluid problems controlled by the Navier-Stokes(N-S)equation system.The approximation solutions provided in this research are based on our developed equal-order mixed PFE,called Pe1Pe1.It is an exciting development based on constructing the mixed scheme method of two equal-order discretisation spaces for both fluid pressure and velocity fields of flows and our proposed stabilisation technique.In this research,to handle the nonlinear problem of N-S,the Picard iteration scheme is applied.Our proposed method’s performance and convergence are validated by several simulations coded by commercial software,i.e.,MATLAB.For this research,the benchmark is executed with variousReynolds numbers up to the maximum Re=1000.All results then numerously compared to available sources in the literature.

Rescue effects of aqueous seed extracts of Foeniculum vulgare and Carum carvi against cadmium-induced hepatic, renal and gonadal damage in female albino rats

Objective: To investigate the protective effects of aqueous seed extracts of fennel"Foeniculum vulgare"(FVE) and caraway "Carum carvi"(CCE) on liver, kidney and reproductive organs in female rats against cadmium chloride(CC) intoxication.Methods: A total of 36 adult female rats were divided into six groups, six in each group.Control group(fed normal diet), CC-treated group(50 mg CC/kg diet), CCE-treated group(150 mg CCE/kg diet), CCE + CC group, FVE(150 mg/kg diet) and FVE + CC. One month later, all rats were sacrificed and all samples were collected at proestrus phase.Results: The toxic effects of CC were confirmed biochemically by significant increase of serum concentration of liver enzymes(P < 0.05), and creatinine(P < 0.001). Moreover,CC increased significantly the serum level of malondialdehyde(MDA) and decreased the total antioxidant capacity(TAC)(P < 0.001). In addition, serum concentrations of estrogen, progesterone, follicle-stimulating hormone(FSH) and luteinizing hormone(LH)were significantly decreased(P < 0.01). Histopathologically, CC-treated group revealed marked pathological changes in renal, hepatic, ovarian and uterine tissues. All toxic findings observed in liver and kidney with CC treatment were found to be ameliorated markedly after co-treatments with FVE or CCE. Furthermore, co-treatment of FVE with CC improved significantly all studied reproductive parameters(P < 0.01).Conclusions: Both FVE and CCE could be used as efficient treatments for liver and kidney against CC intoxication. Moreover, FVE could be utilized as a potent treatment to protect and improve female fertility from cadmium intoxication.

Combining Committee-Based Semi-Supervised Learning and Active Learning

Many data mining applications have a large amount of data but labeling data is usually difficult, expensive, or time consuming, as it requires human experts for annotation. Semi-supervised learning addresses this problem by using unlabeled data together with labeled data in the training process. Co-Training is a popular semi-supervised learning algorithm that has the assumptions that each example is represented by multiple sets of features （views） and these views are sufficient for learning and independent given the class. However, these assumptions axe strong and are not satisfied in many real-world domains. In this paper, a single-view variant of Co-Training, called Co-Training by Committee （CoBC） is proposed, in which an ensemble of diverse classifiers is used instead of redundant and independent views. We introduce a new labeling confidence measure for unlabeled examples based on estimating the local accuracy of the committee members on its neighborhood. Then we introduce two new learning algorithms, QBC-then-CoBC and QBC-with-CoBC, which combine the merits of committee-based semi-supervised learning and active learning. The random subspace method is applied on both C4.5 decision trees and 1-nearest neighbor classifiers to construct the diverse ensembles used for semi-supervised learning and active learning. Experiments show that these two combinations can outperform other non committee-based ones.

Many underdeveloped countries still struggle with the public health issue of acute illnesses,especially with the emergence of injuries as a major category of acute diseases.Control of the acute disease is challenging particularly when it occurs as an outbreak,either in isolation or as part of a wider epidemic.This narrative review summarizes the definition,epidemiologic transition,pattern and modes of spread,classification,epidemiologic measures,community burden,prevention,control,and future challenges of acute disease.This review is to provide a reference for epidemiologists,clinicians,researchers,and policymakers.

Braking efficiency is characterized by reduced braking time and distance,and therefore passenger safety depends on the design of the braking system.During the braking of a vehicle,the braking system must dissipate the kinetic energy by transforming it into heat energy.A too high temperature can lead to an almost total loss of braking efficiency.An excessive rise in brake temperature can also cause surface cracks extending to the outside edge of the drum friction surface.Heat transfer and temperature gradient,not to forget the vehicle’s travel environment(high speed,heavy load,and steeply sloping road conditions),must thus be the essential criteria for any brake system design.The aim of the present investigation is to analyze the thermal behavior of different brake drum designs during the single emergency braking of a heavy-duty vehicle on a steeply sloping road.The calculation of the temperature field is performed in transient mode using a three-dimensional finite element model assuming a constant coefficient of friction.In this study,the influence of geometrical brake drum configurations on the thermal behavior of brake drums with two different materials in grey cast iron FG200 and aluminum alloy 356.0 reinforced with silicon carbide(SiC)particles is analyzed under extreme vehicle braking conditions.The numerical simulation results obtained using FE software ANSYS are qualitatively compared with the results already published in the literature.
