As the key producer of cerebrospinal fluid（CSF）,the choroid plexus（CP） provides a unique protective system in the central nervous *** components are not invariable and they can change based on the pathological conditions of the central nervous *** purpose of the present study was to assess the effects of non-traumatic and traumatic CSF on the differentiation of multipotent stem-like cells of CP into the neural and/or glial *** epithelial cells were isolated from adult male rats and treated with human non-traumatic and traumatic *** in m RNA expression of Nestin and microtubule-associated protein（MAP2）,as the specific markers of neurogenesis,and astrocyte marker glial fibrillary acidic protein（GFAP） in cultured CP epithelial cells were evaluated using quantitative real-time *** data revealed that treatment with CSF（non-traumatic and traumatic） led to increase in m RNA expression levels of MAP2 and ***,the expression of Nestin decreased in CP epithelial cells treated with non-traumatic CSF,while treatment with traumatic CSF significantly increased its m RNA level compared to the cells cultured only in DMEM/F12 as *** seems that CP epithelial cells contain multipotent stem-like cells which are inducible under pathological conditions including exposure to traumatic CSF because of its compositions.

Multicellular microtissues of primary human hepatocytes(PHHs)co-cultured with other supporting cell types are a promis-ing model for drug screening and toxicological ***,these liver microtissues(LMs)rapidly lose their functions during ex vivo ***,in order to mimic the cellular and structural hepatic microenvironment,we co-cultured PHHs with human mesenchymal stromal cells(MSCs)and human umbilical vein endothelial cells(HUVECs)in the presence of cell-sized microparticles(MPs)derived from liver extracellular matrix(LEMPs).The microwell culture platform enabled biofabrication of size-controlled multicellular microtissues(PHH:HUVEC:MSC=3:2:1)with efficient LEMP incorporation(about 70%at a 2:1 ratio of cells:MP).The biofabricated liver microtissues(BLMs)were cultured ex vivo for 14 days and compared to the cell-only LM in terms of gene and protein expression,functional activity,cytochrome P450(CYP450)enzyme inducibility,and drug *** results supported superior hepatic-related gene expression,functional activity,and polarity for PHH in BLM compared to ***450 enzyme inducibility and dose-responsive sensitivity to toxic drugs were significantly higher in the BLM *** conclusion,microtissue engineering by incorporation of tissue-specific microparticles within a multicellular microtissue can offer some advantages for drug discovery studies and cell transplantation *** the near future,this approach could generate a scalable platform of several functional biofabricated microtissues representing different organs.

Antioxidant activity of Ziziphora tenuoir methanolic extracts and comparison of the essential oil in two stages of growth

Flavonoids, anthocyanins, antioxidant capability, total phenolic, ascorbic acid contents, and essential oils compounds of Ziziphora tenuoir(Lamiaceae) for polar and non-polar subfraction in different stages of growth(pre-flowering and flowering) were evaluated in this work. Antioxidant activities of the samples were determined by three testing systems namely DPPH, β-carotene/linoleic acid, and reducing power assay. In the DPPH system, the highest radical scavenging activity was seen by the nonpolar subfraction of the methanol extract in the flowering stage(IC50 43.17 ± 3.68 μg·mL–1). Large differences in the amount of phenolic compounds, antioxidant activity, anthocyanins, and flavonoids of Z. tenuoir in two stages of growth were detected. The chemical composition of the hydrodistilled essential oil of the aerial parts of Z. tenuoir was analyzed by GC/MS and compared in the two stages of growth. The main constituents of the 28 identified compounds in the oil were found to be pulegone(67.34% vs 59.61%), β-humulene(3.24% vs 3.25%), and limonene(5.06% vs 2.57%) at the flowering and pre-flowering stages, respectively.
