The triggering receptor expressed on myeloid cells-1(TREM-1)is a receptor expressed on innate immune *** promoting the amplification of inflammatory signals that are initially triggered by Toll-like receptors(TLRs),TREM-1 has been characterized as a major player in the pathophysiology of acute and chronic inflammatory diseases,such as septic shock,myocardial infarction,atherosclerosis,and inflammatory bowel ***,the molecular events leading to the activation of TREM-1 in innate immune cells remain ***,we show that TREM-1 is activated by multimerization and that the levels of intracellular Ca 2+release,reactive oxygen species,and cytokine production correlate with the degree of TREM-1 ***-1 activation on primary human monocytes by LPS required a two-step process consisting of upregulation followed by clustering of TREM-1 at the cell surface,in contrast to primary human neutrophils,where LPS induced a rapid cell membrane reorganization of TREM-1,which confirmed that TREM-1 is regulated differently in primary human neutrophils and *** addition,we show that the ectodomain of TREM-1 is able to homooligomerize in a concentration-dependent manner,which suggests that the clustering of TREM-1 on the membrane promotes its *** further show that the adapter protein DAP12 stabilizes TREM-1 surface expression and ***-1 multimerization at the cell surface is also mediated by its endogenous ligand,a conclusion supported by the ability of the TREM-1 inhibitor LR12 to limit TREM-1 *** results provide evidence for ligand-induced,receptor-mediated dimerization of ***,our findings uncover the mechanisms necessary for TREM-1 activation in monocytes and neutrophils.

Expression Patterns of CAPN1 and CAPN8b Genes during Embryogenesis in Xenopus laevis

Calpains are a superfamily of Ca2+-dependent cysteine proteases, implicated in various cellular processes and thus probably necessary in all the stages of cell life. The first extended report of quantification of total RNAs within the developmental stages of Xenopus laevis was described in this study. Decreases of total RNAs were positively associated with waves of apoptotic cell death (onset of gastrulation, and morphogenesis). Using qPCR, the temporal expression pattern of CAPN1 and CPAN8b (XCL-2) were characterized during the Xenopus laevis embryogenesis. Transcripts of the CAPN1 and CAPN8 genes were detectable from gastrula stage and their levels oscillated throughout development. The expression of the CAPN1 (mu/I) gene was observed in earliest stage, indicating a maternal origin, while expression of the CAPN8b gene was detectable after midblastula transition. The levels of the two transcripts then started to rise again obviously as a result of zygotic expression (stage 11). The CAPN1 gene expression was particularly expressed at tailbud stage, while the CAPN8 transcripts were found at gastrula, neurula and tailbud stages. This is the first report of quantification of mRNAs CAPN8b and CAPN1 (mu/I) within the developmental stages of Xenopus laevis by qPCR.

Land Use and Land Cover Change Analysis in Ajei Upland Watershed Community Forest, North West Region, Cameroon

This study was carried out in Ajei upland watershed community forest in the North West Region of Cameroon with the aim to analyze spatial and temporal dynamics of land cover and land use from 1988 to 2018 and to identify and characterize the agents, drivers and pressures of this change. The databases used were made up of 3 Lands at satellite images (5 TM of 1988, 7ETM+ of 2003 and 8OLI of 2018). Field survey, interview and focus group discussion methods were used to identify the activities carried out by the local population and to determine agents, drivers and pressures of land use and land cover change. The Ajei community forest is consisted of four main land cover namely;dense vegetation, sparse or degraded vegetation, savanna and bare soil. Between 1988 and 2018, the proportions of dense vegetation and savanna decreased and that of bare soil and sparse vegetation increased. Analysis showed ongoing deforestation with forest cover loss of about 240 ha in 30 years. For the considered time periods 1988-2003 and 2003-2018, deforestation rates were 1.83% per year and 1.49% per year respectively, signifying on average 8 hectares of forest cover undergoing conversion each year. This lost results from the actions of farmers, cattle grazers and wood extractors who through farming, grazing and lumbering activities pressurize and convert the dense forest cover. Economic motives notably the need to increase household income from a frequent demand for farm and wood products in nearby markets represent the drivers of forest cover change. Controlling grazing activities notably in the dry season to check out the use of fires, community sensitization especially among cattle headmen on the importance of the community forest, reforestation activities through natural regeneration or tree planting are needed in the forest.

Tryptophan:A gut microbiota-derived metabolites regulating inflammation

Inflammatory bowel diseases(IBD), which comprise Crohn's disease and ulcerative colitis, are chronic intestinal disorders with an increased prevalence and incidence over the last decade in many different regions over the world. The etiology of IBD is still not well defined, but evidence suggest that it results from per-turbation of the homeostasis between the intestinal microbiota and the mucosal immune system, with the involvement of both genetic and environmental factors. Genome wide association studies, which involve large-scale genome-wide screening of potential polymorphism, have identified several mutations associated with IBD. Among them, Card9, a gene encoding an adapter molecule involved in innate immune response to fungi(via type C-lectin sensing) through the activation of IL-22 signaling pathway, has been identified as one IBD susceptible genes. Dietary compounds, which represent a source of energy and metabolites for gut bacteria, are also appreciated to be important actors in the etiology of IBD, for example by altering gut microbiota composition and by regulating the generation of short chain fatty acids. A noteworthy study published in the June 2016 issue of Nature Medicine by Lamas and colleagues investigates the interaction between Card9 and the gut microbiota in the generation of the microbiota-derived tryptophan metabolite. This study highlights the role of tryptophan in dampening intestinal inflammation in susceptible hosts.

Determining the Relative Age of Fault Activity through Analyses of Gouge Mineralogy and Geochemistry: A Case Study from Vápenná(Rychleby Mts), Czech Republic

The relative age of fractures can be determined through structural analyses in the field or through the detailed mineralogical (XRD) and chemical analyses (AAS method, volumetric, and gravimetric analysis) of fault gouge in the laboratory. The aim of this work was to compare these approaches. It was hypothesised that the two methods would yield consistent results. The studied faults were located in the Rychleby Mts, part of the Sudetic Marginal Fault Zone. The relative age of the faults was determined in the field through the application of the intersection law. The fault gouges were sampled in a crystalline limestone quarry near the village of Vápenná. The mineralogical composition of the fault gouges has been established by XRD analysis of powder samples and analysis of preferentially oriented clay minerals. From our result, it is clear that these two approaches yielded consistent results with regard to the relative age of the faults.

This work explores ceramic additive manufacturing in combination with liquid metal infiltration for the production of degradable interpenetrating phase magnesium/hydroxyapatite(Mg/HA) composites. Material extrusion additive manufacturing was used to produce stoichiometric,and calcium deficient HA preforms with a well-controlled open pore network, allowing the customization of the topological relationship of the composite. Pure Mg and two different Mg alloys were used to infiltrate the preforms by means of an advanced liquid infiltration method inspired by spark plasma sintering, using a novel die design to avoid the structural collapse of the preform. Complete infiltration was achieved in 8 min, including the time for the Mg melting. The short processing time enabled to restrict the decomposition of HA due to the reducing capacity of liquid Mg. The pure Mg-base composites showed compressive yield strength above pure Mg in cast state. Mg alloy-based composites did not show higher strength than the bare alloys due to grain coarsening, but showed similar mechanical properties than other Mg/HA composites that have significantly higher fraction of metallic phase. The composites showed faster degradation rate under simulated body conditions than the bare metallic component due to the formation of galvanic pairs at microstructural level. Mg dissolved preferentially over HA leaving behind a scaffold after a prolonged degradation period. In turn, the fast production of soluble degradation products caused cell metabolic changes after 24 h of culture with not-diluted material extracts. The topological optimization and reduction of the degradation rate are the topics for future research.

The Advanced Approaches to Nutritional and Breadmaking Quality of Wheat, Barley and Rye Flour

This work is focused on the characterization and rapid analytical determination of cereal flour quality with regard to nutritional and breadmaking quality. Starch, protein and non-starch polysaccharides are the main components of cereals. The content and quality of proteins and content of damaged starch is important because of the technological quality of flours. The high content of high molecular weight proteins is substantial for bread technology especially, while soluble protein fractions and non-starch polysaccharides are important for nutrition. The set of wheat, barley and rye flours and their blends were analyzed and their properties and their qualitative parameters were determined. Principal component analysis （PCA） was used on Fourier transform-infrared （FT-IR） spectra in the 1,200-800 cm1 wavenumber region and significant correlations of various nutritional and breadmaking parameters were observed. Results showed that the FT-IR spectroscopy and PCA can serve for rapid screening and classification of cereal flour quality.

Background: The high doses of zinc oxide(Zn O) administered orally to piglets for the prevention of diarrhea and increase of growth rate can contaminate pig farms and the surrounding environment. Therefore, there is a need to find a replacement of high doses of dietary Zn O with an equally effective alternative. In the present study, the effect of two formulations of zinc phosphate-based nanoparticles(Zn A and Zn C NPs) on growth performance,intestinal microbiota, antioxidant status, and intestinal and liver morphology was evaluated. A total of 100 weaned piglets were randomly divided into 10 equal groups with the base diet(control) or the base diet supplemented with Zn A, Zn C, or Zn O at concentrations 500, 1000, and 2000 mg Zn per kilogram of diet. Supplements were given to animals for 10 days. Fecal samples were collected on day 0, 5, 10 and 20. At the end of the treatment(day 10),three piglets from each group were sacrificed and ***: Comparing to that of control, the significantly higher piglet weight gain was observed in all piglet groups fed with Zn A(P < 0.05). Differences in the total aerobic bacteria and coliform counts in piglet feces after NPs supplementation compared to that of control and Zn O groups were also found(P < 0.05). The majority of aerobic culturable bacteria from the feces represented Escherichia(28.57–47.62%), Enterococcus(3.85–35.71%), and Streptococcus(3.70–42.31%) spp. A total of 542 Escherichia coli isolates were screened for the virulence genes STa,STb, Stx2, F4, and F18. The substantial occurrence of E. coli virulence factors was found on day 5, mainly in fimbrillary antigen and thermostable toxins, except for piglets fed by Zn C. Zn treatment decreased Zn blood levels in piglets fed with Zn O and Zn A(500 mg/kg) and increased in Zn C(2000 mg/kg) compared to that of control(P < 0.05).
