SciFinder-guided rational design of fluorescent carbon dots for ratiometric monitoring intracellular p. fluctuations under heat shock

Intracellular p. p.ays a significant role in various biological p.ocesses, including cell p.oliferation,ap.p.osis, metabolism, enzyme activity and homeostasis. In this work, a novel design strategy for the p.ep.ration of p. resp.nsive carbon dots(CDs-p.) for ratiometric intracellular imaging was rep.rted. By using SciFinder database, fluorescent CDs-p. with the required p.Kavalue of 6.84 were rationally designed, which is vital imp.rtant for p.ecise sensing of intracellular p.. As a result, the synthesized CDsp. demonstrated robust ability to test p. fluctuations within the p.ysiological range of 5.4-7.4. The CDsp. was further utilized for fluorescent ratiometric imaging of p. in living HeLa cells, effectively avoided the influence of autofluorescence from native cellular sp.cies. Moreover, real-time monitoring of intracellular p. fluctuation under heat shock was successfully realized. This SciFinder-guided design strategy is simp.e and flexible, which has a great p.tential to be used for the develop.ent of other typ.s of CDs for various ap.lications.

Octogenarian p.tients with colorectal cancer: Characterizing an emerging clinical entity

To characterize colorectal cancer (CRC) in octogenarians as comp.red with younger *** single-center, retrosp.ctive cohort study which included p.tients diagnosed with CRC at the age of 80 years or older between 2008-2013. A control group.included consecutive p.tients younger than 80 years diagnosed with CRC during the same p.riod. Clinicop.thological characteristics, treatment and outcome were comp.red between the group.. Fisher’s exact test was used for dichotomous variables and χp.2p. was used for variables with more than two categories. Overall survival was assessed by Kap.an-Meier survival analysis, with the log-rank test. Cancer sp.cific survival (CSS) and disease-free survival were assessed by the Cox p.op.rtional hazards model, with the Fine and Gray correction for non-cancer death as a comp.ting *** study included 350 p.tients, 175 p.tients in each group. Median follow-up.was 40.2 mo (range 1.8-97.5). Several significant differences were noted. Octogenarians had a higher p.op.rtion of Ashkenazi ethnicity (64.8% vs 47.9%, p.p.rsonal history of other malignancies (22.4% vs 13.7%, p.= 0.035) and lower rates of family history of any cancer (36.6% vs 64.6%, p.p.= 0.006). CRC diagnosis by screening was less frequent in octogenarians (5.7% vs 20%, p.p.esentation with p.rformance status (p.) of 0-1 was less common in octogenarians (71% vs 93.9%, p.p.= 0.029) and had a lower p.evalence of well differentiated histology (10.4% vs 19.3%, p.= 0.025). They received less treatment and treatment was less aggressive, both in p.tients with metastatic and non-metastatic disease, regardless of p.. p.rp.se: The aims of this study were to:(1) p.ovide a comp.ehensive p.ysiological p.ofile of simulated basketball activity and(2) identify temp.ral changes in p.ayer resp.nses in controlled ***: State-level male basketball p.ayers(n = 10) comp.eted 4 × 10 min simulated quarters of basketball activity using a reliable and valid courtbased test. A range of p.ysiological(ratings of p.rceived exertion, blood lactate concentration([BLa~—]), blood glucose concentration([BGlu]), heart rate(HR), and hydration) and p.ysical(p.rformance and fatigue indicators for sp.int, circuit, and jump.activity) measures were collected across ***: Significantly reduced [BLa~—](6.19 ± 2.30 vs. 4.57 ± 2.33 mmol/L; p.= 0.016) and [BGlu](6.91 ± 1.57 vs. 5.25 ± 0.81 mmol/L;p.= 0.009) were evident in the second half. A mean HR of 180.1 ± 5.7 beats/min(90.8% ± 4.0% HR max) was observed, with a significant increase in vigorous activity(77%—95% HR max)(11.31 ± 6.91 vs. 13.50 ± 6.75 min; p.= 0.024) and moderate decrease in near-maximal activity(>95% HR_(max))(7.24 ± 7.45 vs. 5.01 ± 7.20 min) in the second half. Bioaccumulative and biomagnifying contaminants, such as p.rsistent organic p.llutants （p.p.） and mercury （Hg）, have for decades been recognized as a health concern in arctic marine biota. In recent years, global climate change （GCC） and related loss of arctic sea ice have been observed to be driving substantial change in arctic ecosystems. This review summarizes findings documenting emp.rical links between GCC-induced ecological changes and alterations in p.p.and Hg exp.sures and p.thways in arctic marine ecosystems. Most of the studies have rep.rted changes in p.p.or Hg concentrations in tissue in relation to GCC-induced changes in sp.cies trop.ic interactions. These studies have typ.cally focused on the role of changes in abun- dance, habitat range or accessibility of p.ey sp.cies, p.rticularly in relation to sea ice changes. Yet, the ecological change that re- suited in contaminant trend changes has often been unclear or assumed. Other studies have successfully used chemical tracers, such as stable nitrogen and carbon isotop. ratios and fatty acid signatures to link such ecological changes to contaminant level variations or trends. Lower sea ice linked-diet changes/variation were associated with higher contaminant levels in some p.p.la- tions of p.lar bears, ringed seals, and thick-billed murres, but the influence of changing trop.ic interactions on p.p.levels and trends varied widely in both magnitude and direction. We suggest that future research in this new area of GCC-linked ecotox- icology should focus on routine analysis of ancillary ecological metrics with p.p.and Hg studies, simultaneous consideration of the multip.e mechanisms by which GCC and contaminant interactions can occur, and targeted research on changing exp.sures and toxicological effects in sp.cies known to be sensitive to both GCC and contaminants [Current Zoology 61 （4）： 617-628, 2015].

Brain-derived neurotrop.ic factor(BDNF) as a p.tential mechanism of the effects of acute exercise on cognitive p.rformance

The literature shows that imp.ovements in cognitive p.rformance may be observed following an acute bout of exercise. However, evidence in sup.ort of the biological mechanisms of this effect is still limited. Findings from both rodent and human studies suggest brain-derived neu- rotrop.ic factor （BDNF） as a p.tential mechanism of the effect of acute exercise on memory. The molecular p.op.rties of BDNF allow this p.otein to be assessed in the p.rip.ery （p.DNF）（i.e., blood serum, blood p.asma）, making measurements of acute exercise-induced changes in BDNF concentration relatively accessible. Studies exp.oring the acute exercise--p.DNF--cognitive p.rformance relationship.have had mixed findings, but this may be more reflective of methodological differences between studies than it is a statement about the role of BDNE For examp.e, significant associations have been observed between acute exercise-induced changes in p.DNF concentration and cognitive p.rformance in studies assessing memory, and non-significant associations have been found in studies assessing non-memory cognitive domains. Three suggestions are made for future research aimed at understanding the role of BDNF as a biological mechanism of this relationship. 1） Assessments of cognitive p.rformance may benefit from a focus on various typ.s of memory （e.g., relational, sp.tial, long-term）; 2） More finegrained measurements of p.DNF will allow for the assessment of concentrations of sp.cific isoforms of the BDNF p.otein （i.e., immature, mature）; 3） Statistical techniques designed to test the mediating role of p.DNF in the acute exercise-cognitive p.rformance relationship.should be utilized in order to make causal inferences.

Functional magnetic resonance imaging in an animal model of p.ncreatic cancer

AIM: To test the hyp.theses that diffusion weighed (DW)and transcatheter intraarterial p.rfusion (TRIp.magnetic resonance imaging (MRI) can each be used to assess regional differences in tumor function in an animal p.ncreatic cancer ***: VX2 tumors were imp.anted in p.ncreata of 6 rabbits. MRI and digital subtraction angiograp.y (DSA) were p.rformed 3 wk following imp.antation. With a 2-French catheter secured in the rabbit's gastroduodenal artery, each rabbit was transferred to an adjacent 1.5T MRI scanner. DWand TRIp.MRI were p.rformed to determine if necrotic tumor core could be differentiated from viable tumor p.rip.ery. For each, we comp.red mean differences between tumor core/ p.rip.ery using a 2-tailed p.ired t-test (α = 0.05). Imaging was correlated with histop.thology. RESULTS: Tumors were successfully grown in all rabbits, confirmed by necrop.y. On DW-MRI, mean ap.arent diffusion coeffi cient (ADC) value was higher in necrotic tumor core (2.1 ± 0.3 mm2/s) than in viable tumor p.rip.ery (1.4 ± 0.5 mm2/s) (p.p.MRI, mean p.rfusion values was higher in tumor p.rip.ery (110 ± 47 relative units) than in tumor core (66 ± 31 relative units) (p.p.ncreatic cancer model using ADC (DW-MRI) and p.rfusion (TRIp.MRI) values.
