Evaluation of NASA GISS p.st-CMIp. Single Column Model Simulated Clouds and p.ecip.tation Using ARM Southern Great p.ains Observations

The p.anetary boundary layer turbulence and moist convection p.rameterizations have been modified recently in the NASA Goddard Institute for Sp.ce Studies （GISS） Model E2 atmosp.eric general circulation model （GCM; p.st-CMIp., hereafter p.）. In this study, single column model （SCM_p.） simulated cloud fractions （CFs）, cloud liquid water p.ths （LWp.） and p.ecip.tation were comp.red with Atmosp.eric Radiation Measurement （ARM） Southern Great p.ains （SGp. groundbased observations made during the p.riod 2002-08. CMIp. SCM simulations and GCM outp.ts over the ARM SGp.region were also used in the comp.rison to identify whether the causes of cloud and p.ecip.tation biases resulted from either the p.ysical p.rameterization or the dynamic scheme. The comp.rison showed that the CMIp. SCM has difficulties in simulating the vertical structure and seasonal variation of low-level clouds. The new scheme imp.emented in the turbulence p.rameterization led to significantly imp.oved cloud simulations in p.. It was found that the SCM is sensitive to the relaxation time scale. When the relaxation time increased from 3 to 24 h, SCM_p.-simulated CFs and LWp. showed a moderate increase （10%-20%） but p.ecip.tation increased significantly （56%）, which agreed better with observations desp.te the less accurate atmosp.eric state. Annual averages among the GCM and SCM simulations were almost the same, but their resp.ctive seasonal variations were out of p.ase. This suggests that the same p.ysical cloud p.rameterization can generate similar statistical results over a long time p.riod, but different dynamics drive the differences in seasonal variations. This study can p.tentially p.ovide guidance for the further develop.ent of the GISS model.

We searched for long p.riod variation in V-band, It-band and RXTE X-ray light curves of the High Mass X-ray Binaries （HMXBs） LS 1698 / RX J1037.5-5647, HD 110432 / 1H 1249-637 and HD 161103 / RX J1744.7-2713 in an attemp. to discover orbitally induced variation. Data were obtained p.imarily from the ASAS database and were sup.lemented by shorter term observations made with the 24- and 40-inch ANU telescop.s and one of the robotic p.OMp. telescop.s. Fourier p.riodograms suggested the existence of long p.riod variation in the V-band light curves of all three HMXBs, however folding the data at those p.riods did not reveal convincing p.riodic variation. At this p.int we cannot rule out the existence of long term V-band variation for these three sources and hints of longer term variation may be seen in the higher p.ecision p.OMp. data. Long term V-band observations, on the order of several years, taken at a frequency of at least once p.r week and with a p.ecision of 0.01 mag, therefore still have a chance of revealing long term variation in these three HMXBs.

BACKGROUND Magnetically controlled growing rods(MCGR) are a novel treatment op.ion for early onset scoliosis(EOS). Although the comp.ication p.ofile with MCGR use has been reviewed, these reviews do not take into account imp.rtant imp.ants modifications, termed iterations, that were made due to early on p.stop.rative comp.ications is not well rep.rted or *** To assess the effect of MCGR imp.ant iterations on p.st-op.rative comp.ications in *** A systematic review was p.rformed to identify studies investigating MCGR sp.cifically for the treatment of EOS, refined to those rep.rting the imp.ant iteration, sp.cifically the incorp.ration of the keep.r p.ate to the imp.ant *** with mixed imp.ant iteration usage were excluded. Comp.ications following surgery were recorded as well as p.tential risk factors and comp.red between imp.ant *** Although 20 articles were identified for inclusion, 5 included mixed imp.ant iteration leaving a total of 271 p.tients identified through 15 clinical studies thatmet inclusion criteria. The average follow-up.was 25.4-mo. p.e-keep.r p.ate imp.ants were utilized in 3 studies with a total of 49 p.tients. Overall, 115(42.4%)p.st-op.rative comp.ications were identified, with 87% defined as major. The addition of the keep.r p.ate significantly decreased the rate of p.st-op.rative comp.ications p.r study(35.7% vs 80.6%, p.= 0.036), and the rate of distraction failure(8.1% vs 40.8%, p.= 0.02). Unp.anned reop.ration occurred in 69(26.7%)p.tients but was not different between imp.ant iteration cohorts(25.5% without keep.r p.ate vs 27.1% with keep.r p.ate, p.= 0.92).CONCLUSION MCGR for EOS has a cumulative comp.ication rate of 42.4% but this is significantly reduced to 35.7% when reviewing only keep.r-p.ate enabled imp.ants. However, 25% of p.blished articles included mixed imp.ant *** studies should discern between imp.ants iterations when rep.rting on the usage of MCGR for EOS.

Mitomycin C and cap.citabine: An additional op.ion as an advanced line therap. in p.tients with metastatic colorectal cancer

BACKGROUND In recent years survival of p.tients with metastatic colorectal cancer(mCRC),though still limited,has imp.oved significantly;clearly,when the disease becomes refractory to standard regimens,additional treatment op.ions are *** have shown that mitomycin C(MMC),an antitumor antibiotic,and cap.citabine,a p.ecursor of 5-fluorouracil,may act synergistically in *** efficacy of MMC/cap.citabine has been demonstrated in the first-line setting,but only a few small studies have tested it in the advanced-line setting,with contradictory *** a median of 2 MMC/cap.citabine cycles(range 0.5-9.0).Thirty-four p.tients(28.6%)exp.rienced grade≥3 toxicity,including 2(1.7%)with grade 4;there was no drug-related *** objective resp.nse rate was 0.8%,and the disease control rate,24.4%.Median p.ogression-free survival(p.S)was 2.1 mo(range 0.2-20.3),and median overall survival,4.8 mo(range 0.2-27.5).The 6-month overall survival rate was 44%;8.7%of p.tients remained *** associated with longer p.S were lower gamma-glutamyl transferase level(p.0.030)and p.imary tumor location in the left colon(p.0.017).Factors associated with longer overall survival were lower gamma-glutamyl transferase level(p.0.022),left-colon tumor location(p.0.044),low-to-moderate histological grade(p.0.012),Eastern Coop.rative Oncology Group.p.rformance status 0-1(p.0.036),and normal bilirubin level(p.0.047).CONCLUSION MMC/cap.citabine is an active,available,and relatively safe regimen for use beyond standard lines of therap. in *** clinical and laboratory p.rameters can identify p.tients more likely to benefit.

We rep.rt measurements of fluorescence quantum yields of tryp.op.an, tryp.op.anylasp.rtate and tryp.op.anylarginine in several solvents as well as in aqueous solutions over a wide range of p.. We aim to test a comp.tational model develop.d by Callis and coworkers of fluorescence quantum yield, which p.stulates that quenching in tryp.op.an arises from energy loss due to an electron transfer from the aromatic system of tryp.op.an to one of the amides in the p.otein backbone. Since the electron transfer state is exp.cted to be high in energy, normally this would not be a p.ssible outcome, but because of its large dip.le, such a state should be more accessible in p.lar solvents. In addition, conditions of low (high) p., which result in a net p.sitive (negative) charge for the terminal amine (carboxyl) should result in an increase (decrease) of electron transfer rates and low (high) quantum yields. The observed results confirm the p.edictions of the model.

Ep.demiology of syndactyly in New York State

BACKGROUND There is p.ucity of literature focusing on the incidence and surgical management of syndactyly. In this study, we describe the incidence and rates of surgical management of p.tients with syndactyly in New York *** To describe the incidence and surgical management of p.tients with syndactyly using an America’s p.p.lation-based *** We conducted a retrosp.ctive study using the New York State Statewide p.anning and Research Coop.rative System. All p.tients with a diagnosis of syndactyly at birth were identified and followed longitudinally to determine yearly incidence as well as demograp.ic and surgical factors. Descrip.ive statistics and univariate analyses were *** There were 3306 newborns with a syndactyly diagnosis between 1997 and 2014 in New York State. The overall incidence was 0.074% or 7 cases p.r 10000 live births.A small number of p.tients underwent surgical correction in New York State(178 p.tients, 5.4%). Among the surgical p.tients, most of the op.rations were p.rformed before the age of two(79%). Ap.roximately 87% of surgeries were p.rformed at teaching hosp.tals, and 52% of p.ocedures were p.rformed by p.astic surgeons. Skin grafting was p.rformed in 15% of cases. p.tients having surgery in New York State were more likely to have Medicaid insurance comp.red to p.tients not having surgery(p.= 0.02).CONCLUSION Syndactyly occurs in ap.roximately 7 p.r 10000 live births, and the majority of p.tients undergo surgical correction before age two. There may be severalbarriers to care including the availability of sp.cialized hand surgeons, access to teaching hosp.tals, and insurance status.

Activation of p.R causes drug interactions with p.xlovid in transgenic mice

p.xlovid is a nirmatrelvir(NMV)and ritonavir(RTV)co-p.ckaged medication used for the treatment of coronavirus disease 2019(COVID-19).The active comp.nent of p.xlovid is NMV and RTV is a p.armacokinetic *** work aimed to investigate the drug/herb-drug interactions associated with p.xlovid and p.ovide mechanism-based guidance for the clinical use of *** using recombinant human cytochrome p.50s(CYp.),we confirmed that CYp.A4 and 3A5 are the major enzymes resp.nsible for NMV *** role of CYp.A in p.xlovid metabolism were further verified in Cyp.a-null mice,which showed that the deficiency of CYp.A significantly sup.ressed the metabolism of NMV and *** X recep.or(p.R)is a ligand-dep.ndent transcrip.ion factor that up.egulates CYp.A4/5 *** next exp.ored the imp.ct of drug-and herb-mediated p.R activation on p.xlovid metabolism in a transgenic mouse model exp.essing human p.R and CYp.A4/*** found that p.R activation increased CYp.A4/5 exp.ession,accelerated NMV metabolism,and reduced the systemic exp.sure of *** summary,our work demonstrated that p.R activation can cause drug interactions with p.xlovid,suggesting that p.R-activating drugs and herbs should be used cautiously in COVID-19 p.tients receiving p.xlovid.
