Phylogenetic relationships within the genus Aspidisca(Protozoa,Ciliophora,Euplotida) revealed by ITs1-5.8s-ITs2 region sequences

Abstract The internal transcribed spacer regions （ITs1 and ITs2） and 5.8s rRNA genes were sequenced in six populations of four Aspidisca species, namely A. leptaspis, A. orthopogon, A. rnagna and A. aculeata. Phylogenetic trees were constructed by means of Bayesian inference （BI）, Maximum Parsimony （MP）, Neighbor-Joining （NJ）, and Maximum Likelihood （ML） to assess the inter- and intra-species relationships within the genus Aspidisca. all trees show similar topologies with stable supports and indicate that：（1） four well known groups, i.e., Oligotrichia, stichotrichia, Choreotrichia and Hypotrichia, are distinctly outlined within the class spirotrichea, and all are monophyletic other than Hypotrichia; （2） members of Aspidisca can be distinguished well, based on the ***-ITs2 region sequences, and A. leptaspis and A. magna shared a closer relationship than other species; （3） Aspidisca and Euplotes branch early in the subclass Hypotrichia. To compare the phylogenetic relationships based on different genes, ssU rRNA trees were also constructed with nearly the same species inclusion, which revealed different topologies of inter-species, inter-genera and inter-subclasses.

Phylogenetic and taxonomic studies on ciliate protists using molecular approaches have been demonstrated to be very reliable to form strong conclusions and results. In the present work, species separation of some morphologically similar stichotrichous ciliates, two species of Pseudokeronopsis and two species of Apokeronopsis, was reexamined using amplified ribosomal DNA restriction analysis (PCR-RFLP). Five of 10 restriction enzymes revealed species-specific polymorphic patterns, of which four similar stichotrichs could be significantly separated and identified. Among them, EcoR I offered almost no significantly different restriction fragment patterns, but the four species could be separated from one another and identified with Hae Ⅲ. Distinctly different restriction digestion haplotypes and simi-larity indices separated the species, and were used to construct a phylogeny. Phylogenies based on ITs2 nucleotide sequences and ITs2 secondary structures supported the separation of Pseudokeronopsis and Apokeronopsis using RFLP analysis, although three Pseudoker-onopsis carnea populations did not cluster together. In addition, phylogenetic analyses using multiple algorithms confirmed that these two genera formed two distinct groups within the urostylids.

Coating with viscous formulations has been essential in numerous industries as it can be a means for providing functionalization, additional properties, as well as other benefits. However, there have been scarce studies that have investigated and proposed methodologies in literature. Continuous coating of powders with viscous liquids poses as a promising technology, which has been mentioned in some studies, but has not yet been thoroughly investigated. This paper employs the use of image processing and analysis, in combination with statistical analysis of particles to evaluate the effectiveness of foams and liquids as a means of coating powder beds. Two different sizes of twin screw mixers that are working in continuous operation are employed, and a new continuous foaming device is fabricated and used for the experiments of coating. The effect of materials and process parameters (as for example rotational speed, and flowrate) on the quality of coating are investigated. Image analysis is used to assess the coating quality. The results clearly showcase the potential of using twin screw mixers for coating purposes and not only for mixing. The hypothesis that using large bubble foams to improve the coating of viscous liquids on particles is proven correct, as they provide higher quality coatings compared to their equivalent liquids, when used in the twin screw mixer. surprisingly, using a larger scale twin screw mixer, does not show a substantial effect on the mixing, regarding quality, however there is still a requirement for mix optimization for achieving scale-up of this process. These results provide a new pathway for coating powders with viscous formulations in industrial applications, requiring less energy and effort in this process, and can pave the way towards introducing more sustainable industrial methodologies for coating.

The characteristics and infraciliature of a new euplotid ciliate, Euplotes qingdaoensis n. sp., discovered from the coastal waters of Jiaozhou Bay, northern China, were studied using live observation and silver impregnation. The new marine species is characterized by a slightly flattened body （90-110 um×70- 80 um, in vivo）, an adoral zone of 26-29 membranelles, 10 strong frontoventral and five transverse cirri, two caudal cirri, a single left marginal cirrus, seven dorsal kineties, a middle kinety with about 15 pairs of basal bodies, a silverline system of the double-eurystomus type, and a C-shaped macronucleus.

In this study,a comprehensive analysis of jets and underlying events as a function of charged particle multiplicity in proton-proton(pp)collisions at a center-of-mass energy of √s=7 TeV is *** Monte Carlo(MC)event generators,including Pythia8.308,EPOs 1.99,EPOsLHC,EPOs4_(Hydro),and EPOs4_(noHydro),are employed to predict particle *** predictions from these models are compared with experimental data from the CMs *** charged particles are categorized into those associated with underlying events and those linked to jets,and the analysis is restricted to charged particles with|η|0.25 GeV/*** comparing the MC predictions with CMs data,we find that EPOs4_(Hydro),EPOsLHC,and Pythia8 consistently reproduce the experimental results for all charged particles,underlying events,intrajets,and leading charged *** charged jet rates with p_T^(***)>5 GeV/c,EPOs4_(Hydro)and Pythia8 perform exceptionally *** the case of charged jet rates with p_T^(***)→30 GeV/c,EPOsLHC reproduces satisfactorily good results,whereas EPOs4 Hydro exhibits good agreement with the data at higher charged particle multiplicities compared to the other *** can be attributed to the conversion of energy into flow when"Hydro=on"leading to an increase in *** EPOsLHC model describes the data better owing to the new collective flow effects,correlated flow treatment,and parameterization compared to EPOs ***,the examination of the jet p_T spectrum and normalized charged p_T density reveals that EPOs4_(Hydro),EPOs4_(noHydro),and EPOsLHC exhibit good agreement with the experimental results,whereas Pythia8 and EPOs 1.99 do not perform as well owing to the lack of correlated flow treatment.
