Purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RPE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from Polycaprolactone (PCL), and different concentrations of gelatin (5%, 15% and 30%). ARPE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARPE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and ZO-1) qRT-PCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPE-19 cells grew on PCL/Gelatin membranes mainly in PCL/30% gelatin combination, which had not cytotoxic effect. RPE65/cytokeratin-18/ and actin-positive ARPE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPE-19 cells fused among, suggesting the presence of tight junctions. The expression of RPE65 and ZO-1 was unchanged. Conclusions: PCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and exhibited RPE-like morphology. Despite being within the intertropical region of the planet,the Mexican territory still has glacier-covered *** recent decades,important advances have been made in studies on glaciology and periglacial environment in Mexico both for current and past ***,in spite of Cofre de Perote volcano(4200 m a.s.l.)being a strategically located mountain,it has not yet been studied in regards to the glacial and periglacial processes;in fact,those dynamics have modified the mountain massifs in the *** complement the series of studies on glacial history in the high mountain environment of México,this study reconstructs the glacial cover and the periglacial environment of the volcano surface during the final stage of the Late Pleistocene based on climatic retrospective and through the identification of geomorphological *** findings indicate the existence of a large glacier(ice cap)that covered the northern,western,and southern slopes of the mountain;while in the eastern sector there were two small glaciers,one being of cirque type,and the other of valley *** current temperature conditions prevent the occurrence of permanent ice bodies;at the same time,it was found that the periglacial blockfields of the slopes is a legacy of the climatic conditions that prevailed at the end of Late Pleistocene.

Ultraviolet radiation by its wavelength is divided into: UVA, UVB and UVC. Only UVA and UVB manage to penetrate the ozone layer, but due to anthropological activities, all of them are capable of interacting with humans to a greater or lesser extent, and can generate adverse effects such as cellular stress when interacting with intra-and extracellular biomolecules. The skin is the first organ in contact with UV radiation, and the stress it generates can be analyzed by the expression of a bioindicator of cellular damage such as Hsp70. Therefore, the objective of the project was: to determine the effect of UVA, UVB and UVC radiation on HaCaT epithelial cells, by analyzing the expression of Hsp70. Materials and methods: HaCaT cells were cultured in vitro, which were irradiated with UVA, UVB and UVC light at different doses, to subsequently determine the degree of Hsp70 expression by Immunodetection by PAGE-SDS and Western Blot. Results: Basal expression of Hsp70 was observed in no irradiated HaCaT cells. When HaCaT cells were irradiated with UVA, UVB, UVC, an increase in this Hsp70 protein was observed. With UVA, a higher degree of expression was observed at a time of 30 minutes of irradiation. With UVB the highest expression shifted to a time of 20 minutes. With UVC, overexpression was observed after 10 minutes. Conclusion: UV radiation generates cellular stress on HaCaT cells, evaluated by the stress bioindicator Hsp70. According to the wavelength of UV radiation, those that have a shorter wavelength have a greater potential for cellular damage, such as UVC.

Every dam or barrage construction affects the watercourse and the retention of sediment that previously was carried by the river, which can lead to siltation of the reservoir and obstruction of water intakes over time, reducing their capacities. However, the information available regarding the effect of sediment and drawdown parameters, sediment management at reservoirs, as well as different equational approaches,is scarce. The current research aims to evaluate the effect of parameters associated with numerical modeling of sediment management in reservoirs considering scenarios with different drawdowns,transport equations, sediment size distributions, and thickness of the initial sediment layer. The case study of the Aimore s Hydropower Plant(HPP) is used, applying the Delft3D-FLOW model for twodimensional modeling. All parameters influenced the volume of mobilized sediment, among which the initial layer thickness was the parameter that resulted in the greatest changes in simulated results. In general, the results show that the uncertainties in the input parameters outweigh the uncertainties between the techniques, which found large variations in results when evaluating the use of different transport equations. These results indicate the importance of proper estimation of model parameters for predicting effects with accuracy and the need for such studies before planning and management operations are evaluated to avoid environmental harm and energy waste.

目的:研究长链非编码RNA(LncRNA)LUCAT1调控miR-375/高迁移率族蛋白-1(HMGB1)轴对多囊卵巢综合征(PCOS)大鼠胰岛素抵抗的相关分子机制,为疾病的临床干预提供新靶点。方法:选择SD雌性大鼠3周龄(平均体重50 g)20只,随机分为对照组和模型组各10只,模型组采用皮下注射脱氢表雄酮(DHEA)的方法复制PCOS模型。qRT-PCR法检测卵巢组织LncRNA LUCAT1和miR-375表达量,Western blot法检测HMGB1蛋白,常规生化法检测血清空腹血糖(FBG)和空腹胰岛素(FINS),计算胰岛素抵抗指数(HOMA-IR)。随后分离卵巢颗粒细胞(GCs)并采用免疫荧光法检测卵泡刺激素受体(FSHR)进行细胞鉴定。体外构建si-LUCAT1和si-NC并转染GCs,培养48h后检测LUCAT1、miR-375和HMGB1表达量。结果:与对照组大鼠相比,模型组卵巢组织LncRNA LUCAT1和HMGB1蛋白表达量显著升高,而miR-375表达量显著下降(P<0.05);血清FBG和FINS水平以及HOMA-IR值明显升高(P<0.05)。与si-NC组GCs细胞相比,si-LUCAT1组LUCAT1和HMGB1表达量显著下降,而miR-375表达量增加(P<0.05)。结论:LncRNA LUCAT1可能通过调控miR-375/HMGB1轴并参与PCOS的发生以及胰岛素抵抗。

Objective: To evaluate transperineal laser ablation (TPLA) with Echolaser® (Echolaser® TPLA, Elesta S.p.A., Calenzano, Italy) as a treatment for benign prostatic hyperplasia (BPH) and prostate cancer (PCa) using the Delphi consensus ***: Italian and international experts on BPH and PCa participated in a collaborative consensus project. During two rounds, they expressed their opinions on Echolaser® TPLA for the treatment of BPH and PCa answering online questionnaires on indications, methodology, and potential complications of this technology. Level of agreement or disagreement to reach consensus was set at 75%. If the consensus was not achieved, questions were modified after each round. A final round was performed during an online meeting, in which results were discussed and ***: Thirty-two out of forty invited experts participated and consensus was reached on all topics. Agreement was achieved on recommending Echolaser® TPLA as a treatment of BPH in patients with ample range of prostate volume, from 80 mL (80%), comorbidities (100%), antiplatelet or anticoagulant treatment (96%), indwelling catheter (77%), and strong will of preserving ejaculatory function (100%). Majority of respondents agreed that Echolaser® TPLA is a potential option for the treatment of localized PCa (78%) and recommended it for low-risk PCa (90%). During the final round, experts concluded that it can be used for intermediate-risk PCa and it should be proposed as an effective alternative to radical prostatectomy for patients with strong will of avoiding urinary incontinence and sexual dysfunction. Almost all participants agreed that the transperineal approach of this organ-sparing technique is safer than transrectal and transurethral approaches typical of other techniques (97% of agreement among experts). Large quantities of natural aggregates(NA)are required in the manufacturing of asphalt mixtures for *** extraction of NA generates high environmental impacts that negatively affect environmental preservation and *** alternative materials obtained in construction and demolition processes in civil construction worked have been studies as possible replacements for NA,with the purpose of reducing these environmental *** foregoing study presents a state of knowledge review referring to the use of recycled concrete aggregates(RCA)when these are used in the manufacturing of asphalt mixtures(mainly hot-mix asphalt HMA type).Environmental aspects are presented,as well as possible benefits and limitations of using RCA as replacements for NA in asphalt ***,it is not possible to establish a behavior pattern,since the characteristics of the RCA are very heterogeneous and vary from the original source of the *** is highlighted from the literature that RCA has a wide potential for use in the manufacture of asphalt mixtures,since in most studies,the mixtures with these materials comply the quality requirements contemplated by the construction specifications,mainly in low-volume ***,based on the literature review,some recommendations and suggestions are presented for future research.

This letter focuses on a recently published article that provided an exceptional description of the effect of epigenetic modifications on gene expression patterns related to skeletal system ***,it discusses a novel modality of epigenetic regulation,the long noncoding RNAs(lncRNAs),and provides evidence of their involvement in mesenchymal stromal/stem cells osteo-/adipogenic differentiation *** focus on lncRNAs,there is an emerging cross talk between lncRNAs and miRNAs interaction as a novel mechanism in the regulation of the function of the musculoskeletal system,by controlling bone homeostasis and bone regeneration,as well as the osteogenic differentiation of stem ***,we touched on some examples to demonstrate this *** addition,we believe there is still much to discover from the effects of lncRNAs on progenitor and non-progenitor cell *** incorporated data from other published articles to review lncRNAs in normal progenitor cell osteogenic differentiation,determined lncRNAs involved in osteoarthritis pathogenesis in progenitor cells,and provided a review of lncRNAs in non-progenitor cells that are differentially regulated in *** conclusion,we really enjoyed reading this article and with this information we hope to further our understanding of lncRNAs and mesenchymal stromal/stem cells regulation.
