Adipose browning has demonstrated therapeutic potentials in several ***,by conducting transcriptomic profiling at the single-cell and single-nucleus resolution,we reconstituted the cellular atlas in mouse inguinal subcutaneous white adipose tissue(iWAT)at thermoneutrality or chronic cold *** major nonimmune cells within the iWAT,including adipose stem and progenitor cells(ASpCs),mature adipocytes,endothelial cells,Schwann cells,and smooth muscle cells,were recovered,allowing us to uncover an overall and detailed blueprint for transcriptomes and intercellular cross-talks and the dynamics during white adipose tissue brown *** findings also unravel the existence of subpopulations in mature adipocytes,ASpCs,and endothelial cells,as well as new insights on their interconversion and reprogramming in response to *** adipocyte subpopulation competent of major histocompatibility complex class Ⅱ(MHCⅡ)antigen presentation is ***,a subcluster of ASpC with CD74 expression was identified as the precursor of this MHCⅡ^(+)*** adipocytes are transdifferented from preexisting lipid generating adipocytes,which exhibit developmental trajectory from de novo differentiation of amphiregulin cells(Aregs).Two distinct immune-like endothelial subpopulations are present in iWAT and are responsive to *** data reveal fundamental changes during cold-evoked adipose browning.

Expression of alpha-fetoprotein messenger rna in BEL-7404 human hepatoma cells and effect of L-4-oxalysine on the expression

AIM To investigate alpha-fetoprotein (AFp) mrna expression in BEL-7404 human hepatoma cells and the effect of L-4-oxalysine (OXL) on the *** BEl-7404 human hepatoma cells were maintained in RpMI 1640 media. Human AFp cDNA probe was labelled with digoxigenin-11-dUTp by the random primer labelling method. The expression of AFp mrna in Bel-7404 cells was determined by an in situ hybridization technique with digoxigenin-labelled human AFp cDNA probe. The positive intensities of AFp mrna in cells were analyzed by microspectrophotometer and expressed as absorbance at 470nm. For the experiment with OXL, cells were incubated with various concentrations of the agent for *** Essentially all the hepatoma cells contained AFp mrna in the cytoplasm, although in various amounts. The specificity of the hybridization reaction was confirmed by control experiments in which the use of rnase-treated BEL-7404 cells, non-AFp-producing cells (HL-60 human leukemia cells) or a nonspecific cDNA probe resulted in negative hybridization. When the cells were treated with OXL (25, 50mg/L), the content of AFp mrna in the cytoplasm was decreased with the inhibition percentages of 34.3% and 70.1%, respectively (p＜0.05).CONCLUSION AFp mrna was expressed in BEL-7404 human hepatoma cells and OXL suppressed AFp mrna expression in the cells.

目的探究丙型肝炎病毒核糖核酸(HCV-rna)含量与丙氨酸氨基转移酶(ALT)水平的关系。方法 120例HCV患者的血清标本同时以荧光定量聚合酶链式反应(pCR)法检测HCV-rna及全自动生化分析仪检测ALT水平,对两组关系进行统计分析。结果 120例患者的标本中,82例ALT水平异常,其ALT水平及异常率随rna含量的升高而增加,经相关性分析,ALT水平及异常率与HCVrna含量呈正相关性(r=0.986,prna是反映HCV复制的有效指标,通过检测ALT水平可帮助临床了解HCV在体内的复制状况及肝脏的炎性反应状态。

Introduction:Coronavirus disease 2019(COVID-19)may be associated with various skin *** them,figurate erythema(FE)is rare;only a few clinical reports have been ***,we reported an unusual concentric FE lesions and their immunohistochemical profile in a patient with *** presentation:A 52-year-old woman was diagnosed with *** month later,she developed asymptomatic annular erythematous concentric ring lesions on her inner right *** margin of every ring increased in coloration,and the lesions then completely resolved in 2 weeks.A biopsy revealed vasculitis of small vessels,an increase in the fluorescence intensity of CD3 and CD45,and negativity for COVID-19 ***:The results showed an unusual FE that has a clinical mixed type between erythema annulare centrifugum and erythema gyratum repens;the physiopathological mechanism in the FE due to COVID-19 is unknown;one theory involves rapid viral passage into the cutaneous vascular system that triggers an immune and inflammatory *** the results exhibited a negative COVID-19 rna in the tissue,the CD3^(+)/CD45^(+)lymphocyte infiltration could explain the clinical pattern as a direct inflammatory response in the small vessel or as a delayed immune ***:Our findings suggest an immunohistochemical small vessel vasculitis pattern with an unusual clinical FE that differs from other types of COVID-19-associated FE or erythema reported previously.

Despite the success of antiretroviral therapy,human immunodeficiency virus(HIV)cannot be cured because of a reservoir of latently infected cells that evades *** understand the mechanisms of HIV latency,we employed an integrated single-cell rna sequencing(scrna-seq)and single-cell assay for transposase-accessible chromatin with sequencing(scATAC-seq)approach to simultaneously profile the transcriptomic and epigenomic characteristics of~125,000 latently infected primary CD4^(+)T cells after reactivation using three different latency reversing *** expressed genes and differentially accessible motifs were used to examine transcriptional pathways and transcription factor(TF)activities across the cell *** identified cellular transcripts and TFs whose expression/activity was correlated with viral reactivation and demonstrated that a machine learning model trained on these data was 75%-79%accurate at predicting viral ***,we validated the role of two candidate HIV-regulating factors,FOXp1 and GATA3,in viral *** data demonstrate the power of integrated multimodal single-cell analysis to uncover novel relationships between host cell factors and HIV latency.

Metabolic syndrome and the urinary microbiome of patients undergoing percutaneous nephrolithotomy

Objective:To identify possible stone-promoting microbes,we compared the profiles of microbes grown from stones of patients with and without metabolic syndrome(MetS).The association between MetS and urinary stone disease is well established,but the exact pathophysiologic relationship remains *** evidence suggests urinary tract dysbiosis may lead to increased nephrolithiasis ***:At the time of percutaneous nephrolithotomy,bladder urine and stone fragments were collected from patients with and without *** sample types were subjected to expanded quantitative urine culture(EQUC)and 16 S ribosomal rna gene ***:Fifty-seven patients included 12 controls(21.1%)and 45 MetS patients(78.9%).Both cohorts were similar with respect to demographics and non-MetS *** controls had uric acid stone *** EQUC,bacteria were detected more frequently in MetS stones(42.2%)compared to controls(8.3%)(p=0.041).Bacteria also were more abundant in stones of MetS patients compared to *** validate our EQUC results,we performed 16 S ribosomal rna gene *** 12/16(75.0%)sequence-positive stones,EQUC reliably isolated at least one species of the sequenced *** were detected in both“infectious”and“non-infectious”stone ***:Bacteria are more common and more abundant in MetS stones than control *** findings support a role for bacteria in urinary stone disease for patients with MetS regardless of stone composition.
