Lycorine and galantamine are natural alkaloids found in Amaryllidaceae plants, such as narcissus. Narcissus leaves and roots are sometimes accidentally ingested because they resemble vegetables. Lycorine and galantamine are toxic and cause such effects as nausea, vomiting, and abdominal pain, when accidentally ingested. In a case of narcissus poisoning, the detection of lycorine and galantamine in biological samples is vital to determine whether they have been ingested. This study establishes a liquid chromatography-tandem mass spectrometry (LC/MS/MS) method to measure the lycorine and galantamine content of human serum, which can be used for mild to fatal poisoning cases. A serum pretreatment procedure was performed using acetonitrile and QuEChERS AOAC powder. The separation of the compounds was conducted using a pentafluorophenyl column, CAPCELL CORE PFP (2.1 mm I.D. × 100 mm, 2.7 μm). Lycorine, galantamine, and galantamine-d6 (internal standard) were identified by the transitions of m/z 288 → 147, m/z 288 → 213, and m/z 294 → 216, respectively. The calibration curves were linear in the ranges of 0.05 to 5 ng/mL and 5 to 100 ng/mL, with R2 > 0.999. The precision and accuracy were within the permissible range. The matrix effects of lycorine and galantamine were 94.3% - 98.4% and 87.8% - 91.1%, respectively. The extraction recovery rates of lycorine and galantamine were 101.9% - 112.7% and 95.6% - 107.1%, respectively. The present method detected lycorine and galantamine in the sera of three patients with mild poisoning that had accidentally ingested. This method is applicable in cases of lycorine and galantamine poisoning.

The aim in this study is to examine the effect of tirapazamine (TPZ) and mild temperature hyperthermia (MTH) on the repair of radiation-induced damage in pimonidazole-unlabeled quiescent (Q) tumor cells. Labeling of proliferating (P) cells in C57BL/6J mice bearing EL4 tumors was achieved by continuous administration of 5-bromo-2-deoxyuridine (BrdU). Tumors were irradiated with γ-rays at 1 h after the administration of pimonidazole followed by TPZ treatment or MTH. Twenty-four hours later, assessment of the responses of Q and total (= P + Q) cells were based on the frequencies of micronucleation and apoptosis using immunofluorescence staining for BrdU. The response of the pimonidazole-unlabeled tumor cell fractions was assessed by means of apoptosis frequency using immunofluorescence staining for pimonidazole. With γ-rays only, the pimonidazole-unlabeled cell fraction showed significantly enhanced radio-sensitivity compared with the whole cell fraction more remarkably in Q cells than total cells. However, a significantly greater decrease in radio-sensitivity in the pimonidazole-unlabeled than the whole cell fraction, evaluated using a delayed assay, was more clearly observed in Q cells than total cells. Post-irradiation MTH more remarkably repressed the decrease in radio-sensitivity in the Q cell than the total cells. Post-irradiation TPZ administration produced a large radio-sensitizing effect on both total and Q cells, especially on Q cells. On the other hand, in pimonidazole-unlabeled cell fractions in both total and Q cells, TPZ suppressed the reduction in sensitivity due to delayed assay much more efficiently than MTH, whereas no radio-sensitizing effect was produced. Not only through suppressing the recovery from radiation-induced damage but also through radio-sensitizing effect, post-irradiation TPZ administration is very useful for repressing the increase in the difference in radio-sensitivity due to the delayed assay not only between total and Q tumor cells

Background: Recurrent head & neck malignancies (HNM) are often radio-/chemo-resistant and show extensive growth, necessitating a wide resection including surrounding tissues。To avoid severe impairment of oro-facial structures and functions, it is necessary to explore new treatments for HNM。Boron neutron capture therapy (BNCT) is tumor-cell targeted radiotherapy that has significant superiority over conventional radiotherapies in principle。When 10B absorbs thermal neutrons, the alpha and 7Li particles generated by the 10B(n, alpha)7Li reaction are high LET particles, and carry high kinetic energy (2.34MeV), and have short ranges (4-9 micron-meters) of approximately one-cell diameter, resulting in a large relative biological effectiveness and selective destruction of tumor cells containing 10B。We have, first in the world treated with BNCT for a patient with parotid gland carcinoma in 2001。Purpose: To evaluate the effect of BNCT on Recurrent head & neck malignancies。Methods: So far for 6 years, we have treated with 42 times of BNCT for 26 patients with a recurrent HNM who were composed of 19 squamous cell carcinomas (SCC), 4 salivary gland carcinomas and 3 sarcomas。Results: (1) 10B concentration of tumor/normal tissue ratios (T/N ratio) of PET studies wereSCC: 1.8-5.7, sarcoma: 2.5-4.0, parotidtumor: 2.5-3.7。(2)Regression rateswere CR: 12cases (46%), PR: 10cases (39%), PD: 3cases (12%), NE (not evaluated):lcase。

硝酸甘油诱导的血管舒张作为血流介导的血管舒张功能（flow-mediated vasodilation,FMD）的对照试验,用来区别内皮依赖性与非依赖性的血管舒张功能。近来,硝酸甘油诱导的血管舒张已被报道是动脉粥样硬化分级的有用标志物。该研究旨在评估联合FMD与硝酸甘油诱导的血管舒张对未来心血管事件的预测价值。方法与结果：纳入研究对象402例,其中包括心血管病患者,测定FMD与硝酸甘油诱导的血管舒张。
