Adjuvant chemotherapy improves the survival outlook for patients undergoing operations for lung metastases caused by colorectal cancer (CRC). However, a multidisciplinary approach that evaluates several factors related to patient and tumor characteristics is necessary for managing chemotherapy treatment in metastatic CRC patients with lung disease, as such factors dictate the timing and drug regimen, which may affect treatment response and prognosis. In this study, we explore the potential of spatial metabolomics for evaluating metabolic phenotypes and therapy outcomes during the local delivery of the anticancer drug, oxaliplatin, to the lung. 12 male Yorkshire pigs underwent a 3 h left lung in vivo lung perfusion (IVLP) with various doses of oxaliplatin (7.5, 10, 20, 40, and 80 mg/L), which were administered to the perfusion circuit reservoir as a bolus. Biocompatible solid-phase microextraction (SPME) microprobes were combined with global metabolite profiling to obtain spatiotemporal information about the activity of the drug, determine toxic doses that exceed therapeutic efficacy, and conduct a mechanistic exploration of associated lung injury. Mild and subclinical lung injury was observed at 40 mg/L of oxaliplatin, and significant compromise of the hemodynamic lung function was found at 80 mg/L. This result was associated with massive alterations in metabolic patterns of lung tissue and perfusate, resulting in a total of 139 discriminant compounds. Uncontrolled inflammatory response, abnormalities in energy metabolism, and mitochondrial dysfunction next to accelerated kynurenine and aldosterone production were recognized as distinct features of dysregulated metabolipidome. Spatial pharmacometabolomics may be a promising tool for identifying pathological responses to chemotherapy.

Due to a complex geological and biotic history,the Isthmus of Tehuantepec(IT),has been long recognized as a driver for the evolutionary divergence of numerous lowland and highland *** distributed in the lowlands of the American continent,the White-Tipped Dove(Leptotila verreauxi)is a polytypic species with 13 recognized *** of these have been recorded in Mexico,and the distribution of three abuts at the IT,suggesting a contact *** estimate phylogenetic patterns,divergence times and genetic differentiation,we examined two mt DNA(ND2 and COI)and one n DNA(β-fibint 7)*** also used correlative ecological niche models(ENM)to assess whether ecological differences across the IT may have acted as a biogeographical *** estimated paleodistributions during the Middle Holocene,Last Glacial Maximum and Last Interglacial,to evaluate the influence of climate changes on the distribution and demographic *** results showed genetically distinct lineages that diverged approximately 2.5 million years *** and ecological factors may have played a dual role in promoting differentiation,but also in the formation of a secondary contact zone in the southern *** ecological niche comparisons indicated that the ecological niche of sympatric lineages at the IT are not identical,suggesting niches divergence;in addition,environmental niche models across the region indicated no abrupt biogeographic barriers,but the presence of regions with low *** results suggest that genetic differentiation originated by a vicariant event probably related to environmental factors,favored the evolution of different ecological ***,the absence of a biogeographic barrier but the presence of less suitable areas in the contact regions,suggest that secondary contact zones may be also maintained by climatic factors for the eastern group,but also by biotic interactions for the western group.

我们在本文研究了具有不同磁致伸缩系数的Fe_(73.5)Si_(13.5)B_9Nb_3Cu_1和Fe_(73.5)Si_(16.5)B_6Nb_3Cu_1纳米晶带材在0.1-2MHz范围内磁阻和频率的关系。铸态样品分别在施加直流和交流磁场及未施加直流和交流磁场中进行氩气氛560℃退火。在某一固定频率,含13.5％Si的样品加磁场退火和未加磁场退火后相比,可看到性能有了改进。含16.5％Si的样品加磁场退火和未加磁场退火相比,仅观察到磁阻率减小。磁特性分析和X光衍射数据显示,所观察到的磁阻效应改变是由于磁感应各向异性的结果。

Under the present changing climate conditions and the observed temperature increase,it is of high importance to understand its effects on aquatic microbial life,and organisms’adaptations at the biochemical *** adjust to temperature or salinity stress and avoid cell damage,organisms alter their degree of fatty acids(FAs)***,temperature is expected to have strong effects on both the quantity and quality of FAs in aquatic *** we review some recent findings about FAs sensitivity to climate change in contrasting ***,heat waves may induce changes in the relative abundance of polyunsaturated FAs(PUFA).However,the impact of the exposure to warming waters is different in temperate and polar *** cold marine waters,high concentration of omega-3(ω3)FAs such as eicosapentaenoic acid(EPA)is promoted due to the activation of the desaturase *** this way,cells have enough energy to produce or activate antioxidant protection mechanisms and avoid oxidative stress due to heat ***,under high irradiance and heat wave conditions in temperate environments,photosystems’protection is achieved by decreasing EPA concentration due to desaturase *** FAs are transferred in aquatic food ***,any alteration in the production of essential FAs by phytoplankton(the main source ofω3)due to climate warming can be transferred to higher trophic levels,with cascading effects for the entire aquatic ecosystem.

Glioblastoma(GBM,WHO grade IV glioma)is the most common and lethal malignant brain tumor in aduts with a dismal *** extracellular matrix(ECM)supports GBM progression by promoting tumor cell proliferation,migration,and immune *** diphosphate(UDP)-glucose 6-dehydrogenase(UGDH)is the rate-limiting enzyme that catalyzes the biosynthesis of glycosaminoglycans that are the principal component of the CNS *** investigated how targeting UGDH in GBM infuence$the GBM immune microenvironment,including tumor-associated microglia/macrophages(TAMs)and T *** are the main im-mune effector cells in GBM and can directly target tumor cells if properly *** co-cultures of GBM cells and human primary macrophages,UGDH knockdown in GBM cells pro-moted macrophage phagocytosis and M-like *** orthotropic human GBM xeno-grafts and syngeneic mouse glioma models,targeting UGDH decreased ECM deposition,increased TAM phagocytosis marker expression,reduced M2-like TAMS and inhibited tumor *** knockdown in GBM cells also promoted cytotoxic T cell ifltration and activa-tion in orthotopic syngeneic mouse glioma *** potent and in-human-use small mole-cule GAG synthesis inhibitor 4-methylumbelliferone(4-MU)was found to inhibit GBM cell proliferation and migration in vitro,mimic the macrophage and T-cell responses to UGDH knockdown in vitro and in vivo and inhibit growth of orthotopic murine *** study shows that UGDH supports GBM growth through multiple mechanisms and supports the development of ECM-based therapeutic strategies to simultaneously target tumor cells and their microenvi-ronment.

Cyanobacteria are phototrophic organisms with great ecological and economical *** of the genus Microcystis are known for their potential ability to synthesize toxins,notably *** is a growing interest in the evaluation of oxidative stress in relation to the impact of global climate change on natural ecosystems in different trophic *** studies have focused on the analysis of organismal responses to mitigate the damage by controlling the generation of reactive oxygen *** in environ-mental factors caused by climate change generate a situation of oxidative damage in Microcystis aeruginosa as a direct or indirect *** this study we evaluate the effects of ultraviolet radiation and temperature on physiological and biochemical responses of a native ***(strain CAAT 2005-3).The results from the exposure to ultraviolet radiation doses and temperature changes suggest a high ability of *** to detect a potential stress situation as a consequence of reactive species production and to rapidly initiate antioxidant *** catalase activity is an antioxidant protection mechanism in *** for short and long term exposure to different changes in environmental ***,we found a ultraviolet-B radiation threshold dose above which oxidative stress exceeds the antioxidant protection and damage *** addition our results are in agreement with recent findings suggesting that microcystins may act as protein-modulating metabolites and protection against reactive oxygen *** is concluded that cyanobacteria have adaptative mechanisms that could lead to the replacement of species highly susceptible to oxidative stress by others with a higher system of antioxidant protection.
