This paper is the second in a series focused on providing a stable platform for the taxonomy of phytopathogenic *** focuses on 25 phytopathogenic genera:Alternaria,Bipolaris,Boeremia,Botryosphaeria,Calonectria,Coniella,Corticiaceae,Curvularia,Elsinoe,Entyloma,Erythricium,Fomitiporia,Fulviformes,Laetisaria,Limonomyces,Neofabraea,Neofusicoccum,Phaeoacremonium,Phellinotus,Phyllosticta,Plenodomus,Pseudopyricularia,Tilletia,Venturia and Waitea,using recent molecular data,up to date names and the latest taxonomic *** each genus a taxonomic background,diversity aspects,species identification and classification based on molecular phylogeny and recommended genetic markers are *** this study,varieties of the genus Boeremia have been elevated to species ***,Bipolaris,Curvularia,Neofusicoccum and Phyllosticta that were included in the One Stop Shop 1 paper are provided with updated entries,as many new species have been introduced to these genera.

Cytogenetic analysis of chromosomal abnormalities in Sri Lankan children

Background: Cytogenetic analysis is a valuable investigation in the diagnostic work up of children with suspected chromosomal disorders. The objective of this study was to describe the prevalence of various types of chromosomal abnormalities in Sri Lankan children undergoing cytogenetic analysis. Methods: Cytogenetic reports of 1554 consecutive children with suspected chromosomal disorders who underwent karyotyping in two genetic centers in Sri Lanka from january 2006 to December 2011 were reviewed retrospectively. Results: A total of 1548 children were successfully karyotyped. Abnormal karyotypes were found in 783 (50.6%) children. Numerical and structural abnormalities accounted for 90.8% and 9.2%, respectively. Down syndrome was the commonest aneuploidy identifi ed. Other various autosomal and sex chromosomal aneuploidies as well as micro-deletion syndromes were also detected. Conclusions: The prevalence of chromosomal abnormalities in Sri Lankan children undergoing cytogenetic analysis for suspected chromosomal disorders was relatively higher than that in Caucasian and other Asian populations.

Grapevines(Vitis vinifera)are colonized by ubiquitous microorganisms known as endophytes,which may have advantageous or neutral effects without causing disease *** endophytes are uncultivable,so culture-independent approaches such as next generation sequencing(NGS)can help for a better understanding of their ecology and *** date,there are no studies which directly link NGS results with taxa derived from a culturing approach,integrating morphological and multi-gene phylogenetic analysis of *** this study,a culture-dependent and high-resolution culture-independent approach(next generation sequencing)were used to identify endophytes in grapevine *** the culture-dependent approach,a total of 94 isolates were recovered from 84 of 144 healthy grapevine stem fragments(colonization rate=58.3%).The study is unique as we used subsets of combined multi-gene regions to identify the endophytes to species *** on each multi-gene phylogenetic analysis,28 species belong to 19 genera(Acremonium,Alternaria,Arthrinium,Ascorhizoctonia,Aspergillus,Aureobasidium,Bipolaris,Botryosphaeria,Botrytis,Chaetomium,Cladosporium,Curvularia,Hypoxylon,Lasiodiplodia,Mycosphaerella,Nigrospora,Penicillium,Phoma,Scopulariopsis)were identified.A higher number of culturable fungi were obtained from 13 year-old vines,followed by eight and three yearold *** the culture-independent approach,a fungal richness of 59 operational taxonomic units(OTU)was detected,being highest in 13 year-old grapevines,followed by eight and three *** though the cultivation approach detected lower fungal richness,the results related to stem are consistent for fungal community composition and *** of the fungal taxa identified by the two approaches resulted in an overlap of 53%of the fungal *** to interspecific variability of the sequences from NGS,in many cases the OTUs(even with the highly abundant ones)were only assignable to order,family or genus

Fungi have been often neglected,despite the fact that they provided penicillin,lovastatin and many other important *** are an understudied,but essential,fascinating and biotechnologically useful group of *** study of fungi in northern Thailand has been carried out by us since *** studies have been diverse,ranging from ecological aspects,phylogenetics with the incorportation of molecular dating,taxonomy(including morphology and chemotaxonomy)among a myriad of microfungi,to growing novel mushrooms,and DNA-based identification of plant *** this paper,advances in understanding the biodiversity of fungi in the region are discussed and compared with those further *** new species have been inventoried for the region,but many unknown species remain to be described and/or *** example,in the edible genus Agaricus,over 35 new species have been introduced from northern Thailand,and numerous other taxa await *** this relatively well known genus,93%of species novelty is *** the microfungi,which are relatively poorly studied,the percentage of novel species is,surprisingly,generally not as high(55–96%).As well as Thai fungi,fungi on several hosts from Europe have been also *** with the well studied European microfungi an astounding percentage of new taxa(32–76%)have been *** work is just a beginning and it will be a daunting task to document this astonishingly high apparent novelty among fungi.

Botryosphaeria dieback is a serious disease problem for table and grape wine production *** disease however,has been less well-studied in *** this study,we surveyed Botryosphaeria dieback in 72 vineyards of 20 grape-growing regions in China and found that Botryosphaeria dieback occurs in 18 out of 20 *** and multi-gene phylogenetic analyses confirmed that Botryosphaeria dothidea,Diplodia seriata,Lasiodiplodia theobromae and Neofusicoccum parvum are associated with different grapevine dieback *** study also revealed considerable differences in the geographical distribution of Botryosphaeriaceae species in China with Lasiodiplodia theobromae and Neofusicoccum parvum occurring only in subtropical monsoon climate regions,Diplodia seriata occurring only in temperate monsoon climate regions,and Botryosphaeria dothidea occurring in both temperate and subtropical monsoon climate *** of 26 isolates showed that there is little genetic variation within ***’s postulates were satisfied for all species,and pathogenicity testing showed that among the 25 major cultivars growing in China,none was resistant to the four *** current paper represents the first detailed report on Botryosphaeria dieback of grapevines in China.

t Coelomycetous fungi are an artificial taxonomic group which produce conidia inside a cavity *** comprise about,1000 genera and 7000 species,which can be endophytic,pathogenic or *** classification of coelomycetes was previously based on morphology,such as the shape of conidiomata and mode of conidiogenesis,while it was treated as a distinct group *** based taxonomic studies has been used to accommodate asexual fungi in a natural classification system,resolve generic boundaries of polyphyletic genera and species complexes,as well as establish asexual-sexual ***,most of genera lack sequence data,thus,morphology based identification is still important when introducing new genera or *** this paper we illustrate,describe,and provide taxonomic notes for 235 dematiaceous coelomycetous genera,including five new genera ***,Didymellocamarosporium,Melanocamarosporium,Melnikia and *** analyses of combined sequence data are provided to show placements of dematiaceous coelomycetes in Dothideomycetes,Leotiomycetes and ***-hundred and fifty-two(65%)of genera have sequence data,thus,their taxonomic placement in a natural classification system,is listed as an ***,83 genera still lack sequence data,hence,they are treated as Ascomycota,genera incertae *** addition,separate analyses are provided where better taxonomic resolution is needed.
