Detection of HbV DNA integration in plasma cell-free DNA of different HbV diseases utilizing DNA capture strategy

The landscape of hepatitis b virus (HbV) integration in the plasma cell-free DNA (cfDNA) of HbV-infected patients with different stages of liver diseases [chronic hepatitis b (CHb), liver cirrhosis (lC), and hepatocellular carcinoma (HCC)] remains unclear. In this study, we developed an improved strategy for detecting HbV DNA integration in plasma cfDNA, based on DNA probe capture and next-generation sequencing. Using this optimized strategy, we successfully detected HbV integration events in chimeric artificial DNA samples and HbV-infected HepG2-NTCP cells at day one post infection, with high sensitivity and accuracy. The characteristics of HbV integration events in the HbV-infected HepG2-NTCP cells and plasma cfDNA from HbV-infected individuals (CHb, lC, and HCC) were further investigated. A total of 112 and 333 integration breakpoints were detected in the HepG2-NTCP cells and 22 out of 25 (88%) clinical HbV-infected samples, respectively. In vivo analysis showed that the normalized number of support unique sequences (nnsus) in HCC was significantly higher than in CHb or lC patients (P values ll integration breakpoints are randomly distributed on human chromosomes and are enriched in the HbV genome around nt 1800. The majority of integration breakpoints (61.86%) are located in the gene-coding region. both non-homologous end-joining (NHEJ) and microhomology-mediated end-joining (mmEJ) interactions occurred during HbV integration across the three different stages of liver diseases. Our study provides evidence that HbV DNA integration can be detected in the plasma cfDNA of HbV-infected patients, including those with CHb, lC, or HCC, using this optimized strategy. © 2024 The Authors

Plasma electrolytic formation and characterization of mnWO_(4)/WO_(3) film heterostructures

mnWO_(4)/WO_(3) p-n heterojunction films were fabricated using a one-step method consisting of the plasma electrolytic oxidation(PEO)of titanium in homogeneous electrolytes containing paratungstate ions and stable watersoluble EDTA-chelated *** influences of the formation current density and W:mn molar ratio of the electrolyte,which was varied from 1:2 to 2:1,on the composition,morphology,and optical and photocatalytic properties of the resulting coatings were studied.X-ray diffraction analysis,scanning electron microscopy,energy dispersive X-ray analysis,Raman spectroscopy,and ultraviolet diffuse reflectance spectroscopy were used to characterize the formed *** of the W:mn ratio of the electrolyte,the coatings contained crystalline t-WO_(3) and m-mnWO_(4).Depending on the formation conditions,the optical band gap energies of the composites varied from 2.63 to 3.01 *** largest absorption red shift and lowest band gap energy were observed in the film composite formed in an electrolyte with W:mn=2:1,at a current density of 0.2 A cm^(−2).Composites obtained in electrolytes with W:mn ratios of 2:1 and 1:1 exhibited photocatalytic activity in the degradation of rhodamine C and methyl orange dyes in the presence of 10 mmol l^(–1) H2O_(2) under ultraviolet and visible light *** role of hydrogen peroxide in this dye degradation on PEO-coated composites under light irradiation is discussed.

One-neutron stripping process between^(6)li and^(209)bi was studied at 28,30,and 34 meV using the in-beamγ-ray spectroscopy ***γ-γcoincident analysis clearly identified twoγ-rays feeding the ground and long-lived isomeric states,which were employed to determine the cross *** one-neutron stripping cross sections were similar to the cross sections of complete fusion in the^(6)li+^(209)bi system,but the one-neutron stripping cross sections decreased more gradually at the sub-barrier region.A coupled-reaction-channel calculation was performed to study the detailed reaction mechanism of the one-neutron stripping process in^(6)*** calculations indicated that the first excited state of 5 li is critical in the actual one-neutron transfer mechanism,and the valence proton of 209bi can be excited to the low-lying excited state in(^(6)li,^(5)li)reaction,unlike in the(d,p)reaction.

We explore the gapped graphene structure in the two-dimensional plane in the presence of the Rosen-morse potential and an external uniform magnetic *** order to describe the corresponding structure,we consider the propagation of electrons in graphene as relativistic fermion quasi-particles,and analyze it by the wave functions of two-component spinors with pseudo-spin symmetry using the Dirac ***,to solve and analyze the Dirac equation,we obtain the eigenvalues and eigenvectors using the legendre differential *** that,we obtain the bounded states of energy depending on the coefficients of Rosen-morse and magnetic potentials in terms of quantum numbers of principal n and spin-orbit ***,the values of the energy spectrum for the ground state and the first excited state are calculated,and the wave functions and the corresponding probabilities are plotted in terms of coordinates *** what follows,we explore the band structure of gapped graphene by the modified dispersion relation and write it in terms of the two-dimensional wave vectors K_(x) and K_(y).Finally,the energy bands are plotted in terms of the wave vectors K_(x) and K_(y) with and without the magnetic term.

The mesoproterozoic(1.11 Ga)Umkondo large igneous province(lIP)in southern Africa and Antarctica was emplaced inmyr and is dominated by low-Ti tholeiitic doleritic-gabbroic *** is of particular interest because it is the least studied lIP in southern Africa with both sublithospheric and lithospheric mantle sources proposed and it coincides with the early assembly of Rodinia,so it has importance in understanding the nature of magmatism and tectonics in and around the Kalahari craton during the *** this study,we compiled a large database of existing(~750)and new(~100)major and trace element data for the Umkondo province,as well as 42 new Sr-Nd isotopic measurements,to provide constraints on its magma sources and geochemical *** element compositional variations in the low-Ti tholeiites are explained by low-pressure(1 kbar)three-phase fractional crystallisation(olivine,clinopyroxene and plagioclase)of a parent magma with~10 wt.%mgO in oxidising conditions(QFm+1).Inverse models show that the low-Ti tholeiitic magmas were derived as residual melts after the crystallization of 12%-33%olivine from primary komatiitic-basaltic magmas(up to~20 wt.%mgO)in equilibrium with mantle olivine(Fo90).low Sm/Yb and TiO2/Yb-Nb/Yb indicate that the primary magmas were derived by 2%-20%shallow(40-50 km)partial melting of spinel *** Sm/Yb is restricted to dyke swarms and may imply limited magma production from deeper(up to~70 km)garnet lherzolite-like *** low-Ti tholeiites of the Umkondo province are enriched in large ion lithophile elements(Rb-Sr-Cs-K)and depleted in high-field strength elements(Zr-Hf-Nb-Ta),indicating the involvement of crustal material and/or the subcontinental lithospheric *** is supported by covariations in Th/Nb,Nb/Yb,Nb/la and Ce/Sm with generally negativeΔ***-Nd isotopes lend support to the notion that the Umkondo magmas were derived from depleted and/or enriched sublithospheric mantle sources and subs

A Retrospective Analysis of Intoxicated Patients Admitted to the Intensive Care Unit: A Single-Institution Assessment in the Post-Pandemic Period

background: The aim of this study was to evaluate the clinical and epidemiological characteristics of patients with acute intoxication admitted to the Intensive Care Unit (ICU). material and methods: An observational retrospective study was conducted on intoxicated patients who admitted to ICU between January 2022 and January 2024. Data were collected from the patients medical records. The demographic characteristics, causes of intoxications, clinical parameters, the mean stay in the ICU, treatment modalities and prognosis were recorded. Results: A total of 2875 critically ill patients were admitted during the study period, and 109 (3.79%) of them were acute intoxications. Their mean of ages was 38.09 ± 12.29. The female-to-male ratio was 1.37/1. Drugs were found to be the primary cause (62.39%) of intoxications, and analgesics, antidepressants, and antibiotics were the most frequent agents. Suicidal attempts were present in 66 patients, most of them were female (62.13%) and between 17 - 24 years (40.91%). The other common causes of intoxications were carbon monoxide (CO) (22.02%), methyl/ethyl alcohol (8.26%) and mushroom (5.50%). The mean stay in the ICU was 2.69 ± 0.89 days. mechanical ventilation was applied to 10 of our patients. Renal replacement therapy was required in 6 patients. Despite all treatments, 6 of our patients died, and we found the mortality rate to be 5.50%. Conclusion: Intoxications were more frequent in young female patients and drugs were the most common cause with suicidal intent. Unfortunately, CO poisoning continues to be a very important problem in our city. These findings provided significant information about the characteristics of intoxications in Karabuk.
