In this study,three different groups of male broodstocks according to their age （4,5 and 6 respectively） were used to fertilizing of eggs which obtained from nine *** results showed that 6 years old males had maximum body weight （1766.67 g）,total length （56.33 cm） and sperm volume （31.83 ml）.Our results did not show significant difference in values of spermatocrit and spermatozoa concentration among male broodstocks （P＞0.05）.The present study showed maximum fertilization percentage （98.5 %）,survival rate until eyeing percentage （91.17 %）,hatching percentage （94.5 %） and survival rate until absorption of yolk sack stage （97.16 %） when sperm of 4 years old males was used to inseminate of *** positive relationships were detected between sperm production characteristics （spermatozoa concentration,spermatocrit and sperm volume） and fertilization *** on our results,can be concluded that 4 years old males had high efficiency for achieve fertilization success.

Efficient generation of zebrafish maternal-zygotic mutants through transplantation of ectopically induced and Cas9/gRNA targeted primordial germ cells

The clustered regularly interspaced short palindromic repeats(CRISPR)/Cas9 technology has been widely utilized for knocking out genes involved in various biological processes in zebrafish. Despite this technology is efficient for generating different mutations, one of the main drawbacks is low survival rate during embryogenesis when knocking out some embryonic lethal genes. To overcome this problem, we developed a novel strategy using a combination of CRISPR/Cas9 mediated gene knockout with primordial germ cell(PGC) transplantation(PGCT) to facilitate and speed up the process of zebrafish mutant generation, particularly for embryonic lethal genes. Firstly, we optimized the procedure for CRISPR/Cas9 targeted PGCT by increasing the efficiencies of genome mutation in PGCs and induction of PGC fates in donor embryos for PGCT. Secondly, the optimized CRISPR/Cas9 targeted PGCT was utilized for generation of maternal-zygotic(MZ) mutants of tcf7l1a(gene essential for head development), pou5f3(gene essential for zygotic genome activation) and chd(gene essential for dorsal development) at F1 generation with relatively high efficiency. Finally, we revealed some novel phenotypes in MZ mutants of tcf7l1 a and chd, as MZtcf7l1 a showed elevated neural crest development while MZchd had much severer ventralization than its zygotic counterparts. Therefore, this study presents an efficient and powerful method for generating MZ mutants of embryonic lethal genes in zebrafish. It is also feasible to speed up the genome editing in commercial fishes by utilizing a similar approach by surrogate production of CRISPR/Cas9 targeted germ cells.

ANFIS and ANNs model for prediction of moisture diffusivity and specific energy consumption potato,garlic and cantaloupe drying under convective hot air dryer

The main purpose of this study was to develop and apply an adaptive neuro-fuzzy inference system(ANFIS)and Artificial Neural Networks(ANNs)model for predicting the drying characteristics of potato,garlic and cantaloupe at convective hot air *** experiments were conducted at the air temperatures of 40,50,60 and 70C and the air speeds of 0.5,1 and l.5 m/*** properties were including kinetic drying,effective moisture diffusivity(Deff)and specific energy consumption(SEC).The highest value of Deff obtained 9.76×10^-9,0.13×10^-9 and 9.97×10^-10 m^2/s for potato,garlic,and cantaloupe,*** lowest value of SEC for potato,garlic,and cantaloupe were calculated 1.94105,4.52105 and 2.12105 kJ/kg,*** revealed that the ANFIS model had the high ability to predict the Deff(R^2=0.9900),SEC(R^2=0.9917),moisture ratio(R^2=0.9974)and drying rate(R^2=0.9901)during *** ANFIS method had the high ability to evaluate all output as compared to ANNs method.
